Structural Modification of the Black Bengal Buck (Capra hircus) Acrosome During Post-testicular Maturation of Spermatozoa International Journal of Morphology

The present study was conducted to measure various biometric parameters of intact/normal acrosomes (AC) collected respectively from caput, corpus and cauda epididymis and vas deferens of Black Bengal buck. Giemsa stained acrosomes were measured after camera lucida drawings. Observations revealed dimensional characters of the acrosomal cap diminished gradually and significantly (p<0.01, p<0.05) during spermatozoa maturation phases in the different regions of the excurrent duct. Shape and size of the AC were also found to be influenced significantly (p<0.01, p<0.05) by the age and body weight of the animals. ABSTRACT An experiment was conducted to determine the effect of varying dietary supplemental inclusion levels of onion and garlic mixture on semen quality characteristics of Koekoek breeder cocks aged 35-41 weeks. The experimental diets were isocaloric and isonitrogenous but with different supplemental inclusion levels of onion and garlic. A complete randomized design was used for the experiment. The three dietary supplemental levels based on garlic and onion supplemental inclusion levels were Q0 (0 g onion and 0 g garlic per 600 g DM feed), Q1 (2.5 g onion and 2.5 g garlic per 600 g DM feed), and Q2 (5 g onion and 5 g garlic per 600 g DM feed) with each treatment having three replicates. The interaction between acrosome-reacted sperm and zona pellucida proteins is not yet fully understood. Serine protease acrosin and its zymogen proacrosin have been proposed to fulfill this function due to their capacity to bind zona pellucida glycoproteins. However, the molecular mechanism underlying this interaction has been merely speculative. Here we show that fucoidan (a sulfated polysaccharide) and solubilized zona pellucida glycoproteins, but not soybean trypsin inhibitor, are able to detach bound spermatozoa, which suggests that live sperm binds to the zona pellucida in a non-enzymatical way. Interestingly, mild proteolytic digestion with acrosin or trypsin does not modify the structure of the zona pellucida, but rather results in fewer spermatozoa...

Regulated exocytosis is controlled by internal and external signals. The molecular machinery controlling the sorting from the newly synthesized vesicles from the Golgi apparatus to the plasma membrane play a key role in the regulation of both the number and spatial location of the vesicles. In this context the mammalian acrosome is a unique vesicle since it is the only secretory vesicle attached to the nucleus. In this work we have studied the membrane trafficking between the Golgi apparatus and the acrosome during mammalian spermiogenesis. To validate a practical technique of simultaneous evaluation of the plasma, acrosomal and mitochondrial membranes in equine spermatozoa three fluorescent probes (PI, FITC-PSA and MITO) were associated. Four ejaculates from three stallions (n=12) were diluted in TALP medium and split into 2 aliquots, 1 aliquot was flash frozen in liquid nitrogen to induce damage in cellular membranes. Three treatments were prepared with the following fixed ratios of fresh semen: flash frozen semen: 100:0 (T100), 50:50 (T50), and 0:100 (T0). The objective of the present study was to investigate the effects of the direct addition of pentoxifylline (PF) to the ejaculates of men with poor sperm quality before freezing on post-thaw sperm motility, viability, acrosome integrity, and agonist-induced acrosome reaction. Semen specimens from 16 infertile men with impaired sperm count and motility (oligoasthenozoospermia) were divided into two equal aliquots: one received no treatment (control) while the other was incubated with 5 mM PF (treated).
