Diaporthe (Phomopsis) species have often been reported as plant pathogens, non-pathogenic endophytes or saprobes, commonly isolated from a wide range of hosts. The primary aim of the present study was to resolve the taxonomy and phylogeny of a large collection of Diaporthe species occurring on diverse hosts, either as pathogens, saprobes, or as harmless endophytes. In the present study we investigated 243 isolates using multilocus DNA sequence data. Analyses of the rDNA internal transcribed spacer (ITS1, 5.8S, ITS2) region, and partial translation elongation factor 1-alpha (TEF1), beta-tubulin (TUB), histone H3 (HIS) and calmodulin (CAL) genes resolved 95 clades. ABSTRACT We studied the fungal endophyte community of the leaves of Myracrodruon urundeuva in Brazil along with their potential to produce L-asparaginase. In total, 187 endophytes were isolated. The colonization rate of leaf fragments from caatinga (10.41 %) was lower than those from brejo de altitude (39.58 %). Sequences of ITS rDNA from all endophytes revealed relationships with Ascomycota (Botryosphaeriales, Chaetothyriales, Diaporthales, Eurotiales, Glomerellales, Hypocreales, and Pleosporales) and Basidiomycota (Polyporales). ABSTRACT Plants harbour diverse communities of fungal species in their internal compartments. Endophytic fungi help their hosts to establish, survive, and adapt to different environments. Here, we examined the diversity of endophytic fungi in the leaflets and branches of Poincianella pyramidalis, a plant species endemic to the Brazilian tropical dry forest (Caatinga). A total of 360 fragments of leaflets and branches were analysed and 189 endophytic fungi were isolated and distributed among 21 ascomycetous genera based on their ITS and LSU rDNA sequences. Abstract Production of a bioherbicide for biological control of weeds requires a series of steps, from selection of a suitable microbial strain to final formulation. Thus, this study aimed to select fungi for production of secondary metabolites with herbicidal activity using biological resources of the Brazilian Pampa biome. Phytopathogenic fungi were isolated from infected tissues of weeds in the Pampa biome. A liquid synthetic culture medium was used for production of metabolites. The phytotoxicity of fungal metabolites was assessed via biological tests using the plant Cucumis sativus L., and the most promising strain was identified by molecular analysis.
