In vitro growth of Brassocattleya orchid hybrid in different concentrations of KNO3, NH4NO3 and benzylaminopurine Horticultura Brasileira
Cardoso,Jean C; Ono,Elizabeth O.
One of the most important applications of plant tissue culture is mass propagation of ornamental plants. This experiment evaluated the effect of different concentrations of NH4NO3 and KNO3 and BAP on the in vitro growth of orchid hybrid Brassocattleya 'Pastoral'. Seedlings of this orchid hybrid were used as explants and cultivated in medium with mineral salts and vitamins from the MS medium (Murashige & Skoog, 1962), with the macronutrients P, Ca and Mg reduced by half, and with an addition of 25 g L-1 of sucrose, 0.1 g L-1 of myo-inositol and 1.5 g L-1 of activated charcoal. Agar-agar was added (6.5 g L-1) and the pH was adjusted to 5.8. As treatments, four concentrations of the NH4NO3 and KNO3 (2x; 1x; ½ and ¼ MS medium) and three concentrations...

Exogenous application of phytohormones mitigates the effect of salt stress on Carica papaya plants AGRIAMBI
Sá,Francisco V. da S.; Brito,Marcos E. B.; Silva,Luderlândio de A.; Moreira,Rômulo C. L.; Paiva,Emanoela P. de; Souto,Lauter S..
ABSTRACT The salinity has complex effects on the physiological, nutritional and hormonal interactions of plants. This study aimed to evaluate the mitigating action of exogenous application of different types and concentrations of phytohormones on Carica papaya under salt stress. The experiment was conducted in greenhouse, using a randomized block design with treatments formed from a 2 x 3 x 5 factorial scheme, relative to two concentrations of salts in the irrigation water (0.6 and 2.4 dS m-1), three types of phytohormones [Auxin = indole-3-acetic acid; Cytokinin = CPPU (N-(2-chloro-pyridyl)-N-phenylurea) and Gibberellin = gibberellic acid(AG3)], and five concentrations of the phytohormone (0, 12.5, 25.0, 37.5 and 50.0 µM L-1), with three repetitions,...

Micropropagation of Ilex dumosa (Aquifoliaceae) from nodal segments in a tissue culture system Biocell
Luna,C.; Sansberro,P.; Mroginski,L.; Tarragó,J..
Micropropagation of Ilex dumosa var. dumosa R. ("yerba señorita") from nodal segments containing one axillary bud was investigated. Shoot regeneration from explants of six-year-old plants was readily achieved in ¹/4 strength Murashige and Skoog medium (¹/4 MS) plus 30 gr·L-1 sucrose and supplemented with 4.4 mM BA. Further multiplication and elongation of the regenerated shoots were obtained by subculture in a fresh medium of similar composition with 1.5 gr·L-1 sucrose. Rooting induction from shoots were achieved in two steps: 1) 7 days in ¹/4 MS (30 gr·L-1 sucrose, 0.25 % Phytagel®) with 7.3 mM IBA and 2) 21 days in the same medium without IBA and 20 mM of cadaverine added. Regenerated plants were successfully transferred to soil. This micropropagation...

Desiccation tolerance in seeds of Annona emarginata (Schldtl.) H. Rainer and action of plant growth regulators on germination Braz. J. Plant Physiol.
Corsato,Jaqueline Malagutti; Ferreira,Gisela; Barbedo,Claudio Jose.
Annona emarginata (Schldtl.) H. Rainer ("araticum-de-terra-fria") is used as a rootstock for several species of Annonaceae. It is suggested that these seeds should be sown immediately after extraction and, therefore, they could be intolerant to desiccation. There are several mechanisms involved with desiccation tolerance. Soluble sugars, for example, can accumulate and act as osmoprotectants for the membrane system during desiccation. The aim of this study is to assess desiccation tolerance in A. emarginata seeds. In addition, we examined the soluble sugars involved in desiccation tolerance. Finally, we determined the effect of gibberellic acid (GA4+7) and N-(phenylmethyl)-aminopurine in promoting the germination of seeds with different water contents. The...

Growth regulators and essential oil production Braz. J. Plant Physiol.
Prins,Cláudia L; Vieira,Ivo J. C; Freitas,Silvério P.
The aroma and fragrance industry is a billion-dollar world market which grows annually. Essential oils comprise the majority of compounds used by these industries. These sets of metabolites are formed mainly by monoterpenes, which are products of the plants' secondary metabolism. Biosynthesized from mevalonate and methylerythitol phosphate, the essential oil production depends not only on genetic factors and the developmental stage of plants, but also on environmental factors which could result in biochemical and physiological alterations in plants modifying the quantity and quality of the essential oil. These modifications impair aromatic plant production aimed at essential oil by reducing their quality. It is desirable to develop techniques of...

Effects of light and ethylene on endogenous hormones and development of Catasetum fimbriatum (Orchidaceae) Braz. J. Plant Physiol.
Suzuki,Rogério M.; Kerbauy,Gilberto B..
This study attempted to clarify the effects of dark, light and ethylene on plant growth and endogenous levels of indole-3-acetic acid (IAA), cytokinins and abscisic acid in Catasetum fimbriatum. Dark-incubation fully inhibited root and pseudobulb formation as well as leaf growth, but favored shoot elongation. The results of continuous and active growth in dark-incubated shoots (stolons) were induced by strong apical meristem sink activity and by the significantly increased levels of cytokinins in shoots. In fact, shoot length, cytokinin and IAA levels in dark-incubated shoots were about twice as great as for those grown under light conditions. Moreover, the total cytokinin level in shoots of C. fimbriatum under light conditions without ethylene was...

Micropropagation of Stevia rebaudiana plants Ciência Rural
Rokosa,Marta Teresa; Kulpa,Danuta.
ABSTRACT: The aim of the study was to develop optimum composition of plant growth regulators in media for the propagation and rooting of shoots of stevia (Stevia rebaudiana Bertoni) in in vitro cultures. Single-node shoot fragments obtained from plants propagated on MS medium were placed onto media supplemented with: BAP, 2iP and KIN at concentrations: 0.5, 1, 2 and 5 mg∙dm-3, whereas at the rooting stage with addition of: IAA, IBA and NAA at concentrations 1, 2, 4 and 8 mg∙dm-3. The highest number of shoots and leaves was reported for plants propagated on MS medium enriched with 0.5 mg∙dm-3 BAP. The greatest number of the longest roots was developed by stevia on the MS medium enriched with 1 mg∙dm-3 IAA.
