Characterization of Staphylococcus spp. strains in milk from buffaloes with mastitis in Brazil: the need to identify to species level to avoid misidentification Arq. Bras. Med. Vet. Zootec.
Coimbra-e-Souza,V.; Brito,M.A.V.P.; Chamon,R.C.; Laport,M.S.; Giambiagi-deMarval,M..
ABSTRACT Mastitis is an inflammation of the mammary gland that affects dairy cattle worldwide causing economic losses. Coagulase-negative staphylococci (CNS) are the predominant cause of this type of infection. We have recently showed that coagulase-positive staphylococci could be misidentified. So, the aim of this study was to characterize the Staphylococcus spp. strains initially classified as coagulase-negative Staphylococci, isolated from buffalo with subclinical mastitis. Milk of buffaloes with mastitis in herds was collected and 9 strains were identified as CNS by phenotypic tests. Molecular methodologies latter identified the strains as coagulase-negative Staphylococcus chromogenes (5), coagulase-positive Staphylococcus hyicus (2) and...

Estimates of correction factors for lactation length and genetic parameters for milk yield in buffaloes Arq. Bras. Med. Vet. Zootec.
Tonhati,H.; Muñoz,M.F.C.; Duarte,J.M.C.; Reichert,R.H.; Oliveira,J.A.; Lima,A.L.F..
Correction factors for milk yield at 90, 240, 270 and 305 days of lactation of buffaloes and genetic and environmental parameters for milk yield of these lactation periods were estimated. The data used consisted of 3888 lactation records of 1630 buffaloes from 10 herds reared in the State of São Paulo. Genetic parameters were estimated by the derivative free restricted maximum likelihood method, fitting an animal repeatability model. Heritability estimates for milk yield corrected for 90, 240, 270 and 305 days of lactation were 0.17, 0.15, 0.14 and 0.14, respectively. In this same order, repeatability estimates were 0.40, 0.44, 0.41 and 0.41. The genetic correlation estimates between these corrected milk yields ranged from 0.96 to 1.00. The multiplicative...

Lecirelin and Buserelin (Gonadotrophin releasing hormone agonists) are equally effective for fixed time insemination in buffalo Braz. J. Vet. Res. Anim. Sci.
BARUSELLI,Pietro Sampaio; AMARAL,Renato; BARUFI,Francisco Bonomi; VALENTIM,Renato; MARQUES,Marcio de Oliveira.
Buffalo has peculiar reproductive patterns, which make artificial insemination programs a hard and expensive task. Artificial insemination in fixed time is advantaged because females show low incidence of homosexual behaviour and strong dominance relationships, which leads to a poor accuracy in estrus detection. The aim of this experiment was to compare the efficiency of two different GnRH agonists in the GnRH/PGF2alpha/GnRH protocol (Buserelin vs Lecirelin). Two hundred and seventy buffaloes with 45 to 60 days postpartum were synchronized and fixed-time inseminated. The animals were kept on pasture in two farms at São Paulo and Mato Grosso do Sul (Brazil). Cows in Group 1 (n = 132) received, intramuscularly, 20 µg of Buserelin at a random day of the...

In April 1998 urine samples from adult female buffaloes were collected in a farm located in Registro, Vale do Ribeira, São Paulo State, Brazil. The urine samples obtained after furosemide injection were immediately transported to the laboratory in liquid modified EMJH medium and seeded, by the serial dilution technique, into Fletcher's or modified EMJH-0.2% agar, both of them with 5-fluorouracil 100mg/mL. The intraperitoneoum inoculation of 0.5 mL was also performed with each urine sample in young, adult hamsters (Mesocricetus auratus). All samples seeded directly in culture medium were contaminated. The hamsters did not show any sign of disease and were killed at the 21st post inoculation day. At this time kidney cultures of these animals were performed...

Classical nonlinear models to describe the growth curve for Murrah buffalo breed Ciência Rural
Araújo,Ronyere Olegário de; Marcondes,Cintia Righetti; Damé,Maria Cecília Florisbal; Garnero,Analía del Valle; Gunski,Ricardo José; Everling,Dionéia Magda; Rorato,Paulo Roberto Nogara.
With the objective of to adjust nonlinear models for the growth curves for a buffaloes herd raised in floodable lands in Rio Grande do Sul state, monthly records measured from birth to two years-old of 64 males and 63 females born between 1982 and 1989 were used. The models used were: Von Bertalanffy, Brody, Gompertz and Logistic. The parameters were estimated by NLIN procedure and the criteria used to evaluate the adjustment given by the models were: asymptotic standard deviation; coefficient of determination; average absolute deviation of residues and asymptotic index. Von Bertalanffy and Brody models overestimated the male asymptotic weight (A) in 15.9 and 171.3kg, respectively, and the Gompertz and Logistic models underestimated it in 4.5 and 13.4kg,...

