A Casearia sylvestris (Flacourtiaceae) é uma planta popularmente conhecida como "guaçatonga" e é usada por povos indígenas da América do sul (Brasil, Peru e Bolivia) no tratamento de muitas doenças, incluindo câncer. Estudos citotóxicos mostraram que esta planta apresenta um possível e interessante potencial antitumoral devido à presença de moléculas chamadas casearinas. Além disso, a composição do óleo essencial mostrou uma alta concentração de sesquiterpenos de alto potencial citotóxico. Cytotoxic evaluation of essential oil from Zanthoxylum rhoifolium Lam. leaves Acta Amazonica

Zanthoxylum rhoifolium Lam is a plant popularly used as antimicrobial, for malaria and inflammatory treatment. The essential oil of Z. rhoifolium was extracted and its cytotoxic effects against HeLa (human cervical carcinoma), A-549 (human lung carcinoma), HT-29 (human colon adenocarcinoma), Vero (monkey kidney) cell lines and mice macrophages were evaluated. Some of the terpenes of its essential oil (ß-caryophyllene, alpha-humulene, alpha -pinene, myrcene and linalool) were also tested to verify their possible influence in the oil cytotoxic activity. Antimicrobial activity of bergenin from Endopleura uchi (Huber) Cuatrec Acta Amazonica

Endopleura uchi (Huber) Cuatrec. is an Amazon species traditionally used as treatment for inflammations and female disorders. Bergenin was isolated from ethyl acetate fraction of bark of E. uchi by using column chromatography over sephadex LH-20 and then silica gel 60 flash. Its structure was identified on the basis of its NMR spectra. The antimicrobial activity of bergenin and fractions of methanol extract of E. uchi were evaluated against ATCC microorganisms (Escherichia coli, Salmonella enteritidis, Pseudomonas aeruginosa, Enterococcus faecalis, Staphylococcus aureus, Candida albicans, C. guilliermondii, Aspergillus flavus, A. nidulans). Plesiomonas shigelloides exports a lethal cytotoxic-enterotoxin (LCE) by membrane vesicles BJID

ABSTRACT Plesiomonas shigelloides isolated from water in Brazil was previously described as a hemorrhagic heat-labile cytotoxic-enterotoxin producer. We purified this toxin from culture supernatants using ion metallic affinity chromatography (IMAC) followed by molecular exclusion chromatography. The pure toxin presented molecular mass of 50 kDa and isoelectric point (pI) around 6.9 by 2D electrophoresis. When injected intravenously, the purified cytotoxic-enterotoxin induced also severe spasms followed by sudden death of mice. Hence, we entitled it as lethal cytotoxic-enterotoxin (LCE). Cytotoxic factor secreted by Escherichia coli associated with sepsis facilitates transcytosis through human umbilical vein endothelial cell monolayers BJID

Abstract Culture supernatant of sepsis-associated Escherichia coli (SEPEC) isolated from patients with sepsis caused loss of intercellular junctions and elongation of human umbilical vein endothelial cells (HUVEC). The cytotoxic factor was purified from culture supernatant of SEPEC 15 (serogroup O153) by liquid chromatography process. PAGE (polyacrylamide gel electrophoresis) showed that the purified SEPEC cytotoxic factor had a molecular mass of ∼150 kDa and consisted of at least two subunits. At the concentration of 1 CD50 (40 μg/mL) did facilitate transcytosis through the HUVEC cells monolayer of SEPEC 15 as much as E. coli K12 within 30 min without affecting cell viability. Hemagglutinating properties of Salmonella enterica serovar Enteritidis isolated from different sources BJM

Twenty-five strains of Salmonella enterica serovar Enteritidis isolated from different sources were examined for hemagglutinating activity. Bacteria cultured in different media induced hemagglutination of human erythrocytes, but no reaction was observed with erythrocytes from other animal species. The hemagglutinating expression activity was better for cultures on CFA agar at 37ºC than other conditions examined. The hemagglutination was inhibited by D-mannose, D-mannitol, melibiose, D-raffinose, L-rhamnose and sucrose. The absence of cell-surface appendages in electron microscope examinations suggested a nonfimbrial hemagglutinin. Virulence-associated characteristics of Enterococcus faecalis strains isolated from clinical sources BJM

Thirty-two clinical isolates of Enterococcus faecalis were screened for virulence factors. Twenty-four (75%) isolates produced hemolysin on Mueller-Hinton blood agar plates with sheep erythrocytes. However, the cell free heat-stable hemolysin was detected in all isolates (100%) of E. faecalis when grown in BHI-GA (BHI medium supplemented with 1% glucose and 0.03% L-arginine), but not in BHI broth alone. Twenty-four isolates (75%) produced caseinase and 23 (71.9%) lipase, but none of the isolates produced gelatinase. Fifteen (46.9%) culture filtrates caused rounding and membrane alterations with blebbing formation followed by death in HeLa and HEp-2 cells, but not in Vero cells. Virulence factors of Escherichia coli isolated from calves with diarrhea in Brazil BJM

Two hundred and five Escherichia coli strains isolated from calves with diarrhea from mid-western Brazil were screened for the presence of virulence factors associated with bovine colibacillosis. One hundred and two (49.8%) of the E. coli strains produced toxins: Shiga toxins 1 (9.7%) and 2 (6.3%), a-hemolysin (9.7%), enterohemolysin(6.8%), Cytotoxic Necrotizing Factors type 1 (0.5%), and type 2 (4.4%), enterotoxins LT-II (8.3%) and STa (3.9%). No strain produced enterotoxin LT-I. Fimbrial adhesins F5 and F17 were produced by 7.3% and 4.8% of the strains, respectivly, and none expressed F41. Curli fimbria: an Escherichia coli adhesin associated with human cystitis BJM

Abstract Escherichia coli is the major causative agent of human cystitis. In this study, a preliminary molecular analysis carried out by PCR (polymerase chain reaction) demonstrated that 100% of 31 E. coli strains isolated from patients with recurrent UTIs (urinary tract infections) showed the presence of the curli fimbria gene (csgA). Curli fimbria is known to be associated with bacterial biofilm formation but not with the adhesion of human cystitis-associated E. coli. Therefore, this work aimed to study how curli fimbria is associated with uropathogenic E. coli (UPEC) as an adhesion factor. Colibacillosis in lambs is associated to type I heat-stable enterotoxin in a farm in São Paulo State, Brazil Ciência Rural

Twenty seven (48.2%) culture supernatants of 56 Escherichia coli isolated from diarrheic lamb feces (7 to 10 days old) in São Paulo State, Brazil, presented positive results to suckling mice assay (fluid accumulation) but none caused cytopathic effects on Vero and CHO cells, indicating that these strains did not produced LT or VT toxins. PCR assays showed that these 27 E. coli strains harbored estA, that codifies for STa, but not for stx1, stx2 or cnf genes. The positive STa strains were checked for genes that codify for F41, F17 and K99 fimbriae, wich are considered colonization factors in ETEC. Only F17 was detect in two samples (7.4%).
