Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (Banc de Granville and zone X). In 2004, no sample couId be analysed to these two areas because of the difficulty to obtain flat oysters (sparse oyster beds,...

REMONOR : résultats 2005. Evaluation de la mortalité, croissance et qualité des huîtres creuses ArchiMer
Mary, Charlotte; Pien, S; Blin, J.l.; Ropert, Michel.
The regional monitoring network REMONOR, relying on the experience of IFREMER's REMORA national network, each year assesses the mortality, growth and quality for two age classes ("adults" = 2 years and "juveniles" = 1 year) of cup oysters, distributed among various farm sites in the main oyster farming sectors of Lower Normandy. This network, led in collaboration by LERN (Environment Resources Laboratory) and SMEL (Coastline Installations Public-Private Entity), this year welcomes a new farm site in Veules les Roses, in Seine Maritime (76), bringing to 18 the total number of monitored sites. For operational reasons, a reduced monitoring protocol was conducted in 2005 on this last farm site, which will not take on the network's standardised protocol until...

ReMoNor : Mise en place et premier bilan du Reseau Mollusque de NORmandie (1998-1999) ArchiMer
Ropert, Michel; Blin, J.l.; Cornette, Florence; Pacary, S; Pien, S; Leganeur, E; Kopp, Joel; Richard, O.
Since 1998, a growth and quality monitoring study of reared molluscs has been developped as part of a close collaboration between the Shellfish Research Laboratory of Normandy (IFREMER Port-en-Bessin, Calvados) and the SMEL (Syndicat Mixte d'Expansion du Littoral) Experimental Centre (Blainville, Manche). REMONOR (REseau MOllusque de NORmandie) is based on the experience gained through the national monitoring network REMORA, which protocol has been entirely applied. Adults and spat of Pacific oysters Crassostrea gigas were placed in 13 stations all along the oysters rearing areas of Normandy. These oysters had the same origin and history (i.e. age and weight) until they were dispatched on the 13 stations. 4 essential criteria were used to monitor rearing...

Effects of age and environment on survival of summer mortality by two selected groups of the Pacific oyster Crassostrea gigas ArchiMer
Degremont, Lionel; Boudry, Pierre; Ropert, Michel; Samain, Jean-francois; Bedier, Edouard; Soletchnik, Patrick.
This study investigated the effects of age (first or second summer of life) and environment (three study sites on French coasts) on summer mortality in Crassostrea gigas, using three generations of oysters produced during the Morest project. For each generation, two groups of oysters, selected either as 'resistant' (R) or 'susceptible' (S) to summer mortality at the age of 6 months in Rivière d'Auray (RA, South Brittany), were evaluated. In Ronce (Marennes-Oléron Bay) and RA, R oysters had low mortality during their first and second summers, while mortality was higher during the second summer in Baie des Veys (BDV, Normandy). In RA, S oysters exhibited higher mortality during their first summer than during their second, while the opposite result was found...

Complex interactions between oyster, environment and pathogens were observed during the MOREST (2001-2005) project on summer mortality events of Crassostrea gigas oysters in France. The talk will show how nutrition level increasing reproductive intensity generates a risk for pathogen infection under synergistic effect of temperature and stress. Moreover, a genetic component is associated to specific reproductive performances and spawning behaviour. To better understand origin of such resistance or susceptibility to summer mortality, other characteristics as stress susceptibility, energy management and hemocyte activities of oysters from the two phenotypes were compared. This first comparison suggests interactions between stress, reproduction and defence...

Genetic basis of summer mortality in juvenile cupped oysters ArchiMer
Degremont, Lionel; Boudry, Pierre; Bedier, Edouard; Ropert, Michel; Soletchnik, Patrick; Huvet, Arnaud; Moal, Jeanne; Samain, Jean-francois.
The French project "Morest", coordinated by IFREMER, aims to understand the causes of the summer mortalities in Crassostrea gigas. In 2001, three sets of families were bred following a nested half-sib mating design. 17 half-sib families (HSF) were obtained in this first generation (G1) and reared in 3 sites. Significant differences in survival were observed among HSF, and some HSF showed high levels of mortality in all sites, clearly indicating a genetic basis for survival. In 2002, a second generation (G2), including divergent selection and inbred lines, was constituted. Monitoring of survival and growth of G2s were the same as in 2001. Significant differences in survival were found between the offspring of the "high" and "low" selected groups and between...

