REPARACIÓN DEL ADN: UNA POSIBLE RELACIÓN ENTRE LA DEFICIENCIA DE FOLATO Y LA MUERTE NEURONAL Acta biol.Colomb.
RAMÍREZ,NELSON J; ARBOLEDA,GONZALO; ARBOLEDA,HUMBERTO.

ACTIVIDAD ANTIPROLIFERATIVA DE ACEITES ESENCIALES DE PLANTAS CULTIVADAS EN COLOMBIA Acta biol.Colomb.
VELANDIA,Sindi Alejandra; QUINTERO,Elizabeth; STASHENKO,Elena E; OCAZIONEZ,Raquel E.

Apoptotic cell and phagocyte interplay: recognition and consequences in different cell systems Anais da ABC (AABC)
Moreira,Maria Elisabete C.; Barcinski,Marcello A..
Cell death by apoptosis is characterized by specific biochemical changes, including the exposure of multiple ligands, expected to tag the dying cell for prompt recognition by phagocytes. In non-pathological conditions, an efficient clearance is assured by the redundant interaction between apoptotic cell ligands and multiple receptor molecules present on the engulfing cell surface. This review concentrates on the molecular interactions operating in mammalian and non-mammalian systems for apoptotic cell recognition, as well as on the consequences of their signaling. Furthermore, some cellular models where the exposure of the phosphatidylserine (PS) phospholipid, a classical hallmark of the apoptotic phenotype, is not followed by cell death will be discussed.

Calcium and cell death signaling in neurodegeneration and aging Anais da ABC (AABC)
Smaili,Soraya; Hirata,Hanako; Ureshino,Rodrigo; Monteforte,Priscila T.; Morales,Ana P.; Muler,Mari L.; Terashima,Juliana; Oseki,Karen; Rosenstock,Tatiana R.; Lopes,Guiomar S.; Bincoletto,Claudia.
Transient increase in cytosolic (Cac2+) and mitochondrial Ca2+ (Ca m2+) are essential elements in the control of many physiological processes. However, sustained increases in Ca c2+ and Ca m2+ may contribute to oxidative stress and cell death. Several events are related to the increase in Ca m2+, including regulation and activation of a number of Ca2+ dependent enzymes, such as phospholipases, proteases and nucleases. Mitochondria and endoplasmic reticulum (ER) play pivotal roles in the maintenance of intracellular Ca2+ homeostasis and regulation of cell death. Several lines of evidence have shown that, in the presence of some apoptotic stimuli, the activation of mitochondrial processes maylead to the release of cytochrome c followed by the activation of...

Susceptible hosts: a resort for parasites right in the eye of the immune response Anais da ABC (AABC)
DOSREIS,GEORGE A..
Trypanosomatid protozoan parasites express an aggressive strategy of parasitism by infecting host macrophages and inducing extensive T-lymphocyte activation. One goal of such strategy is to drive the immune response of genetically susceptible hosts to a state of unresponsiveness regarding parasite killing. Unresponsiveness is achieved through different mechanisms, depending on the parasite species. In this brief review, recent findings on the molecular and cellular bases of the parasites' exploitation of host immune responses are discussed.

The T-cell anergy induced by Leishmania amazonensis antigens is related with defective antigen presentation and apoptosis Anais da ABC (AABC)
Pinheiro,Roberta O.; Pinto,Eduardo F.; Benedito,Alessandra B.; Lopes,Ulisses G.; Rossi-Bergmann,Bartira.
Leishmania amazonensis is the main agent of diffuse cutaneous leishmaniasis, a disease associated with anergic immune responses. In this study we show that the crude antigen of Leishmania amazonensis (LaAg) but not L. braziliensis promastigotes (LbAg) contains substances that suppress mitogenic and spontaneous proliferative responses of T cells. The suppressive substances in LaAg are thermoresistant (100ºC/1h) and partially dependent on protease activity. T cell anergy was not due to a decreased production of growth factors as it was not reverted by addition of exogenous IL-2, IL-4, IFN-gamma or IL-12. LaAg did not inhibit anti-CD3-induced T cell activation, suggesting that anergy was due to a defect in antigen presentation. It was also not due to cell...

