Electrophoretic characterization of Aspergillus nidulans strains with chromosomal duplications Genet. Mol. Biol.

Pulsed-field gel electrophoresis was used to characterize strains of Aspergillus nidulans with a chromosomal duplication Dp(I-II). Morphologically deteriorated and improved variants of these strains were also analyzed. The electrophoretic karyotype demonstrated that in two duplicated strains (A and B) the 4.2 Mb band, which corresponds to chromosome II, was absent and a new band was observed. Hybridization studies using the uapA (chromosome I) and wA (chromosome II) genes demonstrated that the new band corresponded to chromosome II plus the duplicated segment of chromosome I. Resistência a benzimidazóis por Guignardia citricarpa PAB

O objetivo deste trabalho foi avaliar a resistência de Guignardia citricarpa aos fungicidas carbendazim e piraclostrobina, por meio de avaliação do crescimento em meio de cultura, decomposição de tecido foliar e produção de corpos de frutificação. Para isso, o fungo G. citricarpa foi isolado de lesões de frutos de laranja (Citrus sinensis), produzidos em área com intensa aplicação de fungicida. Os isolados obtidos foram avaliados quanto à sensibilidade aos fungicidas piraclostrobina e carbendazim, nas dosagens de 0,5, 1 e 2 µg mL-1 de i.a., para se verificar o efeito da pressão de seleção causada pelo uso destes compostos em áreas citrícolas. Characterization of fusion products from protoplasts of yeasts and their segregants by electrophoretic karyotyping and RAPD Rev. Microbiol.

In order to characterize fusion products from yeast protoplasts and their segregants, with important features to the wine making industry, electrophoretic karyotyping and RAPD (Random Amplified Polymorphic DNA) were utilized. Electrophoretic karyotyping was performed by the CHEF ("contour-clamped homogeneous electric field electrophoresis") method, which allowed the detection of chromosomal band complementation in fusion products and the presence of patterns of both parental and intermediary strains in segregants. By utilizing two primers, an amplification pattern of DNA fragments was obtained. Conidiogenesis of deteriorated variant of the strain Abnc of Aspergillus nidulans Scientia Agricola

The Abnc strain of A. nidulans carries the bncA1 gene (binucleated conidia), which induces the formation of binucleate and trinucleate conidia, displaying a chromosome I duplicated area and shifted to the chromosome II (I->II), and bringing forth genetic instability with degenerated sectors. This work has considered in a cytological level the conidiogenesis of the deteriorated variants isolated from the Abnc strain of A. nidulans, observing the event at the level of structural alterations, which composes the conidiophore, and the variations in the number of sterigmata and conidia nuclei. Genetic variability and vegetative compatibility of Erythricium salmonicolor isolates Scientia Agricola

The Pink Disease is caused by Erythricium salmonicolor, which attacks broad hosts, such as citrus, coffee, rubber, Eucalyptus spp. and Acacia spp., infecting mainly branches. This disease became a serious problem in Brazil, reducing the citrus production up to 10%. However the genetic diversity and compatibility of the fungus E. salmonicolor from Brazilian citrus plants is not yet evaluated. Pulsed field gel electrophoresis reveals chromosome length and number differences in Brazilian strains of Metarhizium Anisopliae BABT

Electrophoretic karyotypes of eight wild-type strains of Metarhizium anisopliae var. anisopliae were obtained by pulsed-field gel electrophoresis. These strains were isolated from insects of six different Brazilian states. The chromosomal DNA molecules of three strains were separated into seven bands and of five strains into eight bands. Chromosome length polymorphisms were also observed. The size of the chromosomal DNA of all strains varied between 7.7 and 0.9 Mb using the Aspergillus nidulans chromosomes as size standards. The total genome size of these strains was estimated in at least 29.7 Mb. Some correlations between differences in karyotype and occurrence of parasexual cycle likewise the host specificity were discussed.

Genetic characterization of somatic recombination in Trichoderma pseudokoningii BJM

Crossing experiments via hyphal anastomosis between two strains contrasting for auxotrophic markers of Trichoderma pseudokoningii were conducted to characterize the somatic recombination process in this specie. Four crossings were made and a total of 1052 colonies obtained from conidial suspensions of the heterokaryotic colonies were analyzed. Sixty-eight recombinant colonies, from four growing generations, were analyzed for the auxotrophic markers. Of the 68 colonies analyzed, 58 were stable after four generations and the remainders were unstable, reverting to one of the parentals. Limited vegetative compatibility as a cause of somatic recombination in Trichoderma pseudokoningii BJM

With the aim of a better characterization of the somatic recombination process in Trichoderma pseudokoningii, a progeny from crossings between T. pseudokoningii strains contrasting for auxotroph markers was characterized by RAPD markers and PFGE (electrophoretic karyotype). Cytological studies of the conidia, conidiogenesis and heterokaryotic colonies were also performed. The genotypes of the majority of the recombinant strains analyzed were similar to only one of the parental strains and the low frequency of polymorphic RAPD bands suggested that the nuclear fusions may not occur into the heterokaryon. Isolation of herpotrichiellacious fungi from the environment BJM

Herpotrichiellaceous fungi, common agents of chromoblastomycosis and phaeohyphomycosis, were searched in samples of rotten wood, leaf littler, bark and soil of the rhizosphere, collected in the Centro Nacional de Pesquisas de Florestas/EMBRAPA, Colombo, PR, Brazil. Morphological analyses of macro, optic and scanning electron microscopy, as well as the determination of the nutritional pattern of the isolated strains were carried out for a taxonomical study.
