Schistosomes utilize proteinases to accomplish several activities such as tissue penetration, tissue digestion and evasion of host immune responses. Cathepsin L is a cysteine proteinase of the papain superfamily detected in their gut lumen which indicates that this enzyme contributes to the proteolysis of ingested hemoglobin. Due to the roles played in the schistosome biology, proteolytic enzymes are considered potential targets for developing and guiding antischistosomal therapies. Fruits were collected from trees of Coffea arabica cv. Obatã grown at Mococa and Adamantina in São Paulo State, Brazil, which are regions with marked differences in air temperature that produce coffee with distinct qualities. Mococa is a cooler location that produces high-quality coffee, whereas coffee from Adamantina is of lower quality. The amino acid and protein contents, amino acid profile, and proteinase activity and type in endosperm protein extracts were analysed. Proteinase genes were identified, and their expression was assayed. All results indicate that temperature plays a role in controlling proteinase activity in coffee endosperm. In this study, proteins extracted from laticifer cells of three plants were examined by electrophoresis, mass spectrometry (MALDI-TOF) and characterized in respect of proteolytic, chitinolytic and anti-oxidative activities by means of zymography and colorimetric assays. Acidic proteins with molecular masses between 12.5 and 74.5 kDa predominated in laticifers of P. rubra. This profile was not found in laticifers of C. grandiflora and E. tirucalli. The later was poor in respect of proteins.
