Diversity of bats species sent for rabies diagnosis to Pasteur Institute over the period from 2009 to 2012 MV&Z

Bats found in Brazil are classified in 9 families, 64 genus and 168 species. They are an important rabies reservoirs and all species can be infected or be the transmitter of rabies virus, independently of their specie classification or feeding habits. This work aimed a survey of bats species originated from counties of Sao Paulo state, which were sent to the diagnostic laboratory of the Pasteur Institute – Sao Paulo. Rabies is an infectious disease caused by a virus belonging to the family Lissavirus that affects all mammals including humans. Rabies transmission occurs by virus-containing saliva , through biting or licking of an injured tissue. The virus replicates at the site of inoculation until it reaches the nerve endings. At the neuromuscular junctions, the virus binds specifically to the nicotinic acetylcholine receptor. On the other hand, alkaloids have yielded interesting results in terms of drug development. Molecular techniques have been used increasingly as tools for the diagnosis by detecting the rabies virus genome. This study aimed to detect the presence of rabies virus in the wash of skull and in different organs of the genus Artibeus bats using the hemi-nested RT-PCR (hnRT-PCR) and Real Time RT-PCR molecular techniques. From approximately 4,000 specimens of bats received at the Institute Pasteur for rabies diagnosis, 30 bats of the genus Artibeus were selected, with records of positive results for rabies by the traditional techniques of direct fluorescent antibody test (FAT) and inoculation of murine neuroblastoma cell line (N2A). The bats are a major reservoir of rabies virus and have a relevant importance in the disease transmission. The aim of this study was to evaluate the bat population submitted for rabies diagnosis. Data were analyzed from 18.805 bats received by the diagnostic section of the Pasteur Institute of Sao Paulo, originated from several counties of the state of Sao Paulo over the period from 2007 to 2012. The Phyllostomidae family is the most numerous, followed by Vespertilionidae and Molossidae. The fluorescent anti-rabies conjugate (FARC) produced in Instituto Pasteur de São Paulo (IP-SP) is obtained by fluorescein isothiocyanate (FITC) conjugation with specific polyclonal antibodies purified and it recommended storage temperature of 4°C protected from light incidence. The aim of this study was to detect the rabies virus (RABV) presence in different organs of frugivorous bats using molecular techniques such as RT‐PCR, hnRT‐PCR, and the Real Time RT‐PCR. Thirty bats of the genus Artibeus were selected and resulted as positive by the DFA test and N2A‐cells inoculation test using brain tissue in both tests. Samples of salivary gland tissue, urinary bladder tissue, kidney tissue, lung tissue, stool, and skull lavage were collected for molecular assays. The Fluorescent Antibody Virus Neutralization Test (FAVN) is used routinely in many laboratories for the reference measurement of rabies virus neutralizing antibodies (VNA) in serum of animals to confirm the efficacy of vaccination against rabies, which is required to authorize the transit of these animals in countries free of rabies.
