Variation in the electrophoretic karyotype of Brazilian strains of Metarhizium anisopliae Genet. Mol. Biol.

Pulsed-field gel electrophoresis (PFGE) was used to separate chromosome-sized DNA molecules of four strains of Metarhizium anisopliae from Brazil. Metarhizium anisopliae isolates from Japan have been reported as possessing seven chromosomes. Variation was observed among the Brazilian strains and the chromosomal DNA was resolved into eight bands for strain CG46. Densitometric analysis of PFGE gels suggested that the other three Brazilian strains also possess eight chromosomes, with two chromosomes migrating as doublets under the electrophoretic conditions used. The genome size was estimated as varying between 23.39 to 31.88 Mb, not including possible doublet chromosomes.

Genetic variation in the cuticle-degrading protease activity of the entomopathogen Metarhizium flavoviride Genet. Mol. Biol.

Extracellular proteases have been shown to be factors of virulence in fungal entomopathogenicity. We examined the production of the cuticle-degrading extracellular proteases chymoelastase (Pr1) and trypsin (Pr2) in isolates of the fungus Metarhizium flavoviride. Fungal growth was in a mineral medium (MM) containing nitrate, and in MM supplemented with either cuticle from Rhammatocerus schistocercoides or with the non-cuticular substrate casein. The substrates used for growth influenced the expression of both analyzed proteases, the highest protease activities of nearly all isolates having been observed in the medium containing insect cuticle, with more Pr1 than Pr2 being produced. Survival in soil and detection of co-transformed Trichoderma harzianum by nested PCR PAB

The objective of this work was to evaluate the survival of two Trichoderma harzianum co-transformants, TE 10 and TE 41, carrying genes for green fluorescent protein (egfp) and for resistance to benomyl, during four weeks in a contained soil microcosm. Selective culture media were used to detect viable fungal material, whose identity was confirmed by the observation of the fluorescent phenotype by direct epifluorence microscopy. PCR using two nested primer pairs specific to the egfp gene was also used to detect the transformed fungi. Production of hydrolytic enzymes by Trichoderma isolates with antagonistic activity against Crinipellis perniciosa, the causal agent of witches' broom of cocoa BJM

Two isolates of Trichoderma, which reduce the incidence of witches'broom disease caused in cocoa by Crinipellis perniciosa, were evaluated for their potential to produce hydrolases in liquid medium. Very low or no hydrolytic activity was produced in the absence of any substrate. The activities of chitinase, N-acetylglucosaminidase, beta-1,3-glucanase, total cellulase, endoglucanase, aryl- beta-glucosidase, beta-glucosidase, protease and amylase increased dramatically within 72-120 h of growth in the presence of specific substrates. Except for N-acetylglucosaminidase and beta-glucosidase Trichoderma harzianum isolate 1051 produced the largest amounts of hydrolases.
