Polycylic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in the marine environment Their toxicity is mainly linked to the ability of marine species to biotransform them into reactive metabolites PAHs are thus often detected at trace levels in animal tissues For biomonitoring purposes this findings have two main consequences (i) the determination of the PAH tissue concentration is not suitable for the evaluation of individual exposure to PAHs (ii) It can explain sometimes the lack of correlations obtained with relevant markers of toxicity such as genotoxicity biomarkers The aim of the present study was to better investigate the link between PAH exposure and genotoxicity in marine flatfish During a laboratory experiment juvenile soles were exposed... |

Due to hydrophobic and persistent properties, polycyclic aromatic hydrocarbons (PAHs) have a high capacity to accumulate in sediment. Sediment quality criteria, for the assessment of habitat quality and risk for aquatic life, include understanding the fate and effects of PAHs. In the context of European regulation (REACH and Water Framework Directive), the first objective was to assess the influence of sediment composition on the toxicity of two model PAHs, benzo[a]pyrene and fluoranthene using 10-day zebrafish embryo-larval assay. This procedure was undertaken with an artificial sediment in order to limit natural sediment variability. A suitable sediment composition might be then validated for zebrafish and proposed in a new OECD guideline for chemicals... |

As wide-spread pollutants in the marine environment, microplastics (MPs) have raised public concern about potential toxic effects in aquatic organisms, and, among others, MPs were suspected to act as a vector for organic pollutants to biota. The purpose of the present study was to investigate effects by three model pollutants, oxybenzone (BP3), benzo[a]pyrene (BaP), and perfluorooctane sulfonate (PFOS) adsorbed to polyethylene MPs on the basis of a standard assay, the acute fish embryo toxicity test (FET; OECD TG 236) with zebrafish (Danio rerio) supplemented by additional endpoints such as induction of ethoxyresorufin-O-deethylase (EROD) activity, modification of cyp1a gene transcription and changes in larval swimming behavior. FET assays were performed... |

Several bioindicators were used to evaluate the biological and genotoxic effects of marine pollutants near large coastal cities in the northwestern part of the Mediterranean Sea. Three target species of teleosts were selected: red mullet Mullus barbatus and 2 types of comber (Serranus hepatus and S. cabrilla). Induction of ethoxyresorufin-O-deethylase (EROD) activity specific for polycyclic aromatic hydrocarbons (PAH) and polychlorinated biphenyls (PCB) was measured in the livers of the fish, and inhibition of acetylcholinesterase (AChE) by organophosphorus insecticides and carbamates was measured in their muscle tissues. Maximal EROD activities (16.8 +/- 2.7 to 19.4 +/- 4.2 pmol min(-1) mg protein(-1)) recorded in red muller near Barcelona (Spain),... |

The benzo[a]pyrene is a polycyclic aromatic hydrocarbon known to be genotoxic, mutagenic and carcinogenic in higher vertebrates. The aim of this study was to evaluate in vitro the enzymatic and genotoxic effects of BaP in a benthic fish species, Solea solea. Sole hepatocytes were exposed to BaP in order to measure the modulation of ethoxyresorufin-o-deethylase (EROD) activity and the DNA strand breaks induced by BaP metabolism. Exposures were performed in both culture flasks and microplate wells in order to check for the possible miniaturization of the exposure system. Moreover, sole liver microsomes were exposed to BaP in the presence of standard DNA in order to assess the potential formation of DNA adducts in sole. The results demonstrated the ability of... |
