Abstract Background: Studies on toad poison are relevant since they are considered a good source of toxins that act on different biological systems. Among the molecules found in the toad poison, it can be highlighted the cardiotonic heterosides, which have a known mechanism that inhibit Na+/K+-ATPase enzyme. However, these poisons have many other molecules that may have important biological actions. Therefore, this work evaluated the action of the low molecular weight components from Rhinella schneideri toad poison on Na+/K+-ATPase and their anticonvulsive and / or neurotoxic effects, in order to detect molecules with actions of biotechnological interest. Methods: Rhinella schneideri toad (male and female) poison was collected by pressuring their...

Posterior fossa infarct following Viper bite: a paradox J. Venom. Anim. Toxins incl. Trop. Dis.
Deepu,D; Hrishikesh,S; Suma,MT; Zoya,V.
Cerebral infarction after a viper bite is relatively uncommon. A combination of factors has been implicated in the pathophysiology of infarct following snakebite. In this case report, the clinical outcome after a posterior circulation infarct and various possibilities that could lead to such a catastrophic event are discussed. The present study stresses the need to keep hydration, blood pressure and central venous pressure optimal in all snakebite patients. Cerebral infarction should be considered a differential diagnosis, in any patient with neurological deterioration following snakebite. Prognosis of such patients with posterior circulation stroke remains poor and decompressive craniectomy has not been found to be helpful.

Dourado,DM; Matias,R; Almeida,MF; De Paula,KR; Vieira,RP; Oliveira,LVF; Carvalho,PTC.
The present study aimed to assess the effects of low-level laser (660 nm) on myonecrosis caused by the insertion of Bothrops neuwiedi venom in the gastrocnemius muscle of rats. Male Wistar rats were divided into three groups (n = 24 each): Group S (0.9% saline solution); Group V (venom) and Group VLLL (venom plus low-level laser). These categories were subdivided into four additional groups (n = 6) based on the euthanasia timing (3 hours, 24 hours, 3 days and 7 days). The groups V and VLLL were inoculated with 100 µL of concentrated venom (40 µg/mL) in the gastrocnemius muscle. The muscle was irradiated using a gallium-aluminum-arsenide laser (GaAlAs) at 35 mW power and 4 J/cm² energy density for 3 hours, 24 hours, 3 days or 7 days after venom inoculation....

Standardization of anti-lethal toxin potency test of antivenoms prepared from two different Agkistrodon halys venoms J. Venom. Anim. Toxins incl. Trop. Dis.
Lee,K. H.; Won,H. J.; Kim,S. N.; Yoo,S. H.; Shin,I. S.; Shin,K. H.; Hong,S. H.; Lee,S. H.; Min,H. K.; Park,S. N.; Hur,S. J..
In Korea, antivenoms for the treatment of patients bitten by venomous snakes have been imported from Japan or China. Although there is cross-reactivity between these antibodies and venoms from snakes indigenous to Korea (e.g. Agkistrodon genus), protection is not optimal. Antivenoms specifically prepared to neutralize Korean snake venoms could be more effective, with fewer side effects. To this end, we established an infrastructure to develop national standards and created a standardized method to evaluate the efficacy of two horse-derived antivenoms using mouse lethal toxin test. Additionally, we determined the antivenoms neutralizing activity against lethal doses (LD50) of Agkistrodon halys (from Japan) and Jiangzhe Agkistrodon halys (from China) venoms....

Rabies review: immunopathology, clinical aspects and treatment J. Venom. Anim. Toxins incl. Trop. Dis.
Consales,C. A.; Bolzan,V. L..
Among the diseases of viral origin, rabies is unique in its distribution and range of victims since it can afflict all warm-blooded animals. The interaction between the virus and the host population has facilitated the survival of the disease. The rabies virus (RV) has not changed in any significant way and has been capable of taking advantage of conditions suited to the continuance of rabies. Infection by RV is invariably lethal in the absence of protective immune response which, however, can contribute to the pathogenesis of rabies. Proinflammatory cytokines might affect, directly or indirectly, the levels of neurotrophins, growth factors, neurotransmitters and neurotoxins in the brain by activating glia, neurons, and vascular and immune cells. Although...

Morphometric characteristics and seasonal proximity to water of the Cypriot bluntnosed viper Macrovipera lebetina lebetina (Linnaeus, 1758) J. Venom. Anim. Toxins incl. Trop. Dis.
Jestrzemski,Daniel; Kuzyakova,Irina.
Abstract Background: The blunt-nosed viper Macrovipera lebetina (Linnaeus, 1758) is a medically important snake species in the Middle East. Its nominate subspecies Macrovipera l. lebetina is confined to Cyprus, where it is the only dangerously venomous snake species and heavily pursued. Despite the viper's large size, data on its body mass and sex-specific morphological differences are scarce. It is commonly believed that M. l. lebetina prefers freshwater proximity during summer. Hence, we aimed at investigating M. l. lebetina sex-specific morphological differences and its possible attraction to freshwater bodies in late summer. Methods: Morphometric characteristics, proximity to water and conservation status of M. l. lebetina were investigated in Paphos...

