Action mechanisms involved in the bioprotective effect of Lactobacillus harbinensis K.V9.3.1.Np against Yarrowia lipolytica in fermented milk ArchiMer
Mieszkin, Sophie; Hymery, Nolwenn; Debaets, Stella; Coton, Emmanuel; Le Blay, Gwenaelle; Valence, Florence; Mounier, Jerome.
The use of lactic acid bacteria (LAB) as bioprotective cultures can be an alternative to chemical preservatives or antibiotic to prevent fungal spoilage in dairy products. Among antifungal LAB, Lactobacillus harbinensis K.V9.3.1Np showed a remarkable antifungal activity for the bioprotection of fermented milk without modifying their organoleptic properties (Delavenne et al., 2015). The aim of the present study was to elucidate the action mechanism of this bioprotective strain against the spoilage yeast Yarrowia lipolytica. To do so, yeast viability, membrane potential, intracellular pH (pHi) and reactive oxygen species (ROS) production were assessed using flow cytometry analyses after 3, 6 and 10 days incubation in cell-free supernatants. The tested...

Stylicins, a new family of antimicrobial peptides from the Pacific blue shrimp Litopenaeus stylirostris ArchiMer
Rolland, Jean-luc; Abdelouahab, Mahdia; Dupont, J.; Lefevre, F.; Bachere, Evelyne; Romestand, Bernard.
The present study reports the characterization of Ls-Stylicin1, a novel antimicrobial peptide from the penaeid shrimp, Litopenoeus stylirostris. The predicted mature peptide of 82 residues is negatively charged (theoretical pl=5.0) and characterized by a proline-rich N-terminal region and a C-terminal region containing 13 cysteine residues. The recombinant Ls-Stylicin1 has been isolated in both monomeric and dimeric forms. Both display strong antifungal activity against Fusarium oxysporum (1.25 mu M < MIC <2.5 mu M), a pathogenic fungus of shrimp, but lower antimicrobial activity against Gram () bacteria, Vibrio sp. (40 mu M < MIC <80 mu M). However, rLs-Stylicin1 is able to agglutinate Vibrio pennaeicidae in vitro in agreement with its potent...

Detection of griseofulvin in a marine strain of Penicillium waksmanii by ion trap mass spectrometry ArchiMer
Petit, Karina; Mondeguer, Florence; Roquebert, M; Biard, J; Pouchus, Y.
A marine strain of Penicillium waksmanii Zaleski was isolated from a sample of seawater from shellfish-farming area in the Loire estuary (France). The in vitro marine culture showed an important antifungal activity. Bioassay-guided fractionation was used to purify the crude extract. Dereplication by electrospray-ion trap/mass spectrometry (ESI-IT/MS) afforded the identification of the antifungal compound, after a semi-purification consisting of two stages. A comparison of the ionic composition between the active and the non-active fractions allowed the detection of a monocharged ion at m/z 353 containing a chlorine atom, which could be attributed to the antifungal griseofulvin [C17H17ClO6+H](+). Multi-stage fragmentation (MSn) confirmed the identity of the...

ANTIFUNGAL ACTIVITY SCREENING OF ANTARCTIC ACTINOBACTERIA AGAINST PHYTOPATHOGENIC FUNGI Acta biol.Colomb.
SANTOS,Andrés; NÚÑEZ-MONTERO,Kattia; LAMILLA,Claudio; PAVEZ,Mónica; QUEZADA-SOLÍS,Damián; BARRIENTOS,Leticia.
ABSTRACT The extreme weather conditions in the Antarctic have exerted selective pressures favoring differential features in bacteria to survive this untapped environment (i.e., antibiotic molecules). Notably, higher chances of antibiotic discovery from extremophiles have been proposed recently. Although new organic and environmentally friendly sources for helping in the control of plant pathogenic fungi are necessary, the information about anti-phytopathogenic applications of extremophile microorganisms from untapped environments is limited. In this study, we determined the antifungal effect of actinobacterial strains isolated from Antarctic soils and sediments. Co-culture inhibition assays and Minimum Inhibitory Concentration (MIC) determination revealed...