Evaluation of thermorregulatory capacity of dairy buffaloes using infrared thermography REA
Sevegnani,Kelly B; Fernandes,Danielle P. B; Silva,Silvia H. Modenese-Gorla da.
ABSTRACT The objective of this study was to evaluate the thermoregulatory response of dairy buffaloes in pre-milking and post-milking. To identify animal thermoregulatory capacity, skin surface temperatures were taken by an infrared thermometer (SST), a thermographic camera (MTBP) as well as respiratory rate records (RR). Black Globe and Humidity Index (BGHI), radiating thermal load (RTL) and enthalpy (H) were used to characterize the thermal environment. Artificial Neural Networks analyzed those indices as well as animal physiological data, using a single layer trained with the least mean square (LMS) algorithm. The results indicated that pre-milking and post-milking environments reached BGHI, RR, SST and MTBP values above thermal neutrality zone for...

Identification of novel single nucleotide polymorphisms in the DGAT1 gene of buffaloes by PCR-SSCP Genet. Mol. Biol.
Raut,Ashwin A.; Kumar,Anil; Kala,Sheo N.; Chhokar,Vinod; Rana,Neeraj; Beniwal,Vikas; Jaglan,Sundeep; Samuchiwal,Sachin K.; Singh,Jitender K.; Mishra,Anamika.
Diacylglycerol O-acyltransferase 1 (DGAT1) is a microsomal enzyme that catalyzes the final step of triglyceride synthesis. The DGAT1 gene is a strong functional candidate for determining milk fat content in cattle. In this work, we used PCR-SSCP (polymerase chain reaction-single-strand conformation polymorphism) and DNA sequencing to examine polymorphism in the region spanning exon 7 to exon 9 of the DGAT1 gene in Murrah and Pandharpuri buffaloes. Three alleles (A, B and C) and four novel single-nucleotide polymorphisms were identified in the buffalo DGAT1 gene. The frequencies of the alleles differed between the two buffalo breeds, with allele C being present in Murrah but not in Pandharpuri buffalo. The allele variation detected in this work may...

Characterization of PRLR and PPARGC1A genes in buffalo (Bubalus bubalis) Genet. Mol. Biol.
Javed,Ruheena; Gautam,Sanjeev K.; Vijh,Ramesh K.; Tantia,Madhu S..
More than 40 million households in India depend at least partially on livestock production. Buffaloes are one of the major milk producers in India. The prolactin receptor (PRLR) gene and peroxisome proliferators activated receptor-γ coactivator 1-alpha (PPARGC1A) gene are reportedly associated with milk protein and milk fat yields in Bos taurus. In this study, we sequenced the PRLR and PPARGC1A genes in the water buffalo Bubalus bubalis. The PRLR and PPARGC1A genes coded for 581 and 819 amino acids, respectively. The B. bubalis PRLR gene differed from the corresponding Bos taurus at 21 positions and four differences with an additional arginine at position 620 in the PPARGC1A gene were found in the amino acid sequence. All of the changes were confirmed by...

Sequence characterization of coding regions of the myostatin gene (GDF8) from Brazilian Murrah buffaloes (Bubalus bubalis) and comparison with the Bos taurus sequence Genet. Mol. Biol.
Mota,Lígia S.L.S.; Curi,Rogério A.; Palmieri,Darío A.; Borges,Alexandre S.; Lopes,Catalina R.; Barbosa,José D.; Gimenes,Marcos A..
Within about 30 years the Brazilian buffalo (Bubalus bubalis) herd will reach approximately 50 million head as a result of the great adaptive capacity of these animals to tropical climates, together with the good productive and reproductive potential which make these animals an important animal protein source for poor and developing countries. The myostatin gene (GDF8) is important in the physiology of stock animals because its product produces a direct effect on muscle development and consequently also on meat production. The myostatin sequence is known in several mammalian species and shows a high degree of amino acid sequence conservation, although the presence of non-silent and silent changes in the coding sequences and several alterations in the...
