ABSTRACT The aim of this study is the production, purification, and characterisation of thermostable raw starch hydrolyzing α-amylase produced by Bacillus mojavensis SO-10. The maximum production conditions of α-amylase were found at 36th hour, 35 °C and pH 7.0. We utilized three steps to purify the thermostable α-amylase and as a result, 34-fold and 18% yield were obtained. The molecular weight of purified α-amylase was determined as 73 kD. The Km and Vmax rates were detected as 0.010 mM and 3.38 µmol min−1, respectively. This purified α-amylase exhibited the highest activity at pH 5.0-6.0 and 70 ºC and showed stability over a wide variety of pH and temperature at 4.0-8.0, and 40-50 ºC, respectively. This study aimed to investigate the toxicological effects of the LAS-C12 on Mugil platanus (mullet). Fishes exposed to 1.0 mg.L-1 for 24, 48, 72, 96 and 120 hours presented significant increase in specific routine metabolism. At the concentration of 0.5 mgL-1, the active metabolism presented a decreasing trend from 48 h of exposure on. However, only the consumption averages for 72 h were statistically different from the ones obtained for other periods of exposure. The lowest oxygen consumption in this concentration was observed for 24, 48 and 72 hours of exposure. Protease and α-amylase production by a thermophilic Bacillus sp. SMIA-2 cultivated in liquid cultures containing 0.25% (w/v) starch as a carbon source reached a maximum at 18 hours (47 U.mg-1 Protein) and 36 hours (325 U.mg-1 Protein), respectively. Culture medium supplementation with whey protein concentrate (0.1%, w/v) and corn steep liquor (0.3%, w/v) not only improved the production of both enzymes but also enabled them to be produced simultaneously.
