We report the isolation, development and multiplex optimisation of 12 new microsatellite loci for the great scallop, Pecten maximus. Diversity was moderate to high, with number of alleles ranging from 4 to 20 and observed heterozygosity between 0.28 and 0.88. Progeny produced in a commercial hatchery was used to test locus power for parentage assignment. The percentage of offspring that was unambiguously assigned to a unique pair of parents was 97% (software package CERVUS-COLONY). Parentage assignment revealed that 22% of the studied progeny resulted from unplanned crosses. Effective population size of the study progeny was also estimated. Our study illustrates the power of microsatellites for the genetic monitoring of hatchery-produced great scallops. |

The main microbial diseases affecting marine cultured bivalves have been revised on the basis of the etiologic agents, pathogenesis and pathogenicity. Several recent bivalve-interaction models have been studied, including Pecten larvae-Vibrio pectinicida, brown ring disease, juvenile oyster disease, Pacific oyster nocardiosis and summer mortalities of oysters. In addition, the taxonomy and phylogeny of new potential bivalve pathogens and their virulence factors have been established. There is an ongoing need to isolate and purify ceIls in bivalve mollusc disease research. Many separation techniques differentiate cells solely by a single factor and usually require additives in the medium that may damage live cells. Centrifugai elutriation is a technique that separates particles by size and density simultaneously and can be conducted in physiological media without additives. Living cells can be separated without chemical damage and, because the procedure can be conducted aseptically, cells can be maintained in vitro after elutriation. Summer mortality of the Pacific oyster Crassostrea gigas is the result of a complex interaction between oysters, their environment and their pathogens. The physiological status of an oyster, especially its reproductive status, is suspected to play a significant role in the outcome of this interaction. As genetic variability exists for susceptibility to summer mortality, resistant (R) and susceptible (S) oyster lines were produced using a divergent selection scheme. Through differential screening between oyster families selected for high and low summer survival, we have characterized a new transforming growth factor-beta (TGF-beta) superfamily member. This novel factor, named oyster-gonadal-TGF beta-like (og-TGF beta-like), is synthesized as a 307 amino acid precursor and displays 6 of the 7 characteristic cysteine residues of the C-terminal, mature peptide.
