We carried out a comparative morphological study to evaluate the taxonomic value of seed and seedling traits of Entada polystachya and E. simplicata (Leguminosae, Mimosoideae, Mimoseae). Seeds of both species were collected from wild populations in Roraima State, Brazil. Our results show consistent quantitative and qualitative differences between these species regarding their seeds and seedlings. Testa fracture lines, funiculus impression, lens characters, radicle shape and length, and cotyledon lobes length, seedling morphology group, number of pinnae at first node, and hypocotyl length permit a straightforward distinction of E. polystachya from E. simplicata, supporting a recent treatment of the latter taxon as a distinct species, endemic to Roraima... |

Although this study aims to develop an improved method for the preservation of reserve lipids in plant tissues for different uses in plant anatomy, it mostly aims to develop an improved method for the identification of lipid reserves where synthesis or storage occurs. The proposed procedures entail only the utilization of (2-hydroxyethyl)-methacrylate (HEMA) as a dehydration agent. One of the procedures is based on the gradual exchange of aqueous HEMA solutions with increasing concentrations. In another procedure, dehydration and infiltration are induced by the presence of silica gel around a modified microcentrifuge tube containing the aqueous HEMA solution with the plant tissues, thus allowing efficient lipid preservation. Both procedures resulted in... |

The evaluation of the post-germination phases of plants allows the recognition of transitional structures that support the relationships among taxa and their establishment. Together with fruit and seeds, seedlings and saplings provide useful characters for identifying many species. This study aimed to describe and characterize the morphology of the fruits, seeds, seedlings and saplings of Macrolobium acaciifolium, M. bifolium and M. pendulum, as well as to evaluate the ecological, taxonomic and phylogenetic value of the structures identified. In this work, the seeds were sown in plastic trays with sterilized sand and sawdust, without pre-germination treatment. The fruits of the three species are woody legumes, dehiscent fruit or indehiscent fruit. The... |

The common bean (Phaseolus vulgaris L.) is an important source of copper in human nutrition. The objective of this research was to investigate the possible maternal effects on copper concentrations in the seeds of the common bean, to estimate the heritability and the selection gains in early hybrid generations for the characteristic of copper concentration and to evaluate the potential for genetic improvement. The controlled crossings were performed among IAPAR 44 x IAPAR 31 and Diamante Negro x TPS Bonito cultivars. The reciprocal F1, and F2 generations, as well as the backcross populations, were produced by each hybrid combination. The range of variation in the copper concentration among the tested progenies was from 1.07 to 7.75 mg kg-1 dry matter, and... |

In order to produce transgenic goats with hG-CSF, a total of 24 adult Saanen and 48 adult undefined breed goats were used as donors and recipients, respectively. Donors were estrus-synchronized with vaginal sponges and superovulated by a treatment with 200 mg FSH given twice daily in decreasing doses over 3 days starting 48 h before sponge removal. Ovulation was induced by injecting 100µg GnRH 36 h after sponge removal. The recipients also received an estrus synchronization treatment. Donors were mated with fertile Saanen bucks and, approximately 72 h after sponge removal, zygotes were recovered surgically by flushing oviducts. The recovered zygotes were briefly centrifuged to a reliable visualization of the pronuclei. The DNA construct containing hG-CSF... |

In cattle, embryo development is characterized by the appearance of two distinct cell layers, the trophectoderm and the inner cell mass. The latter will undergo differentiation to form the embryonic disc consisting of the epiblast and hypoblast. The aim of this study was to ultrastructurally characterize the bovine embryo from different in vitro production techniques, with emphasis on trophectoderm and inner cell mass cells. Bovine embryos on day 7 (conception = D1) of pregnancy, derived via in vitro production techniques, were fixed for light and transmission electron microscopy processing. Results suggested that embryos produced by nuclear transfer of somatic cells and parthenogenesis showed significant changes in macroscopic and microscopic structure.... |

