Detoxification of Pacific oyster Crassostrea gigas fed on diets of Skeletonema costatum with and without silt, following PSP contamination by Alexandrium minutum ArchiMer
Gueguen, Marielle; Bardouil, Michele; Baron, Regis; Lassus, Patrick; Truquet, Philippe; Massardier, Julie; Amzil, Zouher.
Contamination of shellfish by paralytic shellfish poisoning ( PSP) toxins poses an economic threat to shellfish farmers. As contaminated shellfish cannot be harvested for long periods of time, it would be very useful to develop processes to optimise and shorten their detoxification. In this study, Pacific oysters Crassostrea gigas were first experimentally contaminated over a period of 13 days with a continuous flow of toxic Alexandrium minutum cultures at concentrations ranging from 150 to 200 cell ml(-1) ( toxin content after 13 days of contamination 438 mu g STX equiv. 100 g(-1) wet weight). Then, two different detoxification treatments were tested and showed detoxification rates greater than those observed in coastal environments. The first treatment...

Barille-boyer, Anne-laure; Haure, Joel; Baud, Jean-pierre.
The oyster growth has been studied for a 10 year period, from 1986 to 1995, in four locations of the bay of Bourgneuf (French Atlantic Coast). The relations between oysters growth and the main molluscs stocks (cultivated oysters and wild mussels) or environmental factors (hydrobiology, currents, climatology) are analysed to understand the functioning of this shellfish ecosystem. As the water renewal time is about two months, primary production is essentially autochtonal. Phytoplankton development is controlled by the nutrients brought by the river Loire. An important part of the microalgac is composed by the microphytobenthos resuspended by west wind. High seston concentrations, measured on the North of the bay (150 mg/1 as an average), have a negative...

Impact of atrazine on somatic aneuploidy in cupped oysters, Crassostrea gigas ArchiMer
Bouilly, Karine; Leitao, Alexandra; Mccombie, Helen; Chaves, R.; Guedes Pinto, H; Boudry, Pierre; Lapegue, Sylvie.
In the Pacific oyster, Crassostrea gigas hypodiploid aneuploid cells with 2n=19, 18 or 17 chromosomes have regularly been reported. There is a negative correlation between somatic aneuploidy and growth rate in oysters (Leitão et al., 2001a) as well as evidence for a genetic basis (Leitão et al., 2001b). Furthermore, non-random chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique (Leitão et al., 2001c). This poster summarises different studies used to investigate the effects of the herbicide atrazine on the level of aneuploidy in this species

Arnaud, Christophe; Faury, Nicole; Bouquet, Jean-paul.
The Marennes-Oléron is a well known cupped oyster (Crassostrea gigas) and mussel (Mytilus edulis) reproduction area. Every year, a monitoring of the reproduction of these two species is carried out on behalf of the Regional Shellfish Culture Section of Marennes-Oléron. This monitoring aims to determine the periods of larval emission in order to let the professionals know about the best collection times This study presents the main results obtained in 1999 and compares them with those obtained in 1997 and 1998. The year 1999 showed good results as far as larval emission and development are concerned in both oysters and mussels. The comparison of the three consecutive years shows the disparity of larval evolutions.

L'huître creuse Crassostrea gigas en Bretagne. Résultats du réseau de suivi de 1991 ArchiMer
Le Bec, Claude; Mazurie, Joseph.
The year 1991 is the third one of the regional network on growth assessment of Crassostrea gigas in Brittany, wearing on 9 sites and 3 stations per site. 2 bags of 210 oysters, 18 month old, were placed in each station. Comparatively to previous years, the 1991 growth was moderate, and condition index inferior. However, the survival rates were very high (above 91 %) and most of the slight mortalities occurred a short time after stocking (as revealed by the sizes of dead shells). An improvement was also observed in the infestation rate by the polychaete Polydora sp..Most of the growth differences between sites and stations are still present. An analytical study including environmental variables would be necessary for explanations and will be tempted in 1993.

Oyster hemolymph is a complex and dynamic ecosystem hosting bacteria, protists and viruses ArchiMer
Dupont, Samuel; Lokmer, A.; Corre, E.; Auguet, Jean-christophe; Petton, Bruno; Toulza, E.; Montagnani, Caroline; Tanguy, G.; Pecqueur, D.; Salmeron, C.; Guillou, L.; Desnues, C.; La Scola, B.; Bou Khalil, J.; De Lorgeril, Julien; Mitta, Guillaume; Gueguen, Yannick; Escoubas, Jean Michel.
Background The impact of the microbiota on host fitness has so far mainly been demonstrated for the bacterial microbiome. We know much less about host-associated protist and viral communities, largely due to technical issues. However, all microorganisms within a microbiome potentially interact with each other as well as with the host and the environment, therefore likely affecting the host health. Results We set out to explore how environmental and host factors shape the composition and diversity of bacterial, protist and viral microbial communities in the Pacific oyster hemolymph, both in health and disease. To do so, five oyster families differing in susceptibility to the Pacific oyster mortality syndrome were reared in hatchery and transplanted into a...

