Virulence factors of Escherichia coli isolated from diarrheic calves Arq. Bras. Med. Vet. Zootec.
Rigobelo,E.C.; Gamez,H.J.; Marin,J.M.; Macedo,C.; Ambrosin,J.A.; Ávila,F.A..
One hundred seventy-three Escherichia coli strains isolated from calves from northwestern São Paulo State, having diarrhea were examined for the production of thermolabile (LT) and thermostable (ST) enterotoxins and for the presence of virulence factors associated with bovine colibacillosis. Eighty-five (49.1%) of the E.coli strains produced toxins; 53 isolates were detected as producing STa toxin, and 9 also produced LT toxin. By PCR, 23 isolates were shown to harbor only the LT-II gene. Nine (5.2%) isolates harbored Shiga toxin genes: four carried the stx2 gene, four the stx1 gene and one carried both. Three of the isolates showing stx1 also carried the eae gene. Among the E. coli isolates examined for susceptibility to 10 antimicrobial agents,...

Evaluation of a multiplex PCR method to detect enteroaggregative Escherichia coli Biocell
Rüttler,M.E.; Yanzón,C.S.; Cuitiño,M.J.; Renna,N.F.; Pizarro,M.A.; Ortiz,A.M..
Enteroaggregative Escherichia coli (EAEC) has been implicated in sporadic diarrhea in children and adults and has been identified as the cause of several outbreaks worldwide. The HEp-2 test remains the gold standard for identification of this pathotype. A 60-65 MDa plasmid encodes the aggregative adherence fimbriae (AAF/I and AAF/II), a transcriptional activator ( aggR gene), the enteroaggregative heat-stable enterotoxin EAST1 ( astA gene) and a cytotoxin (Pet). The standard assay for EAEC is performed only in research laboratories, because it is expensive, labor intensive and time-consuming. The Polymerase Chain Reaction (PCR) offers the possibility of rapid diagnosis. In the current study, a multiplex PCR assay which checks aggR and astA genes was...

An outbreak of winter dysentery caused by bovine coronavirus in a high-production dairy cattle herd from a tropical country BABT
Takiuchi,Elisabete; Barry,Aline Fernandes; Alfieri,Alice Fernandes; Filippsen,Patrícia; Alfieri,Amauri Alcindo.
Bovine coronavirus (BCoV) is a known cause of winter dysentery (WD) in adult cattle. The morbidity of the disease is high, that results in a significant decrease in milk production and consequently, economic losses. In the present study, we report on a classical outbreak of WD that affected a high-production Holstein dairy herd raised in a tropical country. The lactating batch included 154 cows, and 138 (90%) presented diarrhea in a short (nine days) period of time. Three (2%) cows died. The other batches of animals did not become ill. The evolution of the disease in the herd, including the clinical signs and epidemiological features, strongly suggested a WD case. Semi-nested PCR and RFLP confirmed that BCoV was the cause of the infection. Samples tested...

Bovine coronavirus detection in a collection of diarrheic stool samples positive for group a bovine rotavirus BABT
Barry,Aline Fernandes; Alfieri,Alice Fernandes; Stipp,Danilo Tancler; Alfieri,Amauri Alcindo.
Neonatal diarrhea is an important cause of economic losses for cattle farmers. The main viral etiologies of enteric diseases are group A rotaviruses (GARV) and the bovine coronavirus (BCoV). Although both viruses infect calves of the same age, the occurrence of mixed infections is still under studied. The present study describes the co-infection of BCoV and GARV in stool samples. Forty-four diarrheic fecal samples from calves up to 60 days old that had previously tested positive for GARV by SS-PAGE were analyzed using semi-nested PCR for BCoV. A product with 251 bp of the BCoV nucleoprotein gene was amplified in 15.9% (7/44) of the samples, demonstrating that co-infection is not an unusual event. These results reinforce the need for testing for both GARV...

Isolation in HRT-18 cells and molecular analysis of a BCoV strain from diarrheic feces of naturally infected calves BABT
Stipp,Danilo Tancler; Barry,Aline Fernandes; Alfieri,Alice Fernandes; Bodnar,Lívia; Alfieri,Amauri Alcindo.
Bovine coronavirus (BCoV) may cause acute diarrhea in newborn calves, leading to significant economic losses for cattle farmers. There are several diagnostic techniques used to detect BCoV in calf fecal samples, but virus isolation still has advantages for antigenic and genomic characterization. This study describes the isolation in HRT-18 cells and molecular characterization of Brazilian BCoV wild-type strains. Three fecal samples from diarrheic 30 day-old calves were inoculated in HRT-18 cell monolayers, which were then evaluated for HA titers and tested using semi-nested PCR followed by RFLP and sequencing. Two samples were successfully isolated and presented HA titers of 16 and 32 units per 25 mL. The results were confirmed using semi-nested PCR and...

Epidemiological and microbiological aspects of acute bacterial diarrhea in children from Salvador, Bahia, Brazil BJID
Diniz-Santos,Daniel R.; Santana,José S.; Barretto,Junaura R.; Andrade,Maria Goreth M.; Silva,Luciana R..
In the few cases of acute childhood diarrhea that require antimicrobial therapy, the correct choice of the drug depends on detailed previous knowledge of local strains. In order to establish such parameters in our city, we reviewed the results of all 260 positive stool cultures of children between 0 and 15 years of age during two years at a pediatric tertiary care facility in Salvador, Brazil. Bacterial strains had been presumptively identified by culturing in selective media and by biochemical testing, and their antimicrobial susceptibility patterns were automatically detected by the MicroScan Walkaway System. Data about patients' sex and age, monthly distribution of the cases, pathogens isolated and their antimicrobial resistance patterns were recorded....

Antibiotics for the empirical treatment of acute infectious diarrhea in children BJID
Diniz-Santos,Daniel R.; Silva,Luciana R.; Silva,Nanci.
While the routine use of antibiotics for infectious diarrhea in children must be avoided, because it brings little benefit in most cases and is associated with the risk of increasing antimicrobial resistance, selected cases may require antimicrobial therapy, and the choice of the antimicrobial agent often has to be made empirically. Physicians prescribing antimicrobials in such a setting have not only to be aware of the most likely pathogens, but also of their characteristic antimicrobial susceptibility pattern and the safety profile of the various drugs. We reviewed the literature on the use of ampicillin, beta-lactamase inhibitors, trimethoprim-sulfamethoxazole, chloramphenicol, tetracyclines, nalidixic acid, fluoroquinolones, third-generation...
