Giongo,Adriana; Beneduzi,Anelise; Gano,Kelsey; Vargas,Luciano Kayser; Utz,Laura; Passaglia,Luciane Maria Pereira.
Microorganisms that live inside and around a plant can supply it with essential substances, such as phytohormones and essential nutrients. The present investigation aimed to isolate and characterize the phyllosphere, the endophytic, and the water tank bacteria associated with Vriesea gigantea and Tillandsia aeranthos. The bacteria were tested for siderophore and indole-3-acetic acid (IAA) production, phosphate solubilization, and presence of the nif H gene. Genetic diversity of the bacterial isolates was evaluated by rep-PCR. Sixty-eight bacterial strains were isolated from 3 different microhabitats of V. gigantea and from 2 microhabitats of T. aeranthos bromeliad plants. Gram-positive, spore-forming bacilli comprised most bacterial isolates. All isolates...

Numoto,Alberto Yuji; Vidigal Filho,Pedro Soares; Scapim,Carlos Alberto; Franco,Antônio Augusto Nogueira; Ortiz,Alex Henrique Tiene; Marques,Odair José; Pelloso,Murilo Fuentes.
ABSTRACT The inoculation of sweet corn seeds with Azospirillum brasilense in association to nitrogen fertilizer may be an agronomic alternative for increasing the crop yield and net income of plant growers. Therefore, the aim of this study was to investigate the effect of the different doses of inoculant (Azospirillum brasilense) associated to the nitrogen fertilization management on the phenotypic traits of one sweet corn hybrid in summer growing periods, under supplemental irrigation, in the Northwestern Paraná state, Brazil. The experiment followed the complete randomized block design with four replications. The treatments were: i) five inoculant doses (0.0, 50, 100, 150 and200 mL∙ha–1) containing Azospirillum brasilense; ii) two N doses (0.0 and 30.0...

Quantification of natural populations of Gluconacetobacter diazotrophicus and Herbaspirillum spp. In sugar cane (Saccharum spp.) Using differente polyclonal antibodies 58
Silva-Froufe,Lúcia Gracinda da; Boddey,Robert Michael; Reis,Veronica Massena.
The species Gluconacetobacterdiazotrophicus, Herbaspirillum seropedicae and H. rubrisubalbicans are endophytic N2-fixing [diazotrophic] bacteria which colonise not only roots, but also the aerial tissue of sugar cane. However, the technique most commonly used to quantify the populations of these microbes in plants is by culturing serial dilutions of macerates of plant tissues in N free semi-solid media which are only semi-selective for the species/genera [the Most Probable Number (MPN) Technique] and each culture must be further subjected to several tests to identify the isolates at the species level. The use of species-specific polyclonal antibodies with the indirect ELISA (enzyme-linked immunosorbent assay) can be an alternative which is rapid and...

Solubilization of Araxá natural phosphate and decomposition of plant residues by bacterial isolates 64
Paredes Filho,Mário Viana; Silva,Adriano Bortolotti da; Florentino,Ligiane Aparecida.
ABSTRACT Brazil imports most of the phosphate fertilizers used in agriculture, however, it has national deposits in its territory that can supply part of the domestic market demand. In order of these to be used effectively, it is necessary to identify techniques to accelerate the release of phosphorus (P) from these rocks. The objective was to evaluate the influence of inoculation with phosphate solubilizing bacterial isolates (PSB) and decomposition of different plant residues in the solubilization of Araxá natural phosphate (ANP). Two experiments were carried out in a completely randomized design in a 5 x 4 factorial scheme, with four replicates. The first experiment evaluated the solubilization of ANP by PSB in culture medium with four different sources...