Interactions between environment, wild animals and human leptospirosis J. Venom. Anim. Toxins incl. Trop. Dis.
Ullmann,LS; Langoni,H.
Leptospirosis, a worldwide distributed zoononis caused by bacteria of the genus Leptospira (antigenically classified into serovars), may be direct or indirectly transmitted through infected urine or environment. Several domestic and wild animals are leptospirosis reservoirs. The disease presents occupational character since it is widely reported in professionals that work in humid environments - such as sewage workers and fishermen - and in places where rodents or susceptible animals are found, like slaughterhouses and veterinary clinics. In developing countries, outbreaks are related to lack of sanitation, overcrowding in inadequate housing and climatic conditions. In developed countries, sporadic cases occur in aquatic recreational activities including...

First serine protease inhibitor isolated from Rhinella schneideri poison J. Venom. Anim. Toxins incl. Trop. Dis.
Shibao,Priscila Y T; Anjolette,Fernando A P; Lopes,Norberto P.; Arantes,Eliane C..
Background Toad secretions are a source of molecules with potential biotechnological application on a wide spectrum of diseases. Toads from theRhinella family have two kinds of poisonous glands, namely granular and mucous glands. Rhinella schneideritoads produce granular secretions that comprise a great number of molecules, including serine proteases inhibitors. Serine proteases, such as trypsin, chymotrypsin and elastase, are enzymes that have a serine amino acid into its catalytic site and can be found in a large number of vertebrate species and pathogenic microorganisms. Therefore, the present work aims to purify a serine protease inhibitor from Rhinella schneiderigranular secretions.Findings This study presents the protocol used to purify a serine...

Canine trypanosomiasis: etiology of infection and implications for public health J. Venom. Anim. Toxins incl. Trop. Dis.
Eloy,LJ; Lucheis,SB.
Canine trypanosomiasis, caused by protozoans of the genus Trypanosoma, is divided into two primary types: the American form (Chagas disease), due to Trypanosoma cruzi infection, and the African form (sleeping sickness or surra), provoked by Trypanosomaevansi. This disease was originally enzootic and affected only wild animals, including mammals and birds, which served as reservoirs. Later, it spread to domestic animals such as horses, cattle and dogs. The disease became a zoonosis when contact between rural inhabitants and natural Trypanosoma foci occurred, due to ecological imbalances and increasing migration. Dogs are significantly involved in this context, because they are the main domestic animals and participate in the transmission and maintenance...

Efficacy of tannins from Mimosa pudica and tannic acid in neutralizing cobra (Naja kaouthia) venom J. Venom. Anim. Toxins incl. Trop. Dis.
Sia,FY; Vejayan,J; Jamuna,A; Ambu,S.
In the present study, the effectiveness of Mimosa pudica tannins (MPT) in neutralizing the lethality of Naja kaouthia venom was compared with commercially derived tannins. Preincubation of MPT with N. kaouthia venom maintained 100% survival of mice after 24 hours. The mouse group in which there was no preincubation, no protection against the effects of the venom was observed. M. pudica tannin was found to be more effective in neutralizing the lethality of N. kaouthia venom when compared to commercial tannic acid. Two protein spots were missing in the two-dimensional gel electrophoresis (2-DE) of the MPT treated mouse indicating the down-regulation of venom proteins. The results from this study indicated that tannins obtained from M. pudica are better than...

Ultramorphological analysis of the venom glands and their histochemical relationship with the convoluted glands in the primitive social paper wasp Polistes versicolor (Hymenoptera: Vespidae) J. Venom. Anim. Toxins incl. Trop. Dis.
Britto,F. B.; Caetano,F. H..
The venom glands are part of the most important defense weapon in Aculeata: the venom apparatus. The arrangement of these glands can vary among species, but in general they are composed of long secretory tubules connected to a muscular sac-like reservoir. Although the occurrence of these variations has been documented, many studies neglected the existence of a well-developed secretory portion in the lumen of the reservoir named convoluted gland. This study is an ultramorphological analysis of the venom glands and their histochemical relationship with the convoluted glands in the primitive social wasp Polistes versicolor. In this wasp, the venom glands are constituted by two tubular portions that penetrate individually in the venom reservoir, inside of...

Abstract Background: Lachesis muta rhombeata is one of the venomous snakes of medical importance in Brazil whose envenoming is characterized by local and systemic effects which may produce even shock and death. Its venom is mainly comprised of serine and metalloproteinases, phospholipases A2 and bradykinin-potentiating peptides. Based on a previously reported fractionation of L. m. rhombeata venom (LmrV), we decided to perform a subproteome analysis of its major fraction and investigated a novel component present in this venom. Methods: LmrV was fractionated through molecular exclusion chromatography and the main fraction (S5) was submitted to fibrinogenolytic activity assay and fractionated by reversed-phase chromatography. The N-terminal sequences of...

On the interference of clinical outcome on rabies transmission an perpetuation J. Venom. Anim. Toxins incl. Trop. Dis.
Brandão,PE.
Rabies is a viral zoonotic infectious disease that affects mammals and is caused by genotypes/species of the Lyssavirus genus (Rhabdoviridae, Mononegavirales), with the genotype 1 (classic rabies virus - RABV) being the most prevalent. Despite continuous efforts, rabies is still an incurable disease that causes thousands of deaths amongst humans worldwide. Due to a wide range of hosts and the different evolutionary paths of RABV in each host, several host-specific variants have arisen in an ongoing process. The result of RABV replication in nervous tissues may lead to two opposite clinical outcomes, i.e., paralytic/dumb form and encephalitic/furious one. The paralytic form creates dead-end hosts mainly amongst herbivores, while the furious form of the...
