L'élément de qualité phytoplancton doit être évalué dans le cadre de la Directive Cadre sur L’Eau grâce à trois indices : biomasse, abondance et composition. Les deux premiers indices, biomasse et abondance, ont déjà été définis. L’indice composition a quant à lui fait l'objet d'une proposition précise en Méditerranée, mais pas en Manche - Atlantique. Les travaux antérieurs montrent que la construction d'un indice de composition doit tenir compte de la diversité de l'ensemble du phytoplancton. Or, les comptages microscopiques ne concernent en général que le micro-phytoplancton. This is a summary of the activities and results of JERICO-NEXT Work Package 3 Innovations in Technology and Methodology, Task 3.1 Automated platform for the observation of phytoplankton diversity in relation to ecosystem services. The aim is to provide an advanced report on the last developments dedicated to the observation of the phytoplankton diversity by applying novel techniques on automated platforms. The work was carried out in close connection with task 4.1 Biodiversity of plankton, harmful algal blooms and eutrophication. Phytoplankton distribution from Western to Central English Channel, revealed by automated flow cytometry during the summer-fall transition ArchiMer

Automated pulse shape-recording flow cytometry was applied to address phytoplankton spatial distribution, at high frequency, in stratified and well mixed water masses in the Western and Central English Channel during the summer-fall transition. Cytometric pulse shapes derived from optical features of single cells allowed the characterization of eight phytoplankton groups. Abundance and total red fluorescence (chlorophyll a autofluorescence) per group were used to define six phytoplankton communities. Their distribution revealed high spatial heterogeneity. This study is the first in situ work comparing rapid light curves (RLC) and non-sequential steady-state light curves (N-SSLC) in their efficiency to characterize phytoplankton photosynthetic activity and acclimation status. Measurements were carried out at two time scales (daily and annual) using the Pulse Amplitude Modulated fluorometry on samples taken in the coastal waters of a macrotidal ecosystem (the Strait of Dover, eastern English Channel). Spectral fluorometric characterization of Haptophyte dynamics using the FluoroProbe: an application in the eastern English Channel for monitoring Phaeocystis globosa ArchiMer

In this study, we examined the possibility of using the FluoroProbe for monitoring the dynamics of the Haptophyte Phaeocystis globosa in the coastal waters of the eastern English Channel. The FluoroProbe was recalibrated by recording a new fingerprint for P. globosa and the use of this new fingerprint was tested through a series of laboratory and in situ experiments. The annual dynamics of P. globosa estimated using the FluoroProbe and by flow cytometry were similar. A strong relationship was found between the FluoroProbe estimates of P. globosa biomass expressed in terms of chlorophyll a equivalent per litre (eq. mu g L(-1)) and flow cytometric cell counts (r = 0.889, P < 0.001, n = 121). The European Union Water Framework Directive (WFD) refers to phytoplankton as one of the biological quality elements that should be regularly monitored and evaluated thanks to three ecological indices: biomass, abundance and composition. The first two have already been defined, but the third one has not yet been proposed in the English Channel-Atlantic zone. The construction of a composition indice must consider the diversity of all the phytoplankton. High spatial variability of phytoplankton assessed by flow cytometry, in a dynamic productive coastal area, in spring:the eastern English Channel ArchiMer

The distribution of phytoplankton (from pico-to microphytoplankton) was investigated, at single-cell level and at high spatial resolution, during an oceanographic cruise across the eastern English Channel (EEC) between April 27 and 29, 2012. Seawater was continuously collected from surface waters and analysed on board at high frequency (one sample every 10 min), by using a new generation of pulse-shape recording scanning flow cytometer (CytoSense, Cytobuoy©). A Bray-Curtis matrix analysis based on phytoplankton composition allowed the discrimination of 4 communities.
