Co-transformation of a tropical maize endophytic isolate of Fusarium verticillioides (synonym F. moniliforme) with gusA and nia genes Genet. Mol. Biol.
Pamphile,João A.; Rocha,Carmen Lúcia M.S.C. da; Azevedo,João L..
A tropical endophytic isolate of the fungus Fusarium verticillioides (synonym Fusarium moniliforme) obtained from Zea mays was co-transformed with plasmid pNH24 containing the Fusarium oxysporum nitrate reductase nia gene and plasmid pNOM 102 carrying the Escherichia coli b-glucuronidase gusA gene. Transformation frequency for the nia marker was 75 transformants mg-1 vector DNA and introduction of the gusA gene by co-transformation was 57.2% as indicated by the presence of the GUS+ phenotype on plates. Southern analyses confirmed the integration of both plasmids into the genome of ten GUS+ transformants. All co-transformants showed mitotic stability in respect of the GUS+ phenotype. To assess the potential of transformed endophytic fungi as vectors for...

Comparative genomics of an endophytic Pseudomonas putida isolated from mango orchard Genet. Mol. Biol.
Asif,Huma; Studholme,David J.; Khan,Asifullah; Aurongzeb,M.; Khan,Ishtiaq A.; Azim,M. Kamran.
Abstract We analyzed the genome sequence of an endophytic bacterial strain Pseudomonas putida TJI51 isolated from mango bark tissues. Next generation DNA sequencing and short read de novo assembly generated the 5,805,096 bp draft genome of P. putida TJI51. Out of 6,036 protein coding genes in P. putida TJI51 sequences, 4,367 (72%) were annotated with functional specifications, while the remaining encoded hypothetical proteins. Comparative genome sequence analysis revealed that the P. putida TJI51genome contains several regions, not identified in so far sequenced P. putida genomes. Some of these regions were predicted to encode enzymes, including acetylornithine deacetylase, betaine aldehyde dehydrogenase, aldehyde dehydrogenase, benzoylformate...

Pathogenicity evaluation of Cytospora eucalypticola isolated from Eucalyptus spp: cankers in Uruguay Trop. Plant Pathol.
Alonso,Raquel; Lupo,Sandra; Bettucci,Lina.
Cytospora eucalypticola has been frequently associated with twig and stem cankers and as endophyte of Eucalyptus globulus and E. grandis in Uruguay. Mycelium discs of two C. eucalypticola isolates obtained from actively growing colonies were inoculated, both superficially and on experimentally wounded stems of E. globulus and E. grandis. No inoculated and control plants have shown any discoloration, gumosis or necrosis nor did they display lesions ten months after inoculation. Callus tissue was formed, partially or wholly occluding the wounds. The ability to penetrate healthy tissues and the inability to produce lesions evidenced that the presence of C. eucalypticola in twig and stem cankers could result from saprotrophic expansion of the endophytic...

Diversity of foliar endophytic fungi from the medicinal plant Sapindus saponaria L . and their localization by scanning electron microscopy Biol. Res.
García,Adriana; Rhoden,Sandro A; Rubin Filho,Celso J; Nakamura,Celso V; Pamphile,João A.
Endophytic fungi inhabit vegetable tissues or organs, without causing them any harm. Endophytes can co-evolve with plant hosts and possess species-specific interactions. They can protect the plant from insect attacks and diseases, and are also able to produce substances of biotechnological interest. In folk medicine, the bark, roots and fruits of Sapindus saponaria is used to produce substances with anxiolytic, astringent, diuretic and expectorant properties, as well as tonics, blood depuratives and cough medicine. This study evaluated the diversity of endophytic fungi present in the leaves of S. saponaria L. and observed the colonization of host plants by endophytes, using light and scanning electron microscopy. We verified that these fungi are found in...

Antagonism and modes of action of Chaetomium globosum species group, potential biocontrol agent of barley foliar diseases Boletín de la Sociedad
Moya,Paulina; Pedemonte,Debora; Amengual,Susana; Franco,Mario E. E; Sisterna,Marina N.
"Net blotch" (Drechslera teres) and "Bipolaris spot blotch" (Bipolaris sorokiniana) are foliar diseases of barley. Biological control is currently considered as an efficient alternative to chemical management of these plant diseases. The aim of the present study was to identify 2 isolates of Chaetomium (C2 and C5), endophytics on barley seedlings and to study the in vitro interactions with D. teres and B. sorokiniana, isolated from seeds of the same host. Cultural and morphological characterization of all microorganisms was done. In addition, molecular characterization of Chaetomium spp. was conducted and dual culture tests were carried out to find, by microscopic observations, the effects of the antagonist on the morphology of the pathogens. The results...

Endophytic fungi found in association with Bacopa monnieri as potential producers of industrial enzymes and antimicrobial bioactive compounds BABT
Katoch,Meenu; Salgotra,Aseem; Singh,Gurpreet.
This study aimed to screen the endophytic fungal species of ethano-medicinal plant Bacopa monnieri (L.) Pennell for their ability to produce antimicrobial substances against Bacillus subtilis, Pseudomonas aeroginosa, Salmonella typhimurium, Escherichia coli, Klebsiella pneumonia, Staphylococcus aureus, and Candida albicans. Endophytes were also screened for their ability to produce amylase, cellulase, protease and lipase to evaluate their ecological role within the host plant. Twenty-six endophytes were isolated and seventeen were identified. All the isolated endophytes exhibited amylolytic activity. Lipolytic, cellulolytic, proteolytic activity was shown by 98, 28 and 31% isolates, respectively. Similarly, all the endophytes (100%) exhibited significant...

A procedure to evaluate the efficiency of surface sterilization methods in culture-independent fungal endophyte studies BJM
Burgdorf,R.J.; Laing,M.D.; Morris,C.D.; Jamal-Ally,S.F..
Extraneous DNA interferes with PCR studies of endophytic fungi. A procedure was developed with which to evaluate the removal of extraneous DNA. Wheat (Triticum aestivum) leaves were sprayed with Saccharomyces cerevisiae and then subjected to physical and chemical surface treatments. The fungal ITS1 products were amplified from whole tissue DNA extractions. ANOVA was performed on the DNA bands representing S. cerevisiae on the agarose gel. Band profile comparisons using permutational multivariate ANOVA (PERMANOVA) and non-metric multidimensional scaling (NMDS) were performed on DGGE gel data, and band numbers were compared between treatments. Leaf surfaces were viewed under variable pressure scanning electron microscopy (VPSEM). Yeast band analysis of the...

Methylobacterium-plant interaction genes regulated by plant exudate and quorum sensing molecules BJM
Dourado,Manuella Nóbrega; Bogas,Andrea Cristina; Pomini,Armando M.; Andreote,Fernando Dini; Quecine,Maria Carolina; Marsaioli,Anita J.; Araújo,Welington Luiz.
Bacteria from the genus Methylobacterium interact symbiotically (endophytically and epiphytically) with different plant species. These interactions can promote plant growth or induce systemic resistance, increasing plant fitness. The plant colonization is guided by molecular communication between bacteria-bacteria and bacteria-plants, where the bacteria recognize specific exuded compounds by other bacteria (e.g. homoserine molecules) and/or by the plant roots (e.g. flavonoids, ethanol and methanol), respectively. In this context, the aim of this study was to evaluate the effect of quorum sensing molecules (N-acyl-homoserine lactones) and plant exudates (including ethanol) in the expression of a series of bacterial genes involved in Methylobacterium-plant...