Diosmin induces caspase-dependent apoptosis in human glioblastoma cells Anais da ABC (AABC)
SOARES,JULIANA M.; FARIA,BRUNA M. DE; ASCARI,LUCAS M.; ALVES-LEON,SONIZA V.; SOUZA,JORGE M. DE; SOARES,ANTONIO G.; CORDEIRO,YRAIMA; ROMÃO,LUCIANA F..
Abstract: Diosmin is a flavone glycoside clinically used as the main component of Daflon for the treatment of venous diseases. Several studies demonstrated that this natural compound can induce apoptosis in different tumors. However, isolated diosmin has not been studied regarding its effects on glioblastoma so far. Since glioblastoma is a highly lethal and fast-growing brain tumor, new therapeutic strategies are urgently needed. Herein, we evaluated the role of this flavonoid against glioblastoma cells using in vitro assays. Diosmin significantly reduced the viability of GBM95, GBM02, and U87MG glioblastoma cells, but not of healthy human astrocytes, as verified by MTT assay. Vimentin immunostaining showed that diosmin induced morphological changes in...

Antifungal and antitumor models of bioactive protective peptides Anais da ABC (AABC)
Rodrigues,Elaine G.; Dobroff,Andrey S.; Taborda,Carlos P.; Travassos,Luiz R..
Peptides are remarkably reactive molecules produced by a great variety of species and able to display a number of functions in uni-and multicellular organisms as mediators, agonists and regulating substances. Some of them exert cytotoxic effects on cells other than those that produced them, and may have a role in controlling subpopulations and protecting certain species or cell types. Presently, we focus on antifungal and antitumor peptides and discuss a few models in which specific sequences and structures exerted direct inhibitory effects or stimulated a protective immune response. The killer peptide, deduced from an antiidiotypic antibody, with several antimicrobial activities and other Ig-derived peptides with cytotoxic activities including antitumor...

Understanding the mechanisms of glutamine action in critically ill patients Anais da ABC (AABC)
Oliveira,Gisele P.; Dias,Cristina M.; Pelosi,Paolo; Rocco,Patricia R.M..
Glutamine (Gln) is an important energy source and has been used as a supplementary energy substrate. Furthermore, Gln is an essential component for numerous metabolic functions, including acid-base homeostasis, gluconeogenesis, nitrogen transport and synthesis of proteins and nucleic acids. Therefore, glutamine plays a significant role in cell homeostasis and organ metabolism. This article aims to review the mechanisms of glutamine action during severe illnesses. In critically ill patients, the increase in mortality was associated with a decreased plasma Gln concentration. During catabolic stress, Gln consumption rate exceeds the supply, and both plasma and skeletal muscle pools of free Gln are severely reduced. The dose and route of Gln administration...

Modulatory effects of rutin on biochemical and hematological parameters in hypercholesterolemic Golden Syrian hamsters Anais da ABC (AABC)
Kanashiro,Alexandre; Andrade,Daiani C.O.; Kabeya,Luciana M.; Turato,Walter M.; Faccioli,Lucia H.; Uyemura,Sérgio A.; Lucisano-Valim,Yara M..
Flavonoids have been reported to exhibit several pharmacological properties, mainly in cardiovascular and inflammatory diseases. In the present study, we observed that rutin, a known glycosylated flavonoid isolated from Dimorphandra mollis, had a lowering effect on plasma triglyceride levels of diet-induced hypercholesterolemic Golden Syrian hamsters, but did not change total cholesterol and high-density lipoprotein cholesterol levels. Moreover, high-fat or rutin supplemented diets showed no immunotoxic effects, since no significant changes were observed on total white blood cells, granulocytes and mononuclear cells, as well as on the neutrophil apoptosis degree, when compared to untreated animals. Therefore, rutin seems to be a selective and non-toxic...

AMIN,BAHAREH; ABNOUS,KHALIL; MOTAMEDSHARIATY,VAHIDEH; HOSSEINZADEH,HOSSEIN.
In our previous study, the ethanolic and aqueous extracts of Crocus sativus elicited antinociceptive effects in the chronic constriction injury (CCI) model of neuropathic pain. In this study, we explored anti-inflammatory, anti-oxidant and anti-apoptotic effects of such extracts in CCI animals. A total of 72 animals were divided as vehicle-treated CCI rats, sham group, CCI animals treated with the effective dose of aqueous and ethanolic extracts (200 mg/kg, i.p.). The lumbar spinal cord levels of proinflammatory cytokines including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β) and interleukin 6 (IL-6), were evaluated at days 3 and 7 after CCI (n=3, for each group). The apoptotic protein changes were evaluated at days 3 and 7 by western blotting....

