Development of genomic probes for detection of hepatitis A virus and enteroviruses in shellfish ArchiMer
Apaire, M; Leguyader, F; Besse, B; Billaudel, S.
The development of molecular biology techniques has modified research procedures for certain viruses, particularly hepatitis A virus (HAV) and enteroviruses. Accordingly, we designed cDNA and RNA genomic probes to detect viruses in shellfish in the environment. Better sensitivity was noted with RNA than cDNA probes: only 17/83 shellfish samples were positive with HAV and enterovirus DNA probes, where as 42/83 were positive with HAV and enterovirus RNA probes.

Human papillomavirus detection and typing using a nested-PCR-RFLP assay BJID
Coser,Janaina; Boeira,Thaís da Rocha; Fonseca,André Salvador Kazantzi; Ikuta,Nilo; Lunge,Vagner Ricardo.
BACKGROUND: It is clinically important to detect and type human papillomavirus (HPV) in a sensitive and specific manner. OBJECTIVES: Development of a nested-polymerase chain reaction-restriction fragment length polymorphism (nested-PCR-RFLP) assay to detect and type HPV based on the analysis of L1 gene. METHODS: Analysis of published DNA sequence of mucosal HPV types to select sequences of new primers. Design of an original nested-PCR assay using the new primers pair selected and classical MY09/11 primers. HPV detection and typing in cervical samples using the nested-PCR-RFLP assay. RESULTS: The nested-PCR-RFLP assay detected and typed HPV in cervical samples. Of the total of 128 clinical samples submitted to simple PCR and nested-PCR for detection of HPV,...

Long-term evaluation of the antimicrobial susceptibility and microbial profile of subgingival biofilms in individuals with aggressive periodontitis BJM
Lourenço,Talita Gomes Baêta; Heller,Débora; Souto,Renata Martins do; Silva-Senem,Mayra Xavier e; Varela,Victor Macedo; Torres,Maria Cynesia Barros; Feres-Filho,Eduardo Jorge; Colombo,Ana Paula Vieira.
This study evaluates the antimicrobial susceptibility and composition of subgingival biofilms in generalized aggressive periodontitis (GAP) patients treated using mechanical/antimicrobial therapies, including chlorhexidine (CHX), amoxicillin (AMX) and metronidazole (MET). GAP patients allocated to the placebo (C, n = 15) or test group (T, n = 16) received full-mouth disinfection with CHX, scaling and root planning, and systemic AMX (500 mg)/MET (250 mg) or placebos. Subgingival plaque samples were obtained at baseline, 3, 6, 9 and 12 months post-therapy from 3–4 periodontal pockets, and the samples were pooled and cultivated under anaerobic conditions. The minimum inhibitory concentrations (MICs) of AMX, MET and CHX were assessed using the microdilution...

Prevalence of "non-oral" pathogenic bacteria in subgingival biofilm of subjects with chronic periodontitis BJM
Souto,Renata; Andrade,Arnaldo Feitosa B. de; Uzeda,Milton; Colombo,Ana Paula Vieira.
The oral cavity may act as a reservoir for several pathogens related to systemic infections. Therefore, the purpose of this study was to determine the prevalence and levels of pathogenic bacteria in the subgingival biofilm of chronic periodontitis lesions and healthy periodontal sites using the checkerboard DNA-DNA hybridization technique. 200 samples of subgingival biofilm from sites with periodontitis (probing pocket depth > 4 mm and /or clinical attachment level > 4 mm) and 200 samples from healthy sites of 14 patients with chronic periodontitis, as well as 200 samples from 3 periodontally healthy patients were obtained. The presence and levels of 11 pathogenic bacteria were determined using whole genomic DNA probes and the checkerboard method,...
