The iron uptake mechanisms of enteroinvasive Escherichia coli BJM

Enteroinvasive Escherichia coli strains (EIEC) of different serotypes isolated from patients with acute diarrhea were examined for the ability to produce siderophores and iron-regulated outer membrane proteins (IROMP). For iron starvation cultures were grown at 37°C in LB supplied with 200 muM of <FONT FACE="Symbol">a</font>-alpha’dypirydil. All strains produced enterobactin and twelve (40%) produced aerobactin. The strains showed IROMP varying from 67-82 kDa. Proteins were either induced or stimulated by the iron starvation. Differences were observed in the electrophoretic profile among the serotypes, originating 5 electrophoretic profiles. ABSTRACT Most Escherichia coli strains live harmlessly in the intestines and rarely cause disease in healthy individuals. Nonetheless, a number of pathogenic strains can cause diarrhea or extraintestinal diseases both in healthy and immunocompromised individuals. Diarrheal illnesses are a severe public health problem and a major cause of morbidity and mortality in infants and young children, especially in developing countries. E. coli strains that cause diarrhea have evolved by acquiring, through horizontal gene transfer, a particular set of characteristics that have successfully persisted in the host. Molecular characterization of enteroinvasive Escherichia coli ipa genes by PCR-RFLP analysis BJM

In this study, polymorphism in ipa genes was found in five out of nine EIEC serotypes studied. When SalI and HindII were used in RFLP-PCR assays many EIEC serotypes showed polymorphism in ipaB and ipaD. On the other hand, no polymorphism was observed in ipaA and ipaC in these strains. The polymorphism present in EIEC strains is serotype-dependent, since restriction patterns were conserved amongst strains belonging to the same serotype. When IpaB deduced amino acid sequences of S. flexneri M90T and FBC124-13 were compared, ten amino acids changes could be observed mainly in the amino-terminal region.