India ink induces apoptosis in the yellow clam Mesodesma mactroides (Deshayes, 1854). Optical and ultrastructural study Anais da ABC (AABC)
CARVALHO,YURI B.M.; JETHRO,JUAN; POERSCH,LUIS H.; ROMANO,LUIS A..
ABSTRACT This paper reports on the acute inflammatory and cellular process in the yellow clam, Mesodesma mactroides, induced by injection of India ink into the muscular foot. Histological observations with optical and electronic microscopy were made at 24 and 48 h after injection. The induced cellular inflammatory response consisted of a general hemocyte infiltration without necrosis and apoptotic activity. Migration of ink-laden phagocytes across the intestinal epithelium was recorded. It appeared that the yellow clam "excreted" ink particles through the gill and kidney. The positive staining for apoptosis was observed in the digestive gland. Electronic microscopy revealed ultrastructural changes of endoplasmic reticulum stress and apoptotic bodies in the...

Inhibition of caspase-8 activity reduces IFN-gamma expression by T cells from Leishmania major infection Anais da ABC (AABC)
Pereira,Wânia F.; Guillermo,Landi V.C.; Ribeiro-Gomes,Flávia L.; Lopes,Marcela F..
Following infection with Leishmania major, T cell activation and apoptosis can be detected in draining lymph nodes of C57BL/6-infected mice. We investigated the mechanisms involved in apoptosis and cytokine expression following Tcellactivation. After two weeks of infection, apoptotic T cells were not detected in draining lymph nodes but activation with anti-CD3 induced apoptosis in both CD4 and CD8 T cells. Treatment with anti-FasLigand, caspase-8 or caspase- 9 inhibitors did not block activation-induced T-cell death. We also investigated whether the blockade of caspase-8 activity would affect the expression of type-1 or type-2 cytokines. At early stages of infection, both CD4 and CD8 T cells expressed IFN-gamma upon activation. Treatment with the...

Active caspase-3 expression levels as bioindicator of individual radiosensitivity Anais da ABC (AABC)
SANTOS,NEYLIANE F.G. DOS; SILVA,RAFAEL F.; PINTO,MARCELA M.P.L.; SILVA,EDVANE B. DA; TASAT,DEBORAH R.; AMARAL,ADEMIR.
ABSTRACT Several molecules and events involved in cell response to radiation-induced damage have been investigated towards a personalized radiotherapy. Considering the importance of active caspase-3 in the proteolytic cascade that ensures radiation-induced apoptosis execution, this research was designed to evaluate the expression levels of this protein as a bioindicator of individual radiosensitivity. Peripheral blood samples of 10 healthy individuals were gamma-irradiated (cobalt-60 source) with 1, 2 and 4 Gy (control: non-irradiated samples), and active caspase-3 expression levels were measured in lymphocytes, by flow cytometry, ex vivo and after different times of in vitro incubation (24, 48 and 72 hours). Short-term incubation of 24 h was the most...

Ouabain exacerbates activation-induced cell death in human peripheral blood lymphocytes Anais da ABC (AABC)
Esteves,Mabel B.; Marques-Santos,Luis F.; Affonso-Mitidieri,Ottília R.; Rumjanek,Vivian M..
Lymphocytes activated by mitogenic lectins display changes in transmembrane potential, an elevation in the cytoplasmic Ca2+ concentrations, proliferation and/or activation induced cell death. Low concentrations of ouabain (an inhibitor of Na+,K+-ATPase) suppress mitogen-induced proliferation and increases cell death. To understand the mechanisms involved, a number of parameters were analyzed using fluorescent probes and flow cytometry. The addition of 100nM ouabain to cultures of peripheral blood lymphocytes activated with 5µg/ml phytohemagglutinin (PHA) did not modify the increased expression of the Fas receptor or its ligand FasL induced by the mitogen. However, treatment with ouabain potentiated apoptosis induced by an anti-Fas agonist antibody. A...
