COMPARING INOCULATION METHODS TO EVALUATE THE GROWTH OF Xanthomonas axonopodis pv. manihotis ON CASSAVA PLANTS Acta biol.Colomb.
MUÑOZ BODNAR,Alejandra; CRUZ GÓMEZ,Leidy Mariel; BERNAL,Adriana; SZUREK,Boris; LÓPEZ CARRASCAL,Camilo Ernesto.
Xanthomonas axonopodis pv. manihotis (Xam) is the causal agent of cassava bacterial blight (CBB), a major disease for cassava crops in South America and Africa. Until now the development of the disease is measured via AUDPC (Area Under Disease Progress Curve) but no reliable quantitative methods are available probably due to high variability of bacterial growth in planta. To establish an accurate method for bacterial quantification during the course of Xam infection within the host tissues, we analyzed bacterial populations upon stem and leaf-puncturing as well as leaf-clipping of cassava varieties MCOL1522 and SG107-35 challenged with the virulent Xam strain CIO151. Here, we show that the movement of bacteria along the tissues and especially in leaves is...

QTL IDENTIFICATION FOR CASSAVA BACTERIAL BLIGHT RESISTANCE UNDER NATURAL INFECTION CONDITIONS Acta biol.Colomb.
SOTO SEDANO,Johana; MORA MORENO,Rubén Eduardo; CALLE,Fernando; LÓPEZ CARRASCAL,Camilo Ernesto.
Cassava, Manihot esculenta Crantz, represents the main food source for more than one billion people. Cassava's production is affected by several diseases, one of the most serious is cassava bacterial blight (CBB) caused by Xanthomonas axonopodis pv. manihotis (Xam). A quantitative trait loci (QTL) analysis for CBB resistance was performed under natural infection conditions, using a mapping population of 99 full-sibs genotypes highly segregant and a SNP-based high dense genetic map. The phenotypic evaluation was carried out in Puerto López, Meta, Colombia, during the rainy season in 2015. Both resistant and susceptible transgressive segregants were detected in the mapping population. Through a non-parametric interval mapping analysis, two QTL were detected,...

DEVELOPMENT OF A MOTORIZED CASSAVA PEELER FOR SMALL AND MEDIUM SCALE ENTERPRISES CIGR Journal
Amanor, Ishmael Nartey; Bobobee, Emmanuel Yaovi Hunnuor.
In Ghana a lot of products are produced from cassava roots, but processing of most cassava product requires peeling. Cassava peeling in Ghana is a major manual activity, which is time consuming and labour-intensive and hence requires mechanization. About 34% of freshly harvested cassava roots is lost along the food chain and faster processing is necessary to curb this situation. The development of an efficient equipment to help fast-track peeling of cassava root is key. A motorized cassava peeler with four different lining materials (concrete, metal, rubber and wood) was developed and tested with two local cassava varieties (Asi-Abayiwa and Dabon). The batch loading weight, peel removal efficiency, percent flesh loss and peeling capacity of the peeler was...

GILES,JOÃO ANTONIO D.; OLIOSI,GLEISON; RODRIGUES,WEVERTON P.; BRAUN,HEDER; RIBEIRO-BARROS,ANA I.; PARTELLI,FÁBIO L..
Abstract The morphoagronomic characterization of 12 genotypes of M. esculenta was performed during the 2013/2014 and 2014/2015 crop years. The 12 genotypes were planted in a randomized block design, with four replicates per genotype. Number of tuberous roots per plant, weight of tuberous roots, root yield, total plant weight, harvest index, plant height, height of first branch, number of shoots, stem diameter, number of buds, leaf dry weight and petiole length were evaluated. Genotypes “Camuquem” and “Goiás” were the most productive, and “Amarela” and “Gema de Ovo” were the most divergent. Seventy percent of genetic diversity was due to petiole length (22.86%), root yield (19.20%), weight of tuberous roots (14.89%) and number of buds (13.72%). Overall, the...

Characterization of chestnut (Castanea sativa, mill) starch for industrial utilization BABT
Demiate,Ivo Mottin; Oetterer,Marília; Wosiacki,Gilvan.
Studies were conducted to characterize the chestnut and its starch. Chemical composition of the chestnuts showed high level of starch. Moisture level in the raw nuts was around 50g/100g in wet basis and starch content, around 80g/100g in dry basis; other nut flour components were protein (5.58 g/100g), lipid (5.39 g/100g), crude fiber (2.34 g/100g) and ash (2.14 g/100g). Starch fraction was chemically characterized in order to identify the granule quality as compared with those of cassava and corn. This fraction showed more lipids and proteins than the other starches. Chestnut starch granules showed peculiar shape, smaller than the control starches and low amount of damaged units. Chemical composition concerning amylose : amylopectin ratio was intermediate...

