Juvenile oysters (Crassostrea gigas) (produced in November 2009) reared under uniform hatchery conditions for 4 months were selected for extreme growth rate differences by repeatedly taking larger and smaller individuals to achieve weight differences > 30x between fast (F) and slow (S) growers. Mortalities of oyster seed of Crassostrea gigas associated with ostreid herpes virus OsHV-1 μVar have been observed in many oyster producing countries since 2008. The objective of this study was to further investigate whether low temperature treatments can offer a viable option to mitigate oyster mortalities. An experiment was set-up to further evaluate the effect of low temperature treatments (10 and 13°C vs. 21°C) and their duration (6 d to 83 d) on the survival of oysters previously infected with OsHV-1 μVar by means of exposure to field conditions in areas where mortalities were occurring. Pacific oyster Crassostrea gigas were inoculated with OsHV-1 at low load (control) or high load (challenged) to better understand the pathogenesis of ostreid herpesvirus 1 (OsHV-1 μVar) and to determine which metabolic pathways might be affected during infection. Animals were sampled for proteomic analysis two days post-injection, at the same time as OsHV-1 initiated an intense replication phase in challenged oysters. Twenty-five abundant protein spots that showed a marked change in accumulated levels were identified using a two-dimensional electrophoresis (2-DE) proteomic approach.
