Effect of environmental and phenological factors on the antimicrobial activity of Cochlospermum regium (Schrank) Pilg. roots Agronomy
Inácio, Marielle Cascaes; Paz, Tiago Antunes; Bertoni, Bianca Waléria; Pereira, Ana Maria Soares.
Cochlospermum regium (Bixaceae) is a medicinal plant species native to the Brazilian savannah, known as the Cerrado. Local communities use its roots to treat infections of the female reproductive system. This study evaluated how seasonality, phenological stage, geographical location, plant age, and plant cultivation substrate affect the antimicrobial activity of Cochlospermum regium roots against Candida albicans. Although the plant displayed antimicrobial action in all the assayed conditions, the roots collected during fall and winter inhibited C. albicans the most effectively. The geographical location in which the plant material was found did not influence its biological response. Plant age and cultivation substrate did impact the biological response;...

Histatin 5 and human lactoferrin inhibit biofilm formation of a fluconazole resistant Candida albicans clinical isolate Anais da ABC (AABC)
CURVELO,JOSÉ A.R.; MORAES,DANIEL C. DE; ANJOS,CAMILA A. DOS; PORTELA,MARISTELA B.; SOARES,ROSANGELA M.A..
Abstract:Candida albicans is the most important fungal pathogen that causes infections in humans. Biofilms are hard-to-treat structures due to their high antifungal resistance. Saliva is a fluid that contains antimicrobial substances acting as the first-line of defense against pathogens, and its immune components may be potential tools for the discovery of new treatments against candidiasis. To evaluate the activity of histatin 5 and human lactoferrin against biofilm formation. A fluconazole-resistant Candida albicans clinical isolate was used as the model microorganism. Morphogenesis was evaluated by differential counting. Biofilm quantification was performed by XTT reduction assay. Thickness and topography of biofilms were assessed through confocal laser...

Ocorrência de Candida albicans em intestinos de bovinos Arq. Bras. Med. Vet. Zootec.
Souza,W.A.; Siqueira,A.M..

Histological Observation and Expression Patterns of antimicrobial peptides during Fungal Infection in Musca domestica (Diptera: Muscidae) Larvae BABT
Jiangfan,Xiu; Tao,Wang; Yu,Wang; Jianwei,Wu; Guo,Guo; Yingchun,Zhang; Xiaoli,Shang.
ABSTRACT Housefly, Musca domestica, has a complicated immune system. However, its underlying operating mechanism remains elusive. Candida albicans is a major pathogen affecting humans by causing deep infection fungous disease, but it is non-symbiotic in houseflies. To investigate the C. albicans infection process in housefly, the changes in morphological and histological and expression patterns of antimicrobial peptide were monitored to indicate the insect's response to fungal infection. The results showed that scattered brown spots were comprising melanized encapsulation and encapsulated fungal cells were initially observed at the inner side of larvae's body wall 3 h post-infection (PI). Between 6 and 36 h PI, the whole body of larvae was densely covered...

Secretory immunoglobulin A: a protective factor in the genital mucosa BJID
Giraldo,Paulo C.; Gonçalves,Ana Katherine; Eleutério Junior,José.
The genital mechanisms of defense are not well understood and are therefore ignored during therapy. This fact results in a great number of cases of treatment failure. The mucosa is an important protective factor of the genital female system, through self-defense mechanisms, and secretor antibodies (immunoglobulin A). The lymphoid tissue exerts protective anti-inflammatory activity, besides inhibiting microorganism adherence, neutralizes viruses and toxins and stabilizes the mucosal flora. Although certain microorganisms, such as viruses and fungus, are controlled by cellular immunity, secretory IgA can also exert an important role in the control of these agents.

Arachidonic acid affects biofilm formation and PGE2 level in Candida albicans and non-albicans species in presence of subinhibitory concentration of fluconazole and terbinafine BJID
Mishra,Nripendra Nath; Ali,Shakir; Shukla,Praveen K..
Candida albicans utilizes arachidonic acid (AA) released during the course of infection (Candidiasis) from phospholipids of infected host cell membranes and synthesizes extracellular prostaglandin(s) which play an important role in hyphae formation and host cell damage. C. albicans biofilms secrete significantly more prostaglandin(s) and evidence suggests that Candida biofilms have dramatically reduced susceptibility to majority of antifungal drugs. AA influences the saturation level of lipids and fluidity of yeast cell membranes. Therefore the aim of this study was to evaluate the effect of AA alone or in combination with antifungal agents on biofilm formation and production of prostaglandin (PGE2) in C. albicans, C. parapsilosis, C. glabrata, C....