Esterase polymorphism marking cultivars of Manihot esculenta, Crantz BABT
Resende,Adriana Gazoli; Vidigal Filho,Pedro Soares; Machado,Maria de Fátima P. S..
Esterase isozymes were used to detected substrate-preference polymorphism in twenty cultivars of Manihot esculenta, and to show cultivar-specific variation of this species. A relatively complex extraction solution of proteins from leaves was needed to show a larger number of esterase isozymes. Similarity between cultivars from six groups ranged from 51 to 96%. The cultivars identified by the same name seemed to be biochemically different regarding esterase isozymes. Esterase isozyme electrophoretic patterns could, therefore, be used to discriminate the cultivars identified by the same name, and to monitor the vegetative propagation of cultivars maintained in the germplasm collection. In breeding strategies, isoesterase analysis could be used to avoid...

Thermal characterization of partially hydrolyzed cassava (Manihot esculenta) starch granules BABT
Lacerda,Luiz Gustavo; Azevedo,Jayme Augusto Menegassi; Carvalho Filho,Marco Aurélio da Silva; Demiate,Ivo Mottin; Schnitzler,Egon; Vandenberghe,Luciana Porto de Souza; Soccol,Carlos Ricardo.
Cassava starch, partially hydrolyzed by fungal á-amylase, was characterized using thermal analysis, light microscopy and X-ray diffraction. Thermal degradation was initiated at lower degradation temperatures after enzymatic treatment and the DSC (Differential scanning calorimetry) analysis showed almost similar range of gelatinization temperature, but the enthalpies of gelatinization were quite increased for the partially hydrolyzed starch granules. The results suggested that the partial degradation of the starch granules was concentrated in the amorphous regions.

Espaçamento para a cultura da Mandioca BABT
Takahashi,Mário; Guerini,Valdir Luis.
The present work had the objective of evaluating the effect of different line spacing upon some agronomic parameters of cassava (Manihot esculenta Crantz) culture. The experiment was conducted in county of Palotina, in the west region of Paraná State, Brazil. The studied spacing were 0.8, 1.0, 1.4 e 1.6 m between lines and 0.6 m between plants, which rendered population densities of 20,830, 16,660, 13,880, 11,900 and 10,410 plants/ha. For the production of fresh and dry root matter and stem fresh matter, the best space between lines was 1.0 m. The highest number of tuberous roots per plant, obtained with the space of 1.40 m between lines did not guarantee the highest productivity of root matter/ha. The production of the vegetative portion was not...

Somatic embryogenesis in cassava genotypes from the northeast of Brazil BABT
Feitosa,Terezinha; Bastos,João Luíz Pinheiro; Ponte,Luíz Ferreira Aguiar; Jucá,Thiago Lustosa; Campos,Francisco de Assis de Paiva.
A method for the induction of somatic embryogenesis in eight cassava genotypes from northeastern Brazil is described. The explants used were shoot apexes isolated both from in vitro grown plants and from shoots that sprouted from stem cuttings. Somatic embryogenesis was achieved in high frequencies by the addition in the induction medium of the auxin picloram over a wide range of concentrations. Green cotyledons of primary somatic embryos were used as explants to induce somatic (cyclic) secondary embryogenesis in an inducing medium supplemented with picloram at 12 mg/L. The method could be used not only for the mass production of plants of the cassava genotypes, but also to generate explants (green cotyledons of somatic embryos) as themselves excellent...

Thermoanalytical and starch content evaluation of cassava bagasse as agro-industrial residue BABT
Lacerda,Luiz Gustavo; Almeida,Rafael Ramires; Demiate,Ivo Mottin; Carvalho Filho,Marco Aurélio Silva; Vasconcelos,Eliane Carvalho; Woiciechowski,Adenise Lorenci; Bannach,Gilbert; Schnitzler,Egon; Soccol,Carlos Ricardo.
Starch nutritional fractions as well as thermal properties and other analysis are essential for food and industrial application. Cassava bagasse is an important agro-industrial residue and its starch content was evaluated using two alternative methods. Thermal characterization and microscopy analyses helped to understand how hydrolysis digests starchy fraction of cassava bagasse. The melting point of cassava starch occurred at 169.2ºC. Regarding TG analyses, after moisture content, there were observed two main mass losses for all samples. Results suggest hydrolysis carried out using enzyme is less effective in order to convert total starch content in cassava bagasse. However, using sulfuric acid, fibers are affected by analyses conditions.

