Laser biosensor use for the microbial metabolic activity assessment of kefir vinegar PAB

Abstract: The objective of this work was to verify the use of a laser biosensor for the microbial metabolic activity assessment of kefir vinegar. Kefir grains were inoculated in apple must and were analyzed daily using the biospeckle technique. The initial biological activity was of 14.21 pixels per absolute value of differences (AVD). The biological activity of kefir grains decreased in the early days of fermentation and increased on the fourth day to 11.51 pixels per AVD; however, on the last day, the biological activity of kefir grains decreased to 7.12 pixels (alcoholic fermentation) and to 6.77 pixels per AVD (acetic fermentation). This new biospeckle methodology facilitates the microbiological control in fermentation processes.

The objective of this study was to evaluate the effect of bacterial inoculants at two inoculation rates on chemical and biological characteristics of maize silage. The treatments consisted of two inoculating rates (5 and 6 log cfu g-1of forage) for each strain of lactic acid bacteria (LAB) identified as Lactobacillus buchneri, L. hilgardii, or L. plantarum. The maize was ensiled in experimental PVC silos. The objective of this research was to evaluate the effect of the addition of two Lactobacillus strains on the aerobic stability of the sugar cane (Saccharum spp.) silages. The experiment was carried out in a completely randomized design with three replicates in split-plot in time scheme, so that the treatments (silages without inoculant and with novel strain or commercial inoculants) were randomized on plot and in times of aerobic evaluation of the silages (zero, two, four, and six days after the opening of the silos) as subplots. The mushroom Agaricus blazei Murrill was discovered in Piedade, state of São Paulo, Brazil, and sent to Japan to be studied for its medicinal properties. Studies in guinea pigs revealed antitumor properties, triggering Japanese importation of A. blazei from Brazil. Because of its high price on the international market, many companies and rural growers produce A. blazei as alternative crop to increase income, but because interest in this mushroom occurred suddenly there has not been enough time for the scientific community to investigate it and, technology used for its cultivation is still based on empirical rules. ABSTRACT Whole-plant corn silage (WPCS) is a commonly used forage for feeding dairy cattle in Brazil. The aim of this study was to evaluate on-farm ensilaging practices and the nutritional value, microbiology and fermentation profiles of WPCS from dairy farms in the south of the state of Minas Gerais, Brazil. Silages from 54 dairy farms located in nine cities were sampled and a questionnaire was distributed for completion to describe the herd profile and ensilage practices. Herd size ranged from 8 to 1,000 lactating cows, and 52 % of the farms had fewer than 50 cows in lactation. Incidence and distribution of filamentous fungi during fermentation, drying and storage of coffee (Coffea arabica L.) beans BJM

The objective of this work was to isolate and characterize filamentous fungi present in different stages of harvest, fermentation, drying and storage of coffee beans processed by natural method. The cherries were hand-picked and then placed on a cement drying platform where they remained until reached 11% of humidity. Microbial counts were found in all samples during fermentation and drying of the coffee beans. Counts of fungi in the coffee cherries collected from the tree (time 0) were around 1.5 x 10³ CFU/g. This number increased slowly during the fermentation and drying reaching values of 2 x 10(5) CFU/g within 22 days of processing. Evaluation of stress tolerance and fermentative behavior of indigenous Saccharomyces cerevisiae BJM

Sixty six indigenous Saccharomyces cerevisiae strains were evaluated in stressful conditions (temperature, osmolarity, sulphite and ethanol tolerance) and also ability to flocculate. Eighteen strains showed tolerant characteristics to these stressful conditions, growing at 42 ºC, in 0.04% sulphite, 1 mol L-1 NaCl and 12% ethanol. No flocculent characteristics were observed. These strains were evaluated according to their fermentative performance in sugar cane juice. The conversion factors of substrates into ethanol (Yp/s), glycerol (Yg/s) and acetic acid (Yac/s), were calculated. Isolation and identification of yeasts and filamentous fungi from yoghurts in Brazil BJM

Seventy-two cartons of yoghurt were sampled three times at monthly intervals from four different local manufacturers. Total counts were close to 6 x 10(7) cells g-1 of yoghurt. Yeast counts varied from 1 to 2,700 g-1. There was no evidence of systematic contamination at source but this longitudinal study revealed that ad hoc contamination and improper storage led to the higher yeast counts. Contamination was generally higher in the hotter months but was lower overall than reported from other countries. A total of 577 yeast isolates were identified belonging to ten species. Microbial ecology and chemical composition of Brazilian kefir beverage was performed. The microorganisms associated with Brazilian kefir were investigated using a combination of phenotypic and genotypic methods. A total of 359 microbial isolates were identified. Lactic acid bacteria (60.5%) were the major isolated group identified, followed by yeasts (30.6%) and acetic acid bacteria (8.9%). Bacaba beverage produced by Umutina Brazilian Amerindians: Microbiological and chemical characterization BJM

Bacaba chicha is a beverage prepared by the indigenous Umutina people from the bacaba fruit (Oenocarpus bacaba), a purple berry that is rich in fat and carbohydrates, as well as a source of phenolic compounds. In this study, samples of bacaba chicha beverage were collected, and the microbial community was assessed using culture-dependent and -independent techniques. The nutritional composition and metabolite profiles were analyzed, and species belonging to lactic acid bacteria (LAB) and yeasts were detected. Cultivo do cogumelo Pleurotus sajor-caju em diferentes resíduos agrícolas Ciência e Agrotecnologia

Diferentes resíduos agrícolas disponíveis na região sul de Minas Gerais foram testados para o cultivo do cogumelo Pleurotus sajor-caju. Foram avaliados os seguintes substratos: palha de feijão pura (PFP), palha de milho pura (PMP), casca de café pura (CCP), palha de feijão enriquecida com 2% de calcário, 2% de gesso e 10% de farelo de trigo (PFE), palha de milho enriquecida (PME) e casca de café enriquecida (CCE). Todos os substratos receberam 2% de inoculante e foram incubados a 24°C. Após a colonização, os sacos foram mantidos abertos em ambiente a 24°C e umidade a 80%. Com este trabalho objetivou-se avaliar a coloração final de basidiocarpos desidratados de Agaricus blazei, após serem submetidos a tratamento com ácidos ascórbico e cítrico. Para tanto, utilizaram-se cogumelos A. blazei, cultivados em substrato apropriado e mantidos em estufa. Os cogumelos foram colhidos com o píleo totalmente fechado e após a limpeza em água corrente, os basidiocarpos foram seccionados ao meio no sentido longitudinal, sendo uma metade submetida ao tratamento experimental e a outra metade representando o tratamento controle.
