Pulse versus continuous peracetic acid applications: Effects on rainbow trout performance, biofilm formation and water quality Organic Eprints
Liu, Dibo; Strauss, David L.; Pedersen, Lars-Flemming; Meinelt, Thomas.
Peracetic acid (PAA) products are being introduced to aquaculture as sustainable disinfectants. Two strategies are used to apply PAA: high dose pulse applications, or low dose continuous application. In the present study, their impacts on fish health and water quality were investigated in triplicate flow-through tanks stocked with rainbow trout. The gentler and shorter water cortisol increase measured along twice-per-week pulse applications of 1 mg L−1PAA indicated a progressive adaptation of fish. In contrast, the continuous application of 0.2 mg L−1PAA caused no stress to fish. Meanwhile, no mortality and no impact on growth or innate cellular immunity were observed. The pulse applications restricted biofilm formation, and partially inhibited...

SHELTERFISH New tools to improve fish health and environment in organic aquaculture Organic Eprints
Jokumsen, Alfred; Gesto, Manuel; Buchmann, Kurt; Lorenzen, Niels; Pedersen, Lars-Flemming; Henriksen, Niels Henrik; Schmidt, Jacob; Madsen, Lone; Kania, Per; Pedersen, Per Bovbjerg.
Rainbow trout farming and in particular organic rainbow trout farms are critically challenged by the relatively high prevalence of skin/gill infections caused by various pathogens, especially the parasite Costia (Ichthyobodo necator) and amoebae, which are ultimately lethal for fry/smaller fish. In addition, a Midichloria-like bacterium causes the non-lethal skin disease Red Mark Syndrome (RMS), which results in downgrading/rejection of up to 30% of the fish when placed on the market. Treatment by use of antibiotics/parasitics/ auxiliary compounds is only possible to a limited extent in organic trout production. Hence, solutions to prevent and/or treat costia, amoebae and RMS are urgently needed, not only to secure production of organic rainbow trout in...

Pedersen, Lars-Flemming; Jokumsen, Alfred.

Confirmation that pulse and continuous peracetic acid administration does not disrupt the acute stress response in rainbow trout Organic Eprints
Gesto, Manuel; Liu, Dibo; Pedersen, Lars-Flemming; Meinelt, Thomas; Strauss, David L.; Jokumsen, Alfred.
Peracetic acid (PAA) is considered an eco-friendly alternative to other antimicrobial agents of common use in aquaculture. The literature suggests that fish can habituate to PAA exposure based on a reduction of the fish corticosteroid response to PAA administration after repeated exposures. If that is true, PAA would also be a good option from the point of view of fish physiology. However, stronger evidence is needed to confirm that the use of PAA is welfare-friendly to fish. Besides habituation, other hypothetical factors such as desensitization, physiological exhaustion or PAA-mediated endocrine disruption could potentially explain the reduction in the corticosteroid response after repeated/prolonged PAA exposure. In this study, rainbow trout that had...

Linking water treatment practices and fish welfare Organic Eprints
Zubiaurre, Claire; Pedersen, Lars-Flemming.
Peracetic acids can be used as sanitizers to control water quality in aquaculture systems. As an alternative to formalin, chloramine-T or copper sulphate, PAA has strong anti-microbial effects, degrades quickly and is relatively safe to use. Its mode of action and associated rapid decay can make optimizing treatment protocols a challenge. Continuous low-dose applications seem to be a promising solution. In this preliminary study behavioral response was used to assess potential correlations with PAA dosage. A behavioral change or response is not necessarily an indication of compromised welfare. Supportive enzymatic, biochemical and physiological biomarkers can be used along with gill and epidermal histological measures to evaluate the effects on water...

Promising water treatment practices with peracetic acid products Organic Eprints
Pedersen, Lars-Flemming; Jokumsen, Alfred; Larsen, Villy J.; Henriksen, Niels Henrik.
Peracetic acid products can be used as sanitizers to control water quality in aquaculture systems. Here we describe recent practical experiences of applying peracetic acid (PAA) as an alternative to formalin or sodium chloride. PAA has strong antimicrobial effect, it is easy degradable and relatively safe to use work-wise. Being a strong oxidant, PAA resembles ozone in its mode of action and the associated rapid decay. This can be a challenge towards optimizing treatment protocols, and continuous low dose applications seem to be a promising solution. PAA is listed among the few disinfectants to be approved to be applied in organic aquaculture where formalin is prohibited (EU, 2014a). As of January 2016, organic trout production fully depends on organic...

”Pulse vs. Continuous Treatment: which is better for applying peracetic acid in RAS?” Organic Eprints
Liu, Dibo; Pedersen, Lars-Flemming; Lazado, Carlo Cabacang; Strauss, David L.; Meinelt, Thomas.
Peracetic acid (PAA) can improve water quality in a way that does not affect the fish, the farmer nor the environment. This presentation included recent findings of ways to improve PAA water treatment strategies. The study compared two distinct way of applying PAA – either as a pulse or as low dose continuous addition. Selected markers to address potential negative impact on fish welfare were used. Cortisol expression was temporarily increased following pulse addition but subsequently fish adapted and cortisole expression remained at low, similar levels as measured among control fish and fish exposed to continuous low dose PAA exposure. Pros and cons about PAA application and its consequences were discussed as well.

ROBUSTFISH: NEW POSSIBILITIES FOR GROWTH AND ROBUSTNESS IN ORGANIC AQUACULTURE Organic Eprints
Jokumsen, Alfred; Høglund, Erik; Lund, Ivar; Madsen, Lone; Pedersen, Lars-Flemming; Nielsen, Max; Nielsen, Thorkild; Larsen, Villy J.; Larsen, Erling P..
The aim of Robustfish project is to answer the following research questions: (1) Is it possible to select robust rainbow trout fry early in development before they enter the more cost intensive steps in the production cycle? (2) Does sustainable (non-fish based) dietary lipid sources affect stress resilience and immune-competence of the fry? (3) Related to (1) and (2): Is improved resistance to RTFS achieved? (4) How will the market respond to a significant increase in production of organic trout, i.e. consumer preferences and competiveness by companies in the value chain? RobustFish will strengthen the development of Danish organic trout production. In this respect the robustness of the fry to diseases is crucial, i.a. RTFS. The robustness of the fry...

Udvikling af dansk økologisk yngelopdræt Organic Eprints
Larsen, Villy J.; Henriksen, Niels Henrik; Pedersen, Lars-Flemming; Jokumsen, Alfred.

The pros and cons of sodium percarbonate Organic Eprints
Pedersen, Lars-Flemming; Jokumsen, Alfred.
The article deals with the use an approved aquaculture sanitizer: sodium-percarbonate (SPC).The substance is an easy to use powder containing hydrogen peroxide; when added to fish ponds hydrogen peroxide gets dissolved and subsequently decomposed into oxygen and water. This article presents the results from measurements in January 2017 in connection with practical application at an organic fish. The study showed that SPC leads to an evenly distribution of hydrogen peroxide in the water and maintained for a few hours. The water treatment causes oxygen liberation at the same time but also a significant pH increase which has to be taken into account for optimal management.
