Profile of the alcohols produced in fermentations with malt contaminated with trichothecenes BABT
Reinehr,Christian Oliveira; Furlong,Eliana Badiale.
In order to study the influence of mycotoxins on the production of alcohols, a fermentative process on a laboratorial scale was simulated. Malt was contaminated with deoxynivalenol and T-2 in different ratios (up to 500 ppb), according to a 3² factorial design, and the alcohols obtained after the fermentation were determined through gas chromatography. The results showed that trichothecenes influenced the profile of the alcohols produced by Saccharomyces cerevisiae during the fermentative process of malt, especially the profile of methyl and isoamyl alcohols.

Incidence and multiplex PCR based detection of trichothecene chemotypes of Fusarium culmorum isolates collected from freshly harvested Maize kernels in Southern India BJM
Venkataramana,M.; Shilpa,P.; Balakrishna,K.; Murali,H.S.; Batra,H.V..
Hundred Fusarium culmorum strains, isolated from freshly harvested maize grain samples from Southern parts of India, were incubated in czapek-dox medium and analyzed for trichothecene (DON/NIV) production. The mPCR assay was standardized targeting trichothecene metabolic pathway genes viz., Tri6, Tri7, Tri13 for detection of trichothecene (DON/NIV) chemotypes and rDNA gene for specific detection of F. culmorum species. Primers for targeted genes were designed and used to predict whether these isolates could produce deoxynivalenol/nivalenol, 94 isolates were able to produce DON/NIV by mPCR assay. Chemical analysis of DON/NIV was carried out for mPCR positive isolates by high performance-thin layer chromatography (HPTLC). To check the practical usefulness of...

Parboiled rice: chemical composition and the occurrence of mycotoxins Ciênc. Tecnol. Aliment.
Dors,Giniani Carla; Bierhals,Vânia da Silva; Badiale-Furlong,Eliana.
The objective of this work was to evaluate the incidence of aflatoxin B1 (AFB B1), deoxynivalenol (DON), ochratoxin A (OTA), and zearalenone (ZEA) in parboiled rice with respect to its chemical composition. Eight lots from five different brands of parboiled rice were collected in four samplings, at different seasons, until the amount of 32 lots. It was observed that: DON was present in 22% of the samples (from 180 to 400 ppb); ZEA in 19% (from 317 to 396 ppb); OTA in 12.5% (from 13 and 26 ppb); and AFB B1 in 9% (from 11 to 74 ppb). The results of the chemical composition were not different from those previously mentioned in the literature concerning parboiled rice. The ash and phenol levels in the contaminated parboiled rice samples suggested that those...

Efficiency of γ-oryzanol against the complex Fusarium graminearum growth and mycotoxins production Ciênc. Tecnol. Aliment.
BEMVENUTI,Renata; RODRIGUES,Marcy Heli; FURLONG,Eliana.
Abstract The ability of γ-oryzanol against Fusarium graminearum growth and mycotoxins production were assessed and compared with the antioxidant properties in order to improve its application in food chain, beyond of the conventional. The γ-oryzanol extracted from rice bran and characterized by HPLC-UV and GG-FD. Its antifungal in vitro, effect on complex F. graminearum CQ244 (halo diameter, ergosterol and glucosamine production) and mycotoxin production (deoxinivalenol, nivalenol and zearalenone) were determined by an adapted method adapted. The antifungal activity of γ-oryzanol was compared with azoxystrobin and trifloxystrobin. MIC and specific inhibitory effects (%inhibition µg γ-oryzanol-1) were also estimated. The antioxidant activity of the...

Technetium-99m-labeled deoxynivalenol from Fusarium mycotoxin alters organ toxicity in BALB/c mice by oral and intravenous route J. Venom. Anim. Toxins incl. Trop. Dis.
Chattopadhyay,P; Pandey,A; Goyary,D; Chaurasia,A; Singh,L; Veer,V.
The toxicity of deoxynivalenol, both intravenously and orally, was investigated in male and female BALB/c mice. Technetium-99m (99m Tc)-labeled deoxynivalenol was administered to mice by tail vein injection and orally dosed. Distribution of labeled deoxynivalenol at 26 hours was monitored by gamma-scintigraphy. In the evaluated organs, the accumulation of radioactive deoxynivalenol was correlated with the amount of radioactivity. In addition, the toxicity of deoxynivalenol was measured by biochemical assays followed by histopathological findings. Kidney and hepatic marker enzymes were significantly increased in intravenously administered deoxynivalenol as compared to orally treated mice. Intravenously treated mice showed severe damage in liver and kidney...

MEIS3 is repressed in A549 lung epithelial cells by deoxynivalenol and the repression contributes to the deleterious effect OAK
Toyotome, Takahito; Takahashi, Hiroki; Kamei, Katsuhiko.
Deoxynivalenol (DON) is an important Fusarium toxin of concern for food safety. The inhalation of powder contaminated with DON is possible and may cause lung toxicity. In this study, we analyzed the gene expression profile of A549 cells treated for 24 hr with 0.2 µg/mL DON by microarray analysis. In total, 16 genes and 5 noncoding RNAs were significantly affected by DON treatment. The repression of B3GALT4, MEIS3, AK7, SEMA3A, KCNMB4, and SCARA5 was confirmed by quantitative PCR. We investigated the DON toxicity on A549 cells that exogenously expressed these 6 genes. The result indicated that A549 cells that transiently expressed MEIS3 were tolerant to the deleterious effects of DON. Our data show that DON affected the expression of genes with various...

Assessing the implications of mycotoxins on productive efficiency of broilers and growing pigs Scientia Agricola
Kipper,Marcos; Andretta,Ines; Ribeiro,Andréa Machado Leal; Pires,Paula Gabriela da Silva; Franceschina,Carolina Schell; Cardinal,Kátia Maria; Moraes,Priscila de Oliveira; Schroeder,Bruna.
ABSTRACT: The effects of mycotoxins on the productive performance of growing pigs and broilers were evaluated using meta-analytical approach. Two databases were constructed: (1) Broilers, with information collected from 51,497 birds and published in 158 scientific papers from 1980 to 2016; and (2) Pigs, with information collected from 7,743 animals and published in 72 scientific papers from 1980 to 2015. The meta-analyses were performed independently for each specie, following three sequential analyses: graphical, correlation, and variance-covariance. Broilers and pigs challenged by mycotoxins reduced (p < 0.05) feed intake by 9 and 6 %, weight gain by 15 and 11 %, and feed efficiency by 6 and 4 % compared with non-challenged animals, respectively....