The effect of cryopreservation in IFN-tau, from bovine embryos produced in vitro was evaluated. Two treated groups (G1= fresh bovine embryos, n=59 and G2= freezed embryos, n=84) were used to study the effect of cryopreservation on IFN-tau secretion. After reaching the blastocyst phase, the embryos were kept on individual culture for additional period of 7 days. On days 3 and 7 after the beginning of embryos cultivation, samples of the media culture were taken for IFN-tau secretion titration. Oocysts taken from follicles ranging from 3 to 5mm in diameter were obtained from ovaries of females at slaughterhouse. The embryos were frozen, after being dehydrated with ethylene glycol (1.8m), conditioned on 0.5ml palletes and frozen. Frozen embryos secreted lower... |

Temperature and rainfall were analyzed daily during six years to evaluate their influence on in vitro production of bovine embryos. Weekly replications (n=480) were performed on 14,778 ovaries collected at slaughterhouses. Cumulus oocyte complexes (n=19,180) were fertilized with a pool of Bos taurus taurus semen in one incubator with 5% CO2. Presumable zygotes were cultured in gasified plastic bags with 5% CO2, 5% O2, and 90% N2. In the first year, cleavage and embryo yield were 60.3% and 15.6%, respectively, being lower (P<0.05) than in the following years. Average cleavage rates were always lower in winter (P<0.0001), thus producing less embryos. Winter climatic conditions had a negative influence on in vitro production, when cleavage and embryo... |

ABSTRACT Piau porcine blastocysts were submitted to MALDI-TOF to identify the main phospholipids (PL). After that, in vivo blastocysts (D6) were vitrified (n=52), non-vitrified were used as control (n=42). After warming, blastocysts were in vitro cultured to assess re-expansion and hatching at 24 and 48 hours. Finally, at 48 hours, hatched blastocysts were submitted to RT-qPCR searching for BCL2A1, BAK, BAX and CASP3 genes. For MALDI-TOF, the ion intensity was expressed in arbitrary units. Blastocyst development was compared by Qui-square (P< 0.05). Among the most representative PL was the phosphatidylcholine [PC (32:0) + H]+; [PC (34:1) + H]+ and [PC (36:4) + H]+. Beyond the PL, MALDI revealed some triglycerides (TG), including PPL (50:2) + Na+, PPO... |

Cryopreservation is a long-term storage technique to preserve the biological material without deterioration for extended period of time at least several thousands of years. The ability to preserve and store both maternal and paternal gametes provides a reliable source of fish genetic material for scientific and aquaculture purposes as well as for conservation of biodiversity. Successful cryopreservation of fish sperm have been achieved for more than 200 fish species and many fish species have been adequated for the purpose of cryobanking. Cryopreservation of fish embryo is not viable, mainly because of the same limitations as in fish oocytes, i.e., high chilling sensitivity and low membrane permeability. However, cryopreservation of isolated embryonic... |

Eggs of the Southern Brazilian flounder Paralichthys orbignyanus were obtained through spawning induction and reared in laboratory. The eggs released were free-floating or pelagic. Other important characteristics were: spherical shape, smooth chorion, narrow perivitelline space, and a single oil globule. Egg and oil globule diameter were about 0.792 ± 0.008 mm and 0.114 ± 0.003 mm, respectively. The blastodisc was observed approximately 1 h after fertilization and followed by blastodisc cleavage. Blastula stage started after about 5 h and gastrula stage after 9 h. Approximately 20 h after fertilization, blastopore closure was observed. Neurula or early embryo occurred after 24 h. Cardiac beats and movements of the free embryonic tail were recorded after 40... |

The function of the visceral yolk sac (VYS) is critical for embryo organogenesis until final fetal development in rats, and can be affected by conditions such as diabetes. In view of the importance of diabetes during pregnancy for maternal and neonatal health, the objective of this study was to assess fetal weight, VYS cell markers, and viability in female Wistar rats (200-250 g) with induced diabetes (alloxan, 37 mg/kg) on the 8th gestational day (gd 8). At gd 15, rats from control (n=5) and diabetic (n=5) groups were anesthetized and laparotomized to remove the uterine horns for weighing of fetuses and collecting the VYS. Flow cytometry was used for characterizing VYS cells, and for determining mitochondrial activity, cell proliferation, DNA ploidy, cell... |