Vargas,Rafael S.; Bataiolli,Renata; da Costa,Pedro B.; Lisboa,Bruno; Passaglia,Luciane Maria P.; Beneduzi,Anelise; Vargas,Luciano K..
The aim of this study was to evaluate the impact of different grazing pressures on the activity and diversity of soil bacteria. We performed a long-term experiment in Eldorado do Sul, southern Brazil, that assessed three levels of grazing pressure: high pressure (HP), with 4% herbage allowance (HA), moderate pressure (MP), with 12% HA, and low pressure (LP), with 16% HA. Two reference areas were also assessed, one of never-grazed native vegetation (NG) and another of regenerated vegetation after two years of grazing (RG). Soil samples were evaluated for microbial biomass and enzymatic (β-glucosidase, arylsulfatase and urease) activities. The structure of the bacterial community and the population of diazotrophic bacteria were evaluated by RFLP of the 16S...

Growth promotion of pineapple 'vitória' by humic acids and burkholderia spp. during acclimatization 90
Baldotto,Lílian Estrela Borges; Baldotto,Marihus Altoé; Canellas,Luciano Pasqualoto; Bressan-Smith,Ricardo; Olivares,Fábio Lopes.
In vitro propagation of pineapple produces uniform and disease-free plantlets, but requires a long period of acclimatization before transplanting to the field. Quicker adaptation to the ex vitro environment and growth acceleration of pineapple plantlets are prerequisites for the production of a greater amount of vigorous, well-rooted planting material. The combination of humic acids and endophytic bacteria could be a useful technological approach to reduce the critical period of acclimatization. The aim of this study was to evaluate the initial performance of tissue-cultured pineapple variety Vitória in response to application of humic acids isolated from vermicompost and plant growth-promoting bacteria (Burkholderia spp.) during greenhouse...

THE RESURRECTION PLANT TRIPOGON SPICATUS (POACEAE) HARBORS A DIVERSITY OF PLANT GROWTH PROMOTING BACTERIA IN NORTHEASTERN BRAZILIAN CAATINGA 90
Fernandes-Júnior,Paulo Ivan; Aidar,Saulo de Tarso; Morgante,Carolina Vianna; Gava,Carlos Alberto Tuão; Zilli,Jerri Édson; Souza,Layane Silva Barbosa de; Marinho,Rita de Cássia Nunes; Nóbrega,Rafaela Simão Abrahão; Brasil,Marivaine da Silva; Seido,Sirando Lima; Martins,Lindete Míria Vieira.
Plant species that naturally occur in the Brazilian Caatinga(xeric shrubland) adapt in several ways to these harsh conditions, and that can be exploited to increase crop production. Among the strategic adaptations to confront low water availability, desiccation tolerance stands out. Up to now, the association of those species with beneficial soil microorganisms is not well understood. The aim of this study was to characterize Tripogon spicatusdiazotrophic bacterial isolates from the Caatingabiome and evaluate their ability to promote plant growth in rice. Sixteen bacterial isolates were studied in regard to their taxonomic position by partial sequencing of the 16S rRNA gene, putative diazotrophic capacity, in vitro indole-acetic acid (IAA) production and...

Tolerance to and Accumulation of Cadmium, Copper, and Zinc by Cupriavidus necator 90
Vicentin,Rayssa Pereira; Santos,Jessé Valentim dos; Labory,Cláudia Regina Gontijo; Costa,Amanda Monique da; Moreira,Fatima Maria de Souza; Alves,Eduardo.
ABSTRACT Preliminary results of in vitro experiments with multicontaminated soils and solid media indicated that nodulating diazotrophic bacteria of the genus Cupriavidus are promising for the remediation of contaminated environments due to their symbiosis with legumes and metal tolerance. Thus, strains of Cupriavidus spp. (LMG 19424T, UFLA 01-659, UFLA 01-663, and UFLA 02-71) were tested for their ability to tolerate and bioaccumulate cadmium (Cd), copper (Cu), and zinc (Zn) in Luria-Bertani broth. Changes in the growth pattern of Cupriavidus strains in the presence or absence of heavy metals were analyzed by scanning electron microscopy and metal allocation by transmission electron microscopy, to clarify the mechanisms of bioremediation.