Summer mortality of hatchery-produced Pacific oyster spat (Crassostrea gigas). II. Response to selection for survival and its influence on growth and yield ArchiMer
Degremont, Lionel; Bedier, Edouard; Boudry, Pierre.
Response to divergent selection for "high" and "low" survival during the summer period, from July to October, was investigated in juvenile (six-month-old) Pacific oysters, Crassostrea gigas, by producing two sets of progenies in 2002 (Generation 2) and three sets of progenies in 2003 (Generation 3). A strict between-family approach was used and resistance of these selected progenies to summer mortality was assessed in three sites along French coasts, to determine their response to selection and estimate realized heritability of the trait. A significant difference in survival was observed between the "high" and "low" selected groups at all sites for all sets of progenies, indicating a significant genetic component. High realized heritabilities for survival...

Additive transcriptomic variation associated with reproductive traits suggest local adaptation in a recently settled population of the Pacific oyster, Crassostrea gigas ArchiMer
Sussarellu, Rossana; Huvet, Arnaud; Lapegue, Sylvie; Quillen, Virgile; Lelong, Christophe; Cornette, Florence; Jensen, Lasse Fast; Bierne, Nicolas; Boudry, Pierre.
Background Originating from Northeast Asia, the Pacific oyster Crassostrea gigas has been introduced into a large number of countries for aquaculture purpose. Following introduction, the Pacific oyster has turned into an invasive species in an increasing number of coastal areas, notably recently in Northern Europe. Methods To explore potential adaptation of reproductive traits in populations with different histories, we set up a common garden experiment based on the comparison of progenies from two populations of Pacific oyster sampled in France and Denmark and their hybrids. Sex ratio, condition index and microarray gene expression in gonads, were analyzed in each progeny (n = 60). Results A female-biased sex-ratio and a higher condition index were...

Deslous-paoli, Jean-marc; Heral, Maurice.
Caloric values of the ash-free dried sofkt parts of Crassostrea gigas have been characterized, according to season using a microcalorimeter and biochemical composition. This study shows the difference between the two methods. A conversation factor (8,63 kcal-gr(-1) has been determined for lipid of pacific oyster. Thix work shows that reproduction implies a loss of energy (53% to 63%) for non juvenile oysters. The increase of energetic level necessary to growth is more important for juvenile oysters and is upper than what is lost for reproduction.

Impact of 20 : 4n-6 supplementation on the fatty acid composition and hemocyte parameters of the Pacific oyster Crassostrea gigas ArchiMer
Delaporte, Maryse; Soudant, Philippe; Moal, Jeanne; Giudicelli, Emilie; Lambert, Christophe; Seguineau, Catherine; Samain, Jean-francois.
Arachidonic acid (20:4n-6, ArA) and its eicosanoid metabolites have been demonstrated to be implicated in immune functions of vertebrates, fish, and insects. Thus, the aim of this study was to assess the impact of ArA supplementation on the FA composition and hemocyte parameters of oysters Crassostrea gigas. Oyster dietary conditioning consisted of direct addition of ArA solutions at a dose of 0, 0.25, or 0.41 mu g ArA per mL of seawater into tanks in the presence or absence of T-Iso algae. Results showed significant incorporation of ArA into gill polar lipids when administered with algae (up to 19.7%) or without algae (up to 12.1%). ArA supplementation led to an increase in hemocyte numbers, phagocytosis, and production of reactive oxygen species by...

Mary, Charlotte; Pien, S; Blin, J.l.; Ropert, Michel.
The regional monitoring network REMONOR, relying on the experience of IFREMER's REMORA national network, each year assesses the mortality, growth and quality for two age classes ("adults" = 2 years and "juveniles" = 1 year) of cup oysters, distributed among various farm sites in the main oyster farming sectors of Lower Normandy. This network, led in collaboration by LERN (Environment Resources Laboratory) and SMEL (Coastline Installations Public-Private Entity), this year welcomes a new farm site in Veules les Roses, in Seine Maritime (76), bringing to 18 the total number of monitored sites. For operational reasons, a reduced monitoring protocol was conducted in 2005 on this last farm site, which will not take on the network's standardised protocol until...

Degremont, Lionel; Moal, Jeanne; Daniel, Jean-yves; Boudry, Pierre; Van Vormhoudt, A.; Sellos, D.; Samain, Jean-francois.
In the scope of the European program GENEPHYS, which aims to establish the relationship between the genetic base and the physiological characters in relation to the growth in Crassostrea gigas, it has been demonstrated that the most explanatory factors for the growth variability are assimilation performance and aneuploidy. A model linking the assimilation performances and the digestive equipment has been set up and more than 70% of the variability can be explained by two digestive enzymes: laminarinase and amylase. A number of laboratories have decided to focus on the latter.

Barille, Laurent.
Recent feeding-physiology experiments have shown that the Japanese oyster Crassostrea gigas was able to operate a selection among inorganic and organic particles before ingestion. However, the underlying mechanisms are not well understood. Anatomical observations of different structures which may play a raie in the selection processes are brought together and submitted to the actual knowledge on bivalve feeding. The mantle lobes, the ciliary structure of the ordinary filaments, the marginal ventral groove and the labial palps are described.
