Common infectious diseases and skin test anergy in children from an urban slum in Northeast Brazil BJID
Castro,Melânia X.; Soares,Alberto M.; Fonsêca,Walter; Rey,Luís C.; Guerrant,Richard L.; Lima,Aldo A. M..
BACKGROUND: Acute respiratory infection (ARI), diarrheal disease (DD) and infective dermatitis (ID) are important causes of morbidity in children under five, in Northeast Brazil. Objectives: (a) to evaluate the morbidity of ARI, DD and ID; and (b) to determine their association with cellular immunity in poor urban children from Fortaleza, Brazil. MATERIALS AND METHODS: A prospective cohort study. At enrollment, multipuncture skin-tests (Multitest CMI) were performed and interpreted according to standard procedures. Children were followed for infectious diseases by weekly home visits. RESULTS: Seventy-one children aged 6 to 21 months were recruited in an ongoing cohort of newborns. A mean of 39 (6 to 63) home visits per child were made, which detected 184.5...

Interleukin-12 gene adjuvant increases the immunogenicity of virus-like particles of human papillomavirus type 16 regional variant strain BJID
Wei,Lanlan; Chu,Ming; Zhang,Qingmeng; Wang,Yan; Shang,Qinglong; Zhang,Yunyan; Zhang,Guangmei.
Objectives:To analyze the immunogenicity of virus-like particles (VLP) of human papillomavirus type 16 (HPV16) isolated in East China and the adjuvant potential of interleukin-12 (IL-12).Methods:The variant HPV16 L1VLP expressed in sf9 insect cells were purified with cesium chloride gradient centrifugation. BALB/c mice were vaccinated with VLP (L1N), VLP with Freund's adjuvant (L1A) or VLP with IL-12 recombinant plasmid (L1P). HPV16 VLP specific IgG and IFN-γ level in the serum were detected by ELISA, and the percentage of CD4+ and CD8+ in spleen cells was detected with flow cytometry. Results:The titers of serum IgG antibodies in vaccinated groups were higher than in negative control and the serum antibodies mainly recognized conformation-dependent HPV16...

Identification of Mycobacterium bovis antigens by analysis of bovine T-cell responses after infection with a virulent strain BJMBR
Alito,A.; McNair,J.; Girvin,R.M.; Zumarraga,M.; Bigi,F.; Pollock,J.M.; Cataldi,A..
Purification and characterization of individual antigenic proteins are essential for the understanding of the pathogenic mechanisms of mycobacteria and the immune response against them. In the present study, we used anion-exchange chromatography to fractionate cell extracts and culture supernatant proteins from Mycobacterium bovis to identify T-cell-stimulating antigens. These fractions were incubated with peripheral blood mononuclear cells (PBMC) from M. bovis-infected cattle in lymphoproliferation assays. This procedure does not denature proteins and permits the testing of mixtures of potential antigens that could be later identified. We characterized protein fractions with high stimulation indices from both culture supernatants and cell extracts....

Phenoloxidase activity and haemolymph cytology in honeybees challenged with a virus suspension (deformed wings virus DWV) or phosphate buffered suspension (PBS) Ciência Rural
Millanta,Francesca; Sagona,Simona; Mazzei,Maurizio; Forzan,Mario; Poli,Alessandro; Felicioli,Antonio.
ABSTRACT: The innate immune system of honeybees mainly consists in antimicrobial peptides, cellular immunity and melanisation. In order to investigate the immune response of honeybees to immune stressors, three stress degrees were tested. Newly emerged bees naturally DWV-infected were collected from a Varroa mite-free apiary and divided into three experimental groups: naturally DWV infected bees, PBS injected bees, and artificially DWV super infected bees. Phenoloxidase activity and haemolymph cellular subtype count were investigated. Phenoloxidase activity was highest (P<0.05) in DWV-superinfected bees, and the haemocyte population differed within the three observed groups. Although, immune responses following DWV infection have still not been...

Role of cytokines, NO, and H2O2 on the immunopathology of Leptospirosis in genetically selected mice J. Venom. Anim. Toxins incl. Trop. Dis.
Marinho,M.; Langoni,H.; Oliveira,S. L.; Lima,V. M. F.; Peiró,J. R.; Perri,S. H. V.; Carreira,R..
Immune response to leptospirosis is mainly humorally mediated, and involves opsonization of leptospires for phagocytosis by macrophages and neutrophils. However, some aspects are still unknown. For a more detailed analysis of the cellular immune response to leptospirosis infection, trials were carried out in order to determine the hydrogen peroxide and nitric oxide (H2O2 and NO) production stimulated or not by Interferon-gamma. The participation of some specific cytokines, such as Tumor Necrosis Factor-alpha (TNF-alpha); Interferon-gamma (IFN-gamma); Interleukin-6 (IL-6); and Interleukin-4 (IL-4), in the immunopathology of this infection was also investigated. For this purpose, we analyzed the supernatant from peritoneal macrophage cell culture and the...

Candidacy of liver stage antigen-1 for Plasmodium falciparum vaccine development OAK
Taylor-Robinson, Andrew W.; Heal, Karen G..
Malaria is the world's most important parasitic disease, imposing a massive health burden on people living in the tropics, often in the poorest countries. The vast majority of deaths in humans from malaria are caused by one species of the hemoprotozoan, Plasmodium falciparum., against which effective control measures are urgently needed. The global situation has deteriorated in recent times due to increased resistance of the anopheline mosquitoes that transmit P. falciparum to insecticides, and of the parasites themselves to drug therapy. An efficacious and cost-effective vaccine against this parasite is considered a public health priority. A vaccine that targets pre-erythrocytic parasites in the liver could potentially prevent clinical disease by blocking...

Effects of levamisole hydrochloride on cellular immune response and flock performance of commercial broilers Rev. Bras. Ciênc. Avic.
Oladele,OA; Emikpe,BO; Adeyefa,CAO; Enibe,F.
Levamisole hydrochloride (Lev.HCl) has been acclaimed to boost immune response particularly in immunocompromised state. Its routine use as an immunomodulator in poultry production is yet to be well embraced, thus its effects of on cellular immunity and flock performance of commercial broilers were evaluated. One hundred and fifty Anak broiler chicks were separated into two groups of 75 each. Broilers in group 1 were sensitized with 150µg of Staphylococcus aureus antigen each at 4 and 5 weeks, while those in group 2 were not sensitized. Each group was further divided into subgroups A, B, and C. Levamisole hydrochloride (40 mg/kg) was administered orally to 1A and 2A at 45 and 46 days of age and to 1B and 2B at 47 and 48 days of age, while 1C and 2C were not...