Effect of pathogenic yeasts on human platelet aggregation BJID
Neiva,Teresinha de Jesus Carvalho; Santos,Jairo Ivo dos.
We investigated the effects of Candida albicans, Cryptococcus neoformans and the respective culture supernatants on human platelet aggregation (PA). Both yeasts were unable to aggregate the platelets directly. On the other hand, cells of these yeasts significantly (P < 0.01) inhibited PA at concentrations equal to or higher than 1 x 10(6) cells/mL for C. albicans and equal to or higher than 1 x 10(5) cells/mL for C. neoformans. When the supernatants of one-week broth cultures were added to the activated platelets no inhibition in aggregation was observed. Apparently somatic components of these yeasts, but not their metabolic products, exert an antagonistic effect on the aggregation of human platelets, possibly aiding the fungi in their evasion of the...

Chloroquine sensitizes biofilms of Candida albicans to antifungal azoles BJID
Shinde,Ravikumar Bapurao; Raut,Jayant Shankar; Chauhan,Nitin Mahendra; Karuppayil,Sankunny Mohan.
Biofilms formed by Candida albicans, a human pathogen, are known to be resistant to different antifungal agents. Novel strategies to combat the biofilm associated Candida infections like multiple drug therapy are being explored. In this study, potential of chloroquine to be a partner drug in combination with four antifungal agents, namely fluconazole, voriconazole, amphotericin B, and caspofungin, was explored against biofilms of C. albicans. Activity of various concentrations of chloroquine in combination with a particular antifungal drug was analyzed in a checkerboard format. Growth of biofilm in presence of drugs was analyzed by XTT-assay, in terms of relative metabolic activity compared to that of drug free control. Results obtained by XTT-metabolic...

An unusual cause of bilateral scrotal abscess in a preterm infant: Candida albicans BJID
Cetinkaya,Merih; Buyukkale,Gokhan; Payasli,Muge; Ozbek,Sibel; Kavuncuoglu,Sultan.
Acute scrotal abscess is a rare condition in neonates. Most of these abscesses were reported to be unilateral and caused by Staphylococcus and Salmonella spp. Herein, we report a bilateral scrotal abscess in a preterm infant and Candida albicans was isolated from the scrotal fluid culture. To our knowledge, this is the first bilateral scrotal abscess in a preterm infant caused by C. albicans. Therefore, this organism must be suspected in differential diagnosis of acute scrotal abscess in neonates, especially in preterm infants.

Trailing end-point phenotype antibiotic-sensitive strains of Candida albicans produce different amounts of aspartyl peptidases BJMBR
Braga-Silva,L.A.; Mesquita,D.G.A.; Ribeiro,M.D.; Carvalho,S.M.F.; Fracalanzza,S.E.L.; Santos,A.L.S..
Candida albicans is an opportunistic fungal pathogen that causes severe systemic infections in immunosuppressed individuals. C. albicans resistance to antifungal drugs is a severe problem in patients receiving prolonged therapy. Moreover, trailing yeast growth, which is defined as a resistant MIC after 48 h of incubation with triazole antifungal agents but a susceptible MIC after 24 h, has been noted in tests of antifungal susceptibility against some C. albicans isolates. In this context, we recently noticed this phenomenon in our routine susceptibility tests with fluconazole/itraconazole and C. albicans clinical isolates. In the present study, we investigated the production of cell-associated and secreted aspartyl peptidases (Saps) in six trailing...

Synergistic action of starch and honey against Candida albicans in correlation with diastase number BJM
Boukraa,Laïd; Benbarek,Hama; Moussa,Ahmed.
To evaluate the synergistic action of starch on the antifungal activity of honey, a comparative method of adding honey with and without starch to culture media was used. Candida albicans has been used to determine the minimum inhibitory concentration (MIC) of five varieties of honey. In a second step, lower concentrations of honey than the MIC were incubated with a set of concentrations of starch added to media to determine the minimum synergistic inhibitory concentration (MSIC). The MIC for the five varieties of honey without starch against C. albicans ranged between 40% and 45% (v/v). When starch was incubated with honey and then added to media, a MIC drop has been noticed with each variety. It ranged between 7% and 25%. A negative correlation has been...

