Background: Sperm capacitation is a process consists of a series of functional, biochemical, and biophysical modifications that render the ejaculated sperm competent for oocyte fertilization. Secreted by the female reproductive tract epithelium, heparin promotes capacitation by binding to and removing seminal plasma proteins, which are adsorbed to the sperm PM and would inhibit capacitation. There is substantial evidence that cryopreservation promotes capacitation-like changes in bull, ram and buck sperm. Abstract: The success of IVF and subsequent development of embryos are directly related to sperm selection and quality. This study aimed to evaluate the effect of different forces and time of centrifugation in mini-Percoll (MP) techniques for sperm selection on capacitation status and plasma membrane (PM) integrity of ram frozen-thawed sperm. Commercial semen from 10 rams of Santa Inês breed, aging 2-5 years old, were used. At post-thawing (PT), the PM integrity was evaluated by CASA using the SCA® system (Sperm Class Analyzer ? This study evaluated different methods of frozen-thawed sperm selection and capacita-tion in goats for further use in reproductive biotechnologies. In Experiment 1, semen wasprocessed by the following techniques: mini-Percoll, swim-up, or washing by centrifuga-tion. In Experiment 2, mini-Percoll selected-sperm was subjected to capacitation inductionby incubation with: 50 ug/mL heparin, 10 uM, 50 uM, or 100 uM of sodium nitroprusside(SNP). Abstract: Sperm capacitation is an essential event for fertilization; however, it decreases the sperm lifespan and viability. The aim of this study was to evaluate the effects of four sperm selection techniques on sperm capacitation and viability after incubation. A pool of frozen-thawed sperm from 10 Santa Inês rams was used. The samples were submitted to one of the following sperm selection techniques: sperm washing, Percoll gradient, mini-Percoll gradient, Swim-up and control group.
