Hundred Fusarium culmorum strains, isolated from freshly harvested maize grain samples from Southern parts of India, were incubated in czapek-dox medium and analyzed for trichothecene (DON/NIV) production. The mPCR assay was standardized targeting trichothecene metabolic pathway genes viz., Tri6, Tri7, Tri13 for detection of trichothecene (DON/NIV) chemotypes and rDNA gene for specific detection of F. culmorum species. Primers for targeted genes were designed and used to predict whether these isolates could produce deoxynivalenol/nivalenol, 94 isolates were able to produce DON/NIV by mPCR assay. The incidence of the trichothecenes deoxynivalenol (DON), diacetoxyscirpenol (DAS), and T2 toxin (T2) in popcorn was investigated in 90 samples, belonging to 2 commercial and 28 experimental hybrids planted at experimental stations of the Agronomic Institute of Campinas at the locations of Campinas, Mococa, and Capão Bonito, and in 15 samples of popcorn, 9 branded and 6 unbranded, acquired from commercial outlets in the city of Campinas, SP. The samples were analyzed by gas chromatography with flame ionization detector combined with an alumina:carbon cleanup column. The toxicity of deoxynivalenol, both intravenously and orally, was investigated in male and female BALB/c mice. Technetium-99m (99m Tc)-labeled deoxynivalenol was administered to mice by tail vein injection and orally dosed. Distribution of labeled deoxynivalenol at 26 hours was monitored by gamma-scintigraphy. In the evaluated organs, the accumulation of radioactive deoxynivalenol was correlated with the amount of radioactivity. In addition, the toxicity of deoxynivalenol was measured by biochemical assays followed by histopathological findings. Deoxynivalenol (DON) is an important Fusarium toxin of concern for food safety. The inhalation of powder contaminated with DON is possible and may cause lung toxicity. In this study, we analyzed the gene expression profile of A549 cells treated for 24 hr with 0.2 µg/mL DON by microarray analysis. In total, 16 genes and 5 noncoding RNAs were significantly affected by DON treatment. The repression of B3GALT4, MEIS3, AK7, SEMA3A, KCNMB4, and SCARA5 was confirmed by quantitative PCR. We investigated the DON toxicity on A549 cells that exogenously expressed these 6 genes. ABSTRACT: The effects of mycotoxins on the productive performance of growing pigs and broilers were evaluated using meta-analytical approach. Two databases were constructed: (1) Broilers, with information collected from 51,497 birds and published in 158 scientific papers from 1980 to 2016; and (2) Pigs, with information collected from 7,743 animals and published in 72 scientific papers from 1980 to 2015. The meta-analyses were performed independently for each specie, following three sequential analyses: graphical, correlation, and variance-covariance.
