Structure and secretory activity of cultured chondrocytes from patients with osteoarthritis Biocell

Cartilage samples were taken from OA patients in order to describe and quantify pro-inflammatory mediators. Samples were cultured under aseptic conditions in Dulbecco's modified Eagle medium at 37°C for 10 days. Control samples, taken from non-inflammatory cartilage, were cultured under the same conditions. The levels of NO-2 and NO-3 were measured in the supernatant using a spectrophotometric assay. The activity of MMP-1 was quantified by ELISA. The concentration of NO-x was 47.3 ± 4.1 µM in the OA cartilague and 10.7 ± 1.8 µM in the controls. The average MMP-1 activity was 3,650 ± 387 ng/ml in the OA cartilage and 2,150 ± 190 ng/ml in the control samples. Statistical Optimisation of Fermentation Media for Novel Clostridium novyi -NT Using Response Surface Methodology. BABT

ABSTRACT Clostridial collagenase is recognized as one of the important proteolytic enzymes used in treatment of varieties of fibro-proliferative disorders. The present work aimed to optimise the biomass concentration and collagenase enzyme activity of novel bacilli Clostridium novyi-NT. The response surface methodology tool was used to identify the optimal fermentation parameters. Central composite design (CCD) was applied with respect to three influencing factors - pH, proteose peptone and trypticase soya broth. These factors showed significant effect on the overall biomass concentration and collagenase enzyme activity (p<0.05). The treatment of some mesenchymal malignancies has made significant gains over the past few decades with the development of effective systemic therapies. In contrast, the treatment of chondrosarcoma has been limited to surgical resection, with the most significant prognostic indicators being surgical margins and histologic grade. We have reported that MMP-1/TIMP-1 gene expression serves to prognosticate for tumor recurrence in this group of patients. There are few complete reports that fully uncover production, characterization and purification of fungi collagenases.
