Burkholderia kururiensis is a diazotrophic bacterium originally isolated from a polluted aquifer environment and presents a high level of similarity with the rice endophyte "B. brasilensis" species. This work assessed the ability of B. kururiensis to endophytically colonize rice plantlets by monitoring different tissues of root-inoculated plants for the presence of bacterial growth in different media, electron microscopy and by 16S rDNA analysis. Observations of roots, stems and leaves of inoculated rice plantlets by electron microscopy revealed B. kururiensis colonization predominantly on root hair zones, demonstrating endophytic colonization primarily through the endodermis, followed by spreading into xylem vessels, a possible pathway leading to aerial... |

The effects of plant growth promoting rhizobacteria (PGPR) on the rooting and root growth of semi-hardwood and hardwood kiwifruit stem cuttings were investigated. The PGPR used were Bacillus RC23, Paenibacillus polymyxa RC05, Bacillus subtilis OSU142, Bacillus RC03, Comamonas acidovorans RC41, Bacillus megaterium RC01 and Bacillus simplex RC19. All the bacteria showed indole-3-acetic acid (IAA) producing capacity. Among the PGPR used, the highest rooting ratios were obtained at 47.50% for semi-hardwood stem cuttings from Bacillus RC03 and Bacillus simplex RC19 treatments and 42.50% for hardwood stem cuttings from Bacillus RC03. As well, Comamonas acidovorans RC41 inoculations indicated higher value than control treatments. The results suggest that these... |

This paper describes a protocol for the efficient vegetative propagation of Cleome rosea by somatic embryogenesis. Leaf and stem explants from nursery-grown seedlings of C. rosea were cultivated on Murashige and Skoog (MS) medium supplemented with indole-3-acetic acid (IAA), a -naphthaleneacetic acid (NAA), 4-amino-3,5,6-trichloropicolinic acid (picloram) or 2,4-dichlorophenoxyacetic acid (2,4-D). Nodular calli were produced from both explant types in the presence of 4.5 and 9.0 µM 2,4-D. Embryo development and maturation were achieved when calli from stem explants were transferred to media containing a ten-fold reduction of 2,4-D concentration initially used (0.45 and 0.90 µM). Leaf-derived calli did not form embryos with the same treatments. The highest... |

Endophytes are symptomless fungal or bacterial microorganisms found in almost all living plant species reported so far. They are the plant-associated microbes that form symbiotic association with their host plants by colonizing the internal tissues, which has made them valuable for agriculture as a tool in improving crop performance. Many fungal endophytes produce secondary metabolites such as auxin, gibberellin etc that helps in growth and development of the host plant. Some of these compounds are antibiotics having antifungal, antibacterial and insecticidal properties, which strongly inhibit the growth of other microorganisms, including plant pathogens. This article reviews the endophyte isolated from different plants, mode of endophytic infection and... |

This study attempted to clarify the effects of dark, light and ethylene on plant growth and endogenous levels of indole-3-acetic acid (IAA), cytokinins and abscisic acid in Catasetum fimbriatum. Dark-incubation fully inhibited root and pseudobulb formation as well as leaf growth, but favored shoot elongation. The results of continuous and active growth in dark-incubated shoots (stolons) were induced by strong apical meristem sink activity and by the significantly increased levels of cytokinins in shoots. In fact, shoot length, cytokinin and IAA levels in dark-incubated shoots were about twice as great as for those grown under light conditions. Moreover, the total cytokinin level in shoots of C. fimbriatum under light conditions without ethylene was... |

The in vitro culture of pineapple leaves on a shoot induction medium (SIM) results in the formation of protuberances and further development in shoots, and plantlets. The contents of endogenous indoleacetic acid (IAA) and five cytokinins (Cks), N6(2-isopentenyl)adenine (iP), N6(2-isopentenyl)adenosine (iPR), zeatin (Z), zeatin riboside (ZR) and N6-benzyladenine (BA), present in the basal portion of those leaves, were correlated to the organogenic response that occurs over 15 days of culture. The endogenous auxin/cytokinins ratio was lowest on the 3rd day, mainly due to a strong increase in the iP level. It seems that endogenous iP concentration triggered the induction signal for an organogenic response in pineapple leaf bases. The rise in iP content... |

Mango flowering involves hormonal regulation of shoot initiation and induction events resulting in reproductive shoot formation. A balance or ratio of endogenously regulated phytohormones, thought to be auxin from leaves and cytokinins from roots, appears to govern the initiation cycle independently from inductive influences. Induction of reproductive or vegetative shoots is thought to be governed by the ratio of a temperature-regulated florigenic promoter and an age regulated vegetative promoter at the time of shoot initiation. Management of off-season flowering in mango trees is being accomplished in the tropics by successfully synchronizing shoot initiation through tip pruning and use of nitrate sprays coupled with management of the stem age to induce... |

Jatropha curcas L. is a versatile crop since all its plant material is brought to use either as energy source, industrial or medicinal purpose. Several studies are ongoing in different parts of the world to optimize, enhance and exploit the growth, fruiting cycle and different developmental stages of the plant so that the economic yield of the plants can be utilized to the fullest limits. Foliar application of plant growth regulators such as ethrel (an ethylene releasing compound), indole acetic acid (IAA) and naphthalene acetic acid (NAA) at 50, 100 and 150 ppm, was found to influence different morpho-physiological characters in Jatropha curcas L, such as plant height, collar diameter, tree spread, flower initiation, number of inflorescence per plant,... |

Beilschmiedia berteroana ("belloto del sur") is a screrophyllous tree of the family Lauraceae that is endemic to the Central-South region of Chile. It is an endangered species; considered a second priority in the "Extinction Danger" category. This species is known for its timber, fruits and lots of mucilages, also has great potential for its size and ornamental evergreen foliage. This study had as a main objective to develop an in vitro rooting protocol of microshoots, obtained from the proliferation phase, by adding auxin: indole 3-butyric acid (IBA), naphthaleneacetic acid (NAA) and indole 3-acetic acid (IAA) in order to contribute to the recovery of the species. Results are discussed in the rooting phase, where this species is induced or maintained in... |

Background: Cyclamen persicum is an economically important ornamental crop that is propagated exclusively through seeds as vegetative propagation using cuttings is not possible. Therefore a micropropagation method through somatic embryogenesis is of high interest; however the method suffers from low reliability concerning quality and quantity of the produced plantlets. A crucial step of the protocol is the removal of plant growth regulators (PGRs) that triggers embryo development. In order to get a better insight in this crucial step of the propagation process, a gene expression analysis has been set up using five different genes of glutathione S-transferases (GST) as these are known to be auxin responsive as well as stress reactive. Results: One out of... |

This long-term study demonstrates for the first time that it is possible to propagate embryogenic cultures in pelargoniums and to subsequently initiate the differentiation of embryos using the cultivar Madame Layal (Pelargonium x domesticum). Propagation of callus was only possible with combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP), which gave rise to embryos from the primary culture stage on. However, the propagation of cells, as well as the differentiation of embryos, was inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking growth regulators was necessary before the differentiation of embryos occurred again. The consequences for improving the propagation... |
