Cryopreservation of zygotic embryos of the Brazilian Green Dwarf coconut PAB

Abstract: The objective of this work was to evaluate rewarming procedures and recovery media for Brazilian Green Dwarf coconut (Cocos nucifera) embryos cryopreserved by vitrification. The rewarming procedures evaluated were: T1, water bath at 38±2°C; and T2, unloading solution consisting of Y3 medium + 1.2 mol L-1 sucrose. The recovery media assessed were: M1, Y3 medium + 45 g L-1 sucrose + 1 mg L-1 gibberellic acid + 1 g L-1 activated charcoal + 2.2 g L-1 Gelrite; and M2, Y3 medium + 60 g L-1 sucrose + 2.2 g L-1 Gelrite. Rewarming procedures showed no significant effect, and the M1 media induced 72.5% regeneration.

Manitol para a conservação ex situ de coqueiro por crescimento mínimo PAB

O objetivo deste trabalho foi determinar a concentração de manitol para a germinação e crescimento mínimo in vitro do coqueiro (Cocos nucifera). Os seguintes acessos foram utilizados: Anão-vermelho-do brasil-de-gramame (BRDG), Anão-amarelo-da-malásia (MYD), Gigante-do-brasil-praia-do-forte (BRA), Gigante-da-polinésia (PYT) e Gigante-de-rennel (RIT). Os embriões foram colocados em meio de cultura Y3 a diferentes concentrações de manitol (0, 0,1, 0,2, 0,3 e 0,4 mol L-1). Verificou-se inibição do crescimento da parte aérea na presença de 0,1, 0,2 e 0,3 mol L-1, para MYD e BRDG, e de 0,1 e 0,2 mol L-1, para PYT, BRA e RIT. O uso do manitol é uma estratégia promissora para a conservação por crescimento mínimo.

Storage as a means of maintaining the pollen viability is important for the preservation of the genetic variability, facilitates the exchange of germplasm and greatly contributes to the generation of variability obtained from artificial crosses, increasing the efficiency of breeding programs. The objective of this study was to select different culture media for the in vitro germination of pollen grain of dwarf and tall coconut accessions, as well as to determine the viability of pollen grain at room temperature conditions. In vitro morphogenic response from zygotic embryos of Genipa americana Ciência Rural

ABSTRACT: The aim of this study was to evaluate the in vitro morphogenic potential of genipap (Genipa americana L.) zygotic embryos. Seeds obtained from ripe fruits had their zygotic embryos excised and inoculated in MS medium with 4.44µM of 6-benzylaminopurine (BAP) and supplemented with 0.0; 1.07; 2.14 and 3.21µM of naphthalene acetic acid (NAA). The potential of explants regeneration and the shoot length and number of leaves in plantlets were evaluated. The in vitro regeneration of genipap is possible from the conversion of zygotic embryos in a MS medium with 4.44µM BAP supplemented with 3.21µM NAA.

In vitro germination and acclimatization of cambui tree type seedlings Ciência Rural

There are few reports in literature on the in vitro behavior of cambui tree (Myrciaria tenella O. Berg) and acclimatization conditions. The aim of this study was to evaluate the effect of culture media on in vitro germination and the effect of different substrates on the acclimatization of two Myrciaria tenella types. The study was carried out at the Embrapa Tabuleiros Costeiros Laboratory of Plant Tissue Culture, Aracaju, SE. Seeds were extracted from fruits of two Myrciaria tenella types: Orange and Purple Types. Strategies for exchange of coconut germplasm in Brazil Ciência Rural

ABSTRACT: Seed size and its physiology are a barrier for the exchange of coconut genetics resources and the tissue culture techniques can assist the germplasm bank enrichment. The aim of this study was to evaluate the effect of transport and storage procedures of zygotic embryos to promote the exchange of coconut accessions. Mature zygotic embryo from Cameroon red dwarf (CRD), Malayan yellow dwarf (MYD) and Malayan red dwarf (MRD) were used. Proline synthesis and physiological response of cassava genotypes under in vitro salinity Ciência Rural

ABSTRACT: The objective of this research was to evaluate the proline synthesis and physiological response of cassava genotypes which were micro propagated and induced to salinity stress in vitro. Micro cuttings of approximately 1.0cm long with a single bud of genotypes TBRS Tapioqueira, BRS Verdinha and Lagoão which were previously established in vitro were inoculated in a MS medium containing different concentrations of NaCl (0; 25; 50; 75; 100mM) and were analyzed after 90th day for: number of roots, number of leaves and shoot dry mass.