Biodegradable films made from raw and acetylated cassava starch BABT
Larotonda,Fábio D. S.; Matsui,Kátia N.; Soldi,Valdir; Laurindo,João B..
Studies were carried out to produce biodegradable films from cassava starch. Two alternatives were investigated. In the first, films were obtained by starch gelatinization followed by thermopressing and glycerol was used at different concentrations as a plasticizer. In the second, starch acetate films were obtained by solubilization of cassava starch acetate in organic solvents, followed by casting on a glass plate and drying at room temperature. The films obtained by gelatinization were transparent and resistant to traction. The starch acetate films were also transparent but breakable. The use of these starch acetate films in paper impregnation improved the mechanical properties of the paper.

Tissue distribution and deposition pattern of a cellulosic parenchyma-specific protein from cassava roots BABT
Souza,Petrônio A.S.; Gomes-Filho,Enéas; Campos,Francisco A.P..
A protein with a molecular mass of 22kDa was purified from the cellulosic parenchyma of cassava roots. The amino acid composition of the protein was determined and antibodies generated against the purified protein were used to show that the concentration of the protein remains unchanged during root "tuber" formation. By using a tissue printing technique, as well as western blot, it was shown that the cellulosic parenchyma was the only root tissue in which the protein was deposited.

Overcoming crossing barriers between cassava, Manihot esculenta Crantz and a wild relative, M. pohlii Warwa Braz. J. Genet.
Nassar,Nagib M.A.; Carvalho,Claudio G.; Vieira,Clibas.
The use of mentor pollen has enabled successful hybridization between cassava, Manihot esculenta Crantz, and the wild species M. pohlii Warwa. Killed pollen of a cross compatible type produced by freeze-thawing was mixed with incompatible pollen and the mixes were dusted on stigmas. This treatment resulted in production of seed in 4.9% of the total pollinations, compared to 0% in the case of untreated pollinations. The use of a bridge species, M. neusana Nassar, through the hybrid M. pohlii and M. neusana also proved successful in overcoming interspecific barriers between cassava and M. pohlii.

Possible etiologies for tropical spastic paraparesis and human T lymphotropic virus I-associated myelopathy BJMBR
Zaninovic',V..
The epidemiology of tropical spastic paraparesis/human T lymphotropic virus I (HTLV-I)-associated myelopathy (TSP/HAM) is frequently inconsistent and suggests environmental factors in the etiology of these syndromes. The neuropathology corresponds to a toxometabolic or autoimmune process and possibly not to a viral disease. Some logical hypotheses about the etiology and physiopathology of TSP and HAM are proposed. Glutamate-mediated excitotoxicity, central distal axonopathies, cassava, lathyrism and cycad toxicity may explain most cases of TSP. The damage caused to astrocytes and to the blood-brain barrier by HTLV-I plus xenobiotics may explain most cases of HAM. Analysis of the HTLV-I/xenobiotic ratio clarifies most of the paradoxical epidemiology of TSP...

Identification of the bacterial community responsible for traditional fermentation during sour cassava starch, cachaça and minas cheese production using culture-independent 16s rRNA gene sequence analysis BJM
Lacerda,Inayara C. A.; Gomes,Fátima C. O.; Borelli,Beatriz M.; Faria Jr,César L. L.; Franco,Gloria R.; Mourão,Marina M.; Morais,Paula B.; Rosa,Carlos A..
We used a cultivation-independent, clone library-based 16S rRNA gene sequence analysis to identify bacterial communities present during traditional fermentation in sour cassava starch, cachaça and cheese production in Brazil. Partial 16S rRNA gene clone sequences from sour cassava starch samples collected on day five of the fermentation process indicated that Leuconostoc citreum was the most prevalent species, representing 47.6% of the clones. After 27 days of fermentation, clones (GenBank accession numbers GQ999786 and GQ999788) related to unculturable bacteria were the most prevalent, representing 43.8% of the clones from the bacterial community analyzed. The clone represented by the sequence GQ999786 was the most prevalent at the end of the fermentation...