A latitudinal gradient of seasonal temperature variation recorded in oyster shells from the coastal waters of France and The Netherlands ArchiMer
Lartaud, Franck; Emmanuel, Laurent; De Rafelis, Marc; Ropert, Michel; Labourdette, Nathalie; Richardson, Christopher A.; Renard, Maurice.
Cathodoluminescence (CL) microscopy of the foliated calcite shell hinge sections of live-collected oyster Crassostrea gigas collected at seven locations along a latitudinal gradient from the Netherlands in the North Sea to the Atlantic coast of France, revealed variations in luminescence that were attributable to seasonal variations in calcification of the hinge. Photomicrographs of hinge sections and luminescence profiles were analyzed to define a micro-sampling strategy that was adopted to drill the hinge samples to determine their isotopic composition. Reconstructed seasonal seawater temperatures determined from the stable oxygen isotope (delta O-18) composition along growth profiles from 32 oyster shell hinges showed distinct seasonal isotopic cycles...

Caractérisation et déterminisme du développement d'une population de l'annélide tubicole Lanice conchilega (Pallas, 1766) (Polychète Terebellidae) associé à la conchyliculture en Baie des Veys (Baie de Seine Occidentale) ArchiMer
Ropert, Michel.
Since the mid 80's, a tidal population of the polychaete Lanice conchilega has proliferated along the eastern part of the "Baie des Veys" (Western part of the Bay of Seine). Population density 7 000 individuals per square meter. This proliferation affected significantly the local oyster farming industry, by resulting increased sedimentation rates. This work try to improved understanding the phenomenon by analyzing at various temporal and spatial scales the ecosystem and its relationship with the Lanice conchilega population dynamics. Several field works were carried out (i.e. bottom type mapping, topography, hydrology) to analyse the occurrence and development of this population taking into account the overall changes of the "Baie des Veys" ecosystem....

Effect of environmental and nutritive conditionings on defense mechanisms of oyster Crassostrea gigas during an annual cycle ArchiMer
Delaporte, Maryse; Moal, Jeanne; Soudant, Philippe; Lambert, Christophe; Pouvreau, Stephane; Enriquez, Martha; Degremont, Lionel; Soletchnik, Patrick; Gagnaire, Beatrice; Ropert, Michel.
In the frame of MOREST project, a common biological material, resulting of a mixture of different families produced in experimental hatchery, was reared in two different environmental fields : Normandy and Charente. Concomitantly, a pool was conditioned at the Ifremer Argenton hatchery with three different algae levels : 4%, 8% and 12% of algal dry weight per oyster dry weight. During the experiments, five immune parameters were studied in parallel with survival rate and reproductive status (stages and intensity). Site location, seasonal variations and experimental diet level clearly influenced oyster immune parameters. Haemocyte counts were higher for oysters reared in Normandy than those reared in Charente and in hatchery. Granulocyte percentage was...

Mn labelling of living oysters: Artificial and natural cathodoluminescence analyses as a tool for age and growth rate determination of C. gigas (Thunberg, 1793) shells ArchiMer
Lartaud, Franck; De Rafelis, Marc; Ropert, Michel; Emmanuel, Laurent; Geairon, Philippe; Renard, Maurice.
We developed a growth model for Crassostrea gigas oyster shells based on the use of in situ temporal manganese markings to calibrate natural cathodoluminescence (CL) changes in the shell hinge sections. A 30 min to 4-h exposure period with Mn2+ (90-120 mg l(-1)) was sufficient to create a detectable mark in the shells. This makes the Mn2+ markings the fastest mollusc shells marking technique to date. The natural CL from juvenile and adult shells cultured in four standard shellfish-farming locations along the English Channel and French Atlantic coasts, exhibited a seasonal pattern (maximum CL intensity occurring during summer periods, minimum CL intensity occurring during winter). Hydrobiological data recorded at Baie des Veys site allows us to attribute...

Spatio-temporal changes in mortality, growth and condition of the pacific oyster, Crassostrea gigas, in Normandy (France) ArchiMer
Royer, J; Ropert, Michel; Costil, K.
Summer mortalities have been observed in French shellfish areas (including Normandy) since 1994, but origin of this syndrome remains unclear and is suspected to be caused by a combination of several interacting extrinsic (biotic and abiotic) and intrinsic (genetic, physiological, immunological) factors. The French research program, MOREST aimed to identify the origin of oyster summer mortality along the French coast, focusing on the interactions between oysters, their pathogens, and the environment. The present study analyzed spatio-temporal variation in growth, condition and mortality in spat, and half-grown and market-sized oysters reared from February 2000 to October 2003 at six stations within two different areas in the Bay of Veys, Normandy: Grandcamp...

Summer mortality associated with juveniles of the oyster Crassostrea gigas is the result of a complex interaction between the host, pathogens and environment. Divergent selection based on summer survival have been applied to produce families with resistant and susceptible progeny. Here, we describe the use of suppression subtractive hybridization to generate 150 clones that were differentially regulated between resistant and susceptible F2 progeny. The nucleotide sequence of these 150 clones was determined. Among the 25 unique sequences that matched with the products of known genes, those putatively implicated in energy generation (12%) and immune function (16%), systems suspected to be implicated in summer mortality, were extended and analyzed by real...
