Se probó el enraizado de estacas, acodo aéreo e injerto para clonar genotipos de un huerto semillero de P. leiophylla, putativamente resistentes al ataque de Dendroctonus sp. y Toumeyella pinicola Ferris. y que además muestran las mejores características fenotipicas. Con el primer método se clonó a la progenie, mientras que los otros métodos se desarrollaron para clonar a los árboles madre. En el enraizado de estacas se evaluó el efecto del substrato, tipo de estaca y concentración de ácido indolbutírico (AIB). Influence of 2,4-D and BAP on callus growth and the subsequent regeneration of somatic embryos in long-term cultures of Pelargonium x domesticum cv. Madame Layal Electron. J. Biotechnol.

This long-term study demonstrates for the first time that it is possible to propagate embryogenic cultures in pelargoniums and to subsequently initiate the differentiation of embryos using the cultivar Madame Layal (Pelargonium x domesticum). Propagation of callus was only possible with combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP), which gave rise to embryos from the primary culture stage on. However, the propagation of cells, as well as the differentiation of embryos, was inhibited by a continuous application of these growth regulators. A normalized embryoid cDNA library (EON) was constructed based on reassociation kinetics reaction. Results from dot blot hybridization and sequencing of EON cDNA clones clearly indicated that the normalization process reduced the frequency of high abundance transcripts and increased the frequency of low abundance gene transcripts. A total of 553 non-redundant expressed sequence tags (ESTs) were identified, 325 of these were not observed in the standard oil palm cDNA libraries sequenced previously. Abstract Auxin regulates a plethora of events during plant growth and development, acting in concert with other phytohormones. YUCCA genes encode flavin monooxygenases that function in tryptophan-dependent auxin biosynthesis. To understand the contribution of the YUCCA4 (YUC4) gene on auxin homeostasis, plant growth and interaction with abscisic acid (ABA) signaling, 35S::YUC4 seedlings were generated, which showed elongated hypocotyls with hyponastic leaves and changes in root system architecture that correlate with enhanced auxin responsive gene expression. Somatic embryogenesis and plant regeneration from zygotic embryo explants of onion Horticultura Brasileira

ABSTRACT: The study was undertaken to examine the effect of two synthetic auxins on somatic embryogenesis and plant regeneration from mature embryo explants of 16 onion cultivars. Cotyledons were removed from the embryos and remaining portions were cultured on MS medium supplemented with 1.0 mg/L 2,4-D and 5.0 mg/L picloram alone or in combination (1.0 mg/L 2,4-D + 2.5 mg/L picloram) to produce embryogenic callus. MS medium supplemented with 5.0 mg/L picloram was found to be the best one for both embryogenic callus induction (85%) and callus diameter (3.8 mm). This effect may be regarded, in part, as a thigmomorphogenic response, but gravimorphogenetic effect may also be involved, since polar auxin transport is known to be negatively affected in plants with horizontal or hanging stems, which may result in an altered hormone balance at the whole plant level. Com base nos resultados positivos do emprego de 2,4-D associado ao Thiabendazol no enraizamento de mudas na fase "palito", superando inclusive o AIB l000 ppm, testou-se a sua utilizacao no enraizamento de tocos enxertados de seringueira e, se viavel, indica-los como sucedaneos do AIB por serem de baixo custo e mais facil aquisicao.

O pau-de-leite (Sapium glandulatum (Vell.) Pax., Euphorbiaceae) é uma árvore nativa de vários biomas  brasileiros, com grande interesse para a recuperação  de ecossistemas degradados devido à sua rusticidade e ornitocoria intensa. Contudo, sua propagação sexuada é difícil, uma vez que a maioria das flores é masculina  e a porcentagem de germinação das sementes é muito baixa. ABSTRACT: The hormesis effect of 2,4-D, applied at two stages of soybean development, on the physiological components of crop yield, was investigated. Soybean seeds of the cultivar ‘AG 3680 IPRO’ were sown in pots containing 7 L of a substrate and kept in a greenhouse. A randomized complete block design with four replicates was used. Eighteen treatments were tested, arranged in a split-plot scheme. Propagation of the fig tree predominately uses hardwood cuttings obtained from pruning stock plants. With a view to optimising fig tree propagation, the effects of auxin and misting on the rooting of herbaceous and hardwood cuttings from the fig tree were evaluated. An experiment was set up in a 2x2x2 factorial design, with two types of cuttings (softwood and hardwood), two levels of auxin (0 and 1000 mg L-1 indolbutyric acid) and two environments (with and without misting). ABSTRACT A standard protocol to evaluate the effects of biostimulants on plant physiology is still lacking. The proton pumps present in the vacuolar and plasma membranes are the primary agents responsible for the regulation of the electrochemical gradient that energizes the nutrient uptake system and acid growth mechanism of plant cells. In this study, two of these enzymes were characterized as biochemical markers of biostimulant activity. Este trabalho teve como objetivo avaliar a resposta rizogênica in vitro de ápices caulinares de plantas jovens de mamoeiro ‘Tainung 01’, em diferentes tempos de permanência em meios de indução e regeneração. O meio de indução empregado foi o MS, suplementado com complementos orgânicos constituídos de sacarose, mio-inositol, tiamina, piridoxina, ácido nicotínico, além de AIB. O meio de regeneração diferiu do meio de indução por ser desprovido de AIB. Os tratamentos consistiram em tempos variáveis de permanência dos explantes nos meios de indução e de regeneração (indução/regeneração): 0/25; 5/20; 10/15; 15/10; 20/5 e 25/0 dias.
