The spread of major diseases of molluscs around the world and the increase of transfers need the development of adapted prophylactic strategies and the increase of research in epidemiology, immunology and genetics. The prophylactic strategies must be based on common laws with the establishment of a list of declarable pathogens, with basic sampling rules (sample size, frequency, definition of geographical areas) and with reglementation of the internal transfers. A systematic surveillance progam conducted since 1980 along the coasts of Britanny has followed the evolution and epidemiological state of the natural O. eulis oyster beds and seed areas in the intertidal zones and deep water. Presently, Marteilia refringens exists endemically in Brittany estuaries ; the rate and degree of infestation varies from year to year depending on the hydrological conditions and the geographical region. In no case, however, is the return of flat oyster culture envisoned. QTL mapping for resistance to bonamiosis in the European flat oyster Ostrea edulis ArchiMer

A breeding programme has been undertaken by Ifremer to produce selected Ostrea edulis families which are resistant to Bonamia ostreae, a parasite which is the most significant constraint on aquaculture of this oyster. Using such families, we aimed to identify Quantitative Trait Loci (QTLs) of resistance/susceptibility to bonamiosis. We used an F2 family derived from a bi-parental cross between a wild oyster and an individual from one of the selected oyster families. A total of 550 F2 individuals from the family cohabited with wild oysters that were injected with the parasite. From 1988 to 1989, a study has been conducted pertaining to the growth of the flat oyster Ostrea edulis, as well as Marteilia refringens and Bonamia ostreae outbreaks. Commercial mono specific farming (flat oysters), with low density or mixed (flat and hollow oysters) along with higher density, has been taking place in the Arcachon basin oceanographic sectors, where main hydrobiological parameters were acquired simultaneously. The excellent growth of the flat oyster was witnessed in mono specific farming. However this growth was average in mixed farming, probably due to higher total oyster density and interspecies competition. L'ostréiculture représente une part économique régionale et française importante avec une production d'environ 117 106 Tonnes de coquillages par an (FAO, 2003). Cependant le développement de cette activité peut être limité par des mortalités, parfois massives, dues à des maladies infectieuses. L'huître plate Ostrea edulis a subi deux maladies majeures : la Marteiliose due à Marteilia refringens et la Bonamiose due à Bonamia ostreae. Molecular detection and quantification of the protozoan Bonamia ostreae in the flat oyster, Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. L'huître plate, Ostrea edulis, espèce indigène des côtes européennes est présente de la Norvège à la baie d'Agadir au Maroc ainsi que sur le pourtour méditerranéen et en Mer Noire. Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. After its first description in June 1979 in L'Ile Tudy, Brittany, the parasite rapidly spread to all French oyster farming areas and in other European countries through transfers of live molluscs. Bonamiosis and marteiliosis, another protozoan disease appeared in the seventies, drastically reduced the French flat oyster production from 20 000 tonnes in 1970 to less than 2000 tonnes after 1981. Whole-genome amplification: a useful approach to characterize new genes in unculturable protozoan parasites such as Bonamia exitiosa ArchiMer

Bonamia exitiosa is an intracellular parasite (Haplosporidia) that has been associated with mass mortalities in oyster populations in the Southern hemisphere. This parasite was recently detected in the Northern hemisphere including Europe. Some representatives of the Bonamia genus have not been well categorized yet due to the lack of genomic information. In the present work, we have applied Whole-Genome Amplification (WGA) technique in order to characterize the actin gene in the unculturable protozoan B. exitiosa. The in vitro model Ostrea edulis hemocyte - Bonamia ostreae is interesting to investigate host-parasite interactions at the cellular level. Indeed, this unicellular parasite infects the flat oyster Ostrea edulis and multiplies within hemocytes, the central effectors of oyster defenses. Apoptosis is a mechanism used by many organisms to eliminate infected cells.
