Abstract: The aim of this work was to optimize a maceration condition of cannabis (Cannabis sativa L.). A circumscribed central composite experimental design was applied in this work. Temperature and time were varied from 40-80 °C and 30-90 min, respectively. The three responses (i.e., extraction yield, cannabidiol content, and Δ9- tetrahydrocannabinol content) were predicted by computer software. The yield was high when cannabis was macerated using ethanol at high temperature and long duration time. While cannabidiol and Δ9- tetrahydrocannabinol content was high when macerating at a low heating temperature and short duration time. The optimal condition provided the simultaneous high of cannabidiol and Δ9- tetrahydrocannabinol content was 40 °C for 30 min.... |

Cannabinoids, flavonoids, and stilbenoids have been identified in the annual dioecious plant Cannabis sativa L. Of these, the cannabinoids are the best known group of this plant's natural products. Polyketide synthases (PKSs) are responsible for the biosynthesis of diverse secondary metabolites, including flavonoids and stilbenoids. Biosynthetically, the cannabinoids are polyketide substituted with terpenoid moiety. Using an RT-PCR homology search, PKS cDNAs were isolated from cannabis plants. The deduced amino acid sequences showed 51%-73% identity to other CHS/STS type sequences of the PKS family. Cannabis Sativa is a multiuse herb in traditional medicine, its hydroalcoholic extract (10, 50, and 100 mg/kg) administered interaperitoneally for 14 consequent days to Wistar male rats resulted in significant decrease in progressive motility of sperm. Sperm count and seminiferous tubules diameter decreased significantly in comparison with control group. Also decrease in animal body weight in doses of 50 and 100 mg/kg was observed. Changes in testes weight and serum testosterone were not significant. Cannabis sativa extract has negative effect on sperm parameters such as motility, sperm count, and seminiferous tubules diameter. |
