The recent interest in bioconversion of agricultural and industrial wastes to chemical feedstock has led to extensive studies on cellulolytic enzymes produced by microorganisms. In the present study three lignocellulosic substrates viz. sugarcane bagasse, sawdust and water hyacinth were pre-treated with alkali and enzyme and their effect on bioconversion has been investigated. Production of lignocellulolytic enzymes by Aspergillus niger biofilms at variable water activities Electron. J. Biotechnol.

Lignocellulolytic enzyme production by Aspergillus niger was compared both in submerged fermentation (SF) and biofilm fermentation (BF) at varying water activities. Maximal filter paper activity, endoglucanase and xylanase activities were much higher in BF (2.96, 4.7 and 4.61 IU ml-1, respectively) than in SF cultures (1.71, 1.31 and 2.3 IU ml-1, respectively) but biomass yields were lower in BF than in SF (0.338 g g-1 and 0.431 g g-1, respectively). In the presence of 20% ethylene glycol (a w = 0.942) the enzyme activities decreased in both systems but BF still had higher levels (1.0, 1.0 and 2.6 IU ml-1, respectively) than SF cultures (0.6, 0.7 and 1.5 IU ml-1, respectively). Since fungal morphology influences the productivity of fungal fermentations, it is of major importance to well know the fungal behavior during culture for cellulase production. Characterization of extracellular cellulose-degrading enzymes from Bacillus thuringiensis strains Electron. J. Biotechnol.

The gram-positive spore-forming bacteria, Bacillus thuringiensis (Bt) strains produced novel cellulases which could liberate glucose from soluble cellulose, carboxymethyl cellulose (CMC), and insoluble crystalline cellulose. The maximal cellulase activities were obtained after 60 hrs incubation at 28ºC in a LB broth medium with 1% CMC. Maximum CMCase activities were got at 40ºC and pH 4.0, respectively, and more than 50% of its maximal activity was retained at 40-60ºC for 1 hr, while approximately 40% of its maximal activity was also retained after incubating at 70ºC for 1 hr. Thirty-two ½ Angus ½ Nellore crossbred steers at an average age of 12 months and an average initial weight of 422 kg, were kept in a feedlot for 77 days. Use of remazol blue dyed avicel for the determination of cellulolytic activity in basidiomycetes Rev. Microbiol.

A modified method for direct determination of cellulolytic activity using Avicel colored with Remazol Brilliant Blue R (RBBR) in Agar test tubes is discussed. Refinements were introduced in a simple method for quantitation of cellulase activity, based on the release of dye from Avicel-RBBR medium by the enzymatic hydrolysis. Modifications in Avicel-dye preparation were enhanced and a spectrophotometer for direct OD measurement in agar test tubes used. The use of a spectrophotometer improved the precision of the collected data, since absorbance measurements could be done at the maximum wavelenght for RBBR (595 nm).

In this study, we isolated filamentous fungi from the guts of Amazonian aquatic insect larvae to evaluate their cellulolytic activity. Abundance of lignocellulosic biomass provides a good solution to the demands of energy crops in producing biofuel like biodiesel and bioethanol. In this study, bioethanol was produced from sago hampas via the Simultaneous co-Saccharification and Fermentation (Sc-SF) process, at 2.5% and 5.0% (w/v) solid loadings. The processing step in Sc-SF is virtually similar to that of Simultaneous Saccharification and Fermentation (SSF). The present work aimed to study some aspects of sorption and immobilization of cellulase molecules on soil components by the analysis of the reactions of cellulase in a soil treated with different levels of maize residue and incubated for 90 days. The analysis of variance showed that the effects of the treatments of maize straw, incubation time and their interaction on cellulase adsorption, desorption and immobilization were statistically significant. ABSTRACT Rumen fungi inhabit the gastro-intestinal tract of ruminants and the most non-ruminant herbivores. Rumen fungi produce highly active plant cell wall degrading enzymes, therefore they have gained scientific interest. In this study, genes encoding xylanase (xynA-7) and cellulase (celA-5) were amplified from Neocallimastix sp. GMLF7 and Orpinomyces sp. GMLF5, respectively, and expressed in Escherichia coli. XynA-7 was found to be active only on xylan, however CelA-5 had activity both on carboxymethyl cellulose and lichenan. Cellulase activity of a Lentinula edodes (Berk.) Pegl. strain grown in media containing Carboximetilcellulose or microcrystalline cellulose BABT

Endoglucanase and exocellobiohydrolase produced b Lentinula edodes (Berk.) Pegl. strain thatt was cultivated in carboxymetilcellulose (CMC) or microcrystalline cellulose (Avicel) liquid media. The concentration and type of cellulose influenced the enzyme activity and production. Extra-cellular cellobiase activity was not detected in CMC or Avicel media. This enzyme was detected in mycelial extracts only. With 1.7% Avicel liquid medium, the strain did not produce exocellobiohydrolase, but 74 µmol RBBR/mg protein/min was detected with 0.5% Avicel. Twenty-seven thermophilic and thermotolerant fungal strains were isolated from soil, decaying organic matter and sugarcane piles based on their ability to grow at 45ºC on medium containing corn straw and cardboard as carbon sources. These fungi were identified in the genera Aspergillus, Thermomyces, Myceliophthora, Thermomucor and Candida. The efficiency of enzymatic hydrolysis of cellulose can be improved by various pretreatments of the substrate. In order to increase the efficiency of enzymatic saccharification of the wheat straw, we determined the effect of different pretreatments on the physical structure, chemical components and enzymatic saccharification of wheat straw. Our results showed that combination of grinding and sodium hydroxide (NaOH) treatment had high effect on the enzymatic hydrolysis of wheat straws.
