ABSTRACT It has now been 15 years since the HER2-targeted monoclonal antibody trastuzumab was introduced in clinical and revolutionized the treatment of HER2-positive breast cancer patients. Despite this achievement, most patients with HER2-positive metastatic breast cancer still show progression of their disease, highlighting the need for new therapies. The continuous interest in novel targeted agents led to the development of pertuzumab, the first in a new class of agents, the HER dimerization inhibitors. ABSTRACT Insulin-like Growth Factor-1 (IGF-1) gene polymorphism has been associated with an increased risk for breast cancer. IGF-1 is a key regulator of proliferation, cell differentiation and apoptosis. It has important mitogenic and anti-apoptotic activities in normal cells and in breast cancer cells, acting synergistically with estrogen to increase neoplastic cell proliferation. Flow cytometric analysis of DNA content was performed on 28 samples of canine mammary tumors. Nine of them were benign and 19 were malignant. All benign tumors and 11 malignant tumors (57.9%) were diploid (P<0.05). Form the aneuploid tumors, five (26.3%) were hyperdiploid, one (5.3%) hypodiploid, one (5.3%) near triploid and one (5.3%) multiploid. The analysis of the expression of the markers PR and CD31 revealed a significant difference between diploid and aneuploid tumors (P<0.05). C2- and C4-position 17&#946;-estradiol metabolites play an important role in breast carcinogenesis. 2-Hydroxyestradiol and 4-hydroxyestradiol are implicated in tumorigenesis via two pathways. These pathways entail increased cell proliferation and the formation of reactive oxygen species that trigger an increase in the likelihood of deoxyribonucleic acid mutations. 2-Methoxyestradiol, a 17&#946;-estradiol metabolite, however, causes induction of apoptosis in transformed and tumor cells; thus exhibiting an antiproliferative effect on tumor growth. The 4-hydroxyestradiol:2-methoxyestradiol and 2-hydroxyestradiol:2-methoxyestradiol ratios therefore ought to be taken into account as possible indicators of carcinogenesis.

Inhibition of focal adhesion kinase by antisense oligonucleotides enhances the sensitivity of breast cancer cells to camptothecins Biocell

This study shows a strong association between cell attachment to substratum and activation of b1-integrin-signaling with resistance to the camptothecin derivative topotecan (TPT) in breast cancer cells. We propose a mechanistic-driven approach to sensitize the cells to camptothecins. ZR-75-1 anchorage-dependent breast cancer cell line, its derivative 9D3S suspension cells (9D3S-S), and 9D3S cells attached to fibronectin-coated plates (9D3S-A) were treated with TPT (1 mM) or CPT-11 (40 mM) for 48 h. BACKGROUND: BRCA protein interacts with at least 13 different proteins that have been implicated with cancer susceptibility and loss of BRCA function is correlated to sensitivity to DNA crosslinking agents in preclinical models. RESULTS: BRCA2 methylation frequency was 44%, p53 Pro22 allele frequency was 32% and heterozygous frequency of Arg/Pro72 genotype was 60% which could be associated as risk factor for metastasis (p = 0.046 OR = 4.190). Regarding to polymorphism of codon 249 the frequency of Arg249 allele presented 82% which was considered not statistically significant. A sample of 58 familial breast cancer patients from Venezuela were screened for germline mutations in the coding sequences and exon-intron boundaries of BRCA1 (MIM no. 113705) and BRCA2 (MIM no. 600185) genes by using conformation-sensitive gel electrophoresis. Ashkenazi Jewish founder mutations were not found in any of the samples. We identified 6 (10.3%) and 4 (6.9%) patients carrying germline mutations in BRCA1 and BRCA2, respectively. Radioguided surgery for breast cancer: preliminary experience in Piaui BABT

Intraoperatory detection and excision of sentinel lymph node (SLN), radioguided occult lesion localization (ROLL) and lesional resection, or a combination of these two procedures at one time in a same patient (SNOLL), are techniques that represent, at the moment, a significant advance in breast cancer surgery, procedures in clinical practice being established, as can be seen from the medical literature, as well as in this institution. Thirty-one patients with breast cancer were initially treated with the aid of radioguided surgery. Of those, 20 patients were submitted to sentinel lymph node biopsy, six have undergone only ROLL and five were submitted to a combination of the two techniques.

The aim of the work was to analyse the potential influence of different factors on the surgical detection of sentinel lymph node in breast cancer. The procedure has been performed in 704 patients. In 43 out of the 704 cases, the SN was not detected, 24 were palpable and 19 had no palpable tumors. Lymphoscitigraphy was done in 2 h p.i. of 37-55 MBq of 99mTc-nanocolloid. The day before surgery in palpable tumours and 4 h previous to surgery in non-palpable tumours, surgical detection was performed using a gammaprobe. Apoptosis plays an important role in oncogenesis determining tumor growth and aggressiveness. The aim of this study was to examine the relationship between apoptosis, diagnosis and prognosis in canine mammary tumors. Thirty bitches were submitted to tumor extirpation and a method of labeling apoptotic cells, that labels DNA fragments, by terminal deoxynucleotidyl-tranferase (TdT) mediated by 5' deoxy-uridine-triphosphate (dUTP) nick and labeling (TUNEL) was used. The apoptotic cells were counted in ten high power fields (10HPF). Statistical results showed that when there are more than 30 apoptotic cells/10HPF the prognosis is worse (p= 0,0005). Thus suggesting that apoptosis can be used as a prognostic marker in canine breast tumors.

Our objective was to determine the presence of vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 (MMP-2) and MMP-9 and specific tissue inhibitors of matrix metalloproteinase (TIMP-1 and TIMP-2) in tumor samples obtained from patients with primary breast cancer. We attempted to correlate these findings with the status of the sentinel lymph node (SLN) and clinical-pathological characteristics such as age, tumor size, histological type, histological grade, and vascular invasion. Tumor samples from 88 patients with primary breast cancer were analyzed.