Vulvovaginal candidiasis in Mato Grosso, Brazil: pregnancy status, causative species and drugs tests BJM
Dias,Luciana Basili; Melhem,Márcia de Souza Carvalho; Szeszs,Maria Walderez; Meirelles Filho,José; Hahn,Rosane Christine.
Causative agent in majority of VVC is Candida albicans, but infection due to non-C. albicans is common. Use of empiric antifungal therapy in Brazil due to syndromic management of vulvovaginitis could act as risk factor for increase resistance among VVC causative agents. From Mato Grosso patients, 160 with culture-proved among 404 women who had clinical symptoms of VVC, were enrolled in this study. 70 non-pregnant women and 90 pregnant women were included. Candida albicans was the most prevalent, representing 72.9% in the non-pregnant group and 92.3% in the pregnant group. Differences in species distribution were noted between the two groups, being C. parapsilosis the second more prevalent species among non-pregnant women. Susceptibility testing revealed...

Antifungal activity of Cymbopogon winterianus jowitt ex bor against Candida albicans BJM
Oliveira,Wylly Araújo de; Pereira,Fillipe de Oliveira; Luna,Giliara Carol Diniz Gomes de; Lima,Igara Oliveira; Wanderley,Paulo Alves; Lima,Rita Baltazar de; Lima,Edeltrudes de Oliveira.
Candida albicans is an opportunistic yeast and a member of the normal human flora that commonly causes infections in patients with any type of deficiency of the immune system. The essential oils have been tested for antimycotic activity and pose much potential as antifungal agents. This work investigated the activity of the essential oil of Cymbopogon winterianus against C. albicans by MIC, MFC and time-kill methods. The essential oil (EO) was obtained by hydrodistillation using a Clevenger-type apparatus. It was tested fifteen strains of C. albicans. The MIC was determined by the microdilution method and the MFC was determined when an aliquot of the broth microdilution was cultivated in SDA medium. The phytochemical analysis of EO showed presence of...

Effect of alcohols on filamentation, growth, viability and biofilm development in Candida albicans BJM
Chauhan,Nitin M; Shinde,Ravikumar B; Karuppayil,S. Mohan.
In this study we report the potential of alcohols as morphogenetic regulators in Candida albicans. All the alcohols tested influenced various modes of growth like planktonic as well as biofilm forms. Viability was affected at high concentrations. Among the alcohols, the response of C. albicans to amyl alcohol (pentanol) was noteworthy. Amyl alcohol at a concentration 0.5% which was not inhibitory to growth and viability specifically inhibited morphogenetic switching from yeast to hyphal forms. It also inhibited normal biofilm development favoring yeast dominated biofilms. Based on this study we hypothesize that alcohols produced under anaerobic conditions may not favor biofilm development and support dissemination of yeast cells. Since anaerobic conditions...

Treatment of serum with supernatants from cultures of Candida albicans reduces its serum-dependent phagocytosis BJM
Santos,Aderbal Antonio dos; Sá,Eglius Alexandre Colignese de; Gaziri,Luis Carlos Jabur; Felipe,Ionice.
Candida albicans is a potent activator of the complement system, and heat labile opsonins produced by activation of C3 (C3b and iC3b) enhance phagocytosis of C. albicans mediated by complement receptors. In this study we treated mouse serum with supernatants from cultures of a protease producer strain of C. albicans and evaluated the ability of this serum to enhance phagocytosis of C. albicans. Cell-free supernatants from cultures of C. albicans were concentrated 5 fold and added to mouse serum for 30 min at 37ºC, before using this serum for opsonization of glutaraldehyde-fixed yeast cells. We observed that normal mouse serum increased about 3 fold the phagocytosis of C. albicans by mice peritoneal macrophages, whereas supernatant-treated serum did not...

Identification of genes differentially expressed in hyphae of Candida albicans BJM
Melo,Analy Salles de Azevedo; Serafim,Rui Cosme; Briones,Marcelo Ribeiro da Silva.
The ability to switch from yeast to hyphal forms is essential for Candida albicans virulence. This morphological switch involves the expression of hyphal-specific genes under the control of transcriptional factors. To contribute to the discovery of hyphal-specific genes, we used a differential screening method where clones of a genomic DNA library were hybridized with yeast and hyphal cDNA probes. Two clones with increased expression in hyphae were selected for study. Sequencing these clones, we found that they encoded two important metabolic genes, CaHXT7 (high-affinity hexose transporter) and CaYLL34 (member of the AAA ATPase family). CaHXT7 and CaYLL34 ORFs were completely determined. Analyses of the putative proteins show that: (1) CaHxt7p has one...
