Population density of Beauveria bassiana in soil under the action of fungicides and native microbial populations Agronomy
Soares, Flávia Barbosa; Monteiro, Antonio Carlos; Babosa, José Carlos; Mochi, Dinalva Alves.
This study investigated whether populations of naturally-occurring soil bacteria, fungi and actinomycetes influence the effect of fungicides on the survival and growth of Beauveria bassiana. The toxicity of methyl thiophanate, pyraclostrobin, mancozeb and copper oxychloride at the recommended doses was analyzed in culture medium and in soil inoculated with fungus at various time points after addition of fungicides. All fungicides completely inhibited the growth and sporulation of B. bassiana in the culture medium. The fungicides were less toxic in soil, emphasizing the action of the microbial populations, which interfered with the toxic effects of these products to the fungus. Actinomycetes had the greatest influence on the entomopathogen, inhibiting it or...

Polymorphism of the grasshopper Schistocerca pallens (Thunberg) (Orthoptera: Acrididae) and its natural pathogen Metarhizium flavoviride Gams & Rozsypal (Hyphomycetes), revealed by RAPD analysis Anais da SEB
Silveira,Eni B.; Al-Janabi,Salah M.; Magalhães,Bonifácio P.; Carvalho,Luiz J.C.B.; Tigano,Myrian S..
The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters. The program RAPDDIP applied to S. pallens revealed nucleotide diversity of 2.3 and 2.2% for the populations from Rio Grande do Norte (Northeast Brazil) and Federal District (Central Brazil), respectively. These values indicate the presence of high genetic variability within these populations. Conversely, the...

Pulsed field gel electrophoresis reveals chromosome length and number differences in Brazilian strains of Metarhizium Anisopliae BABT
Kava-Cordeiro,Vanessa; Queiroz,Marisa Vieira de; Pizzirani-Kleiner,Aline Aparecida; Azevedo,João Lúcio.
Electrophoretic karyotypes of eight wild-type strains of Metarhizium anisopliae var. anisopliae were obtained by pulsed-field gel electrophoresis. These strains were isolated from insects of six different Brazilian states. The chromosomal DNA molecules of three strains were separated into seven bands and of five strains into eight bands. Chromosome length polymorphisms were also observed. The size of the chromosomal DNA of all strains varied between 7.7 and 0.9 Mb using the Aspergillus nidulans chromosomes as size standards. The total genome size of these strains was estimated in at least 29.7 Mb. Some correlations between differences in karyotype and occurrence of parasexual cycle likewise the host specificity were discussed.

Horizontal transmission and effect of the temperature in pathogenicity of Beauveria bassiana against Diatraea saccharalis (Lepidoptera: Crambidae) BABT
Svedese,Virgínia Michelle; Lima,Elza Áurea de Luna Alves; Porto,Ana Lúcia Figueiredo.
The aim of this work was to evaluate the effect of temperature on the pathogenicity of Beauveria bassiana against Diatraea saccharalis and evaluate the horizontal transmission capacity among the sugarcane borer larvae. The strains were most pathogenic at 26ºC, followed by 32ºC than at 20ºC, with respective mortalities of 100, 50 and 30.3%. The lethal time was also affected by the temperature, with the shortest LT 50 observed at 26ºC (2.2 days) and the longest at 20ºC (16.2 days). The URM2915 strain appeared most effective in all the experiments as having potential for deployment in biological control programs for the sugarcane borer. Results in horizontal transmission of B. bassiana showed that this procedure could also be adopted as control strategy for...

Biocontrol of pigeon tick Argas reflexus (Acari: Argasidae) by entomopathogenic fungus Metarhizium Anisopliae (Ascomycota: Hypocreales) BJM
Tavassoli,Mosa; Pourseyed,Seyed Hassan; Ownagh,Abdulghaffar; Bernousi,Iraj; Mardani,Karim.
The pigeon tick Argas reflexus is a pathogen-transmitting soft tick that typically feeds on pigeons, but can also attack humans causing local and systemic reactions. Chemical control is made difficult due to environmental contamination and resistance development. As a result, there is much interest in increasing the role of other strategies like biological control. In this study, the efficacy of three strains (V245, 685 and 715C) of entomopathogenic fungus Metarhizium anisopliae for biological control of three life stages of pigeon tick A. reflexus including eggs, larvae, engorged and unfed adults was investigated under laboratory conditions. Five concentrations of different strains of M. anisopliae ranging from 10³ to 10(7) conidia/ml were used. All...

Pereira,Aldo; Casals,Pedro; Salazar,Ana María; Gerding,Marcos.
Obscure mealybug, Pseudococcus viburni (Signoret), is a cosmopolitan pest that causes damage by suction of vascular juices and the production of honeydew, as well as for being a quarantine insect. Within control options, entomopathogenic fungi are a good alternative, nevertheless, more research is needed. In this research, the Metarhizium anisopliae var. anisopliae (Metschnikoff) isolate Qu-M984 was evaluated on P. viburni under laboratory conditions. Virulence was evaluated by lethal dose 50 (LD50) and lethal time 50 (LT50), for each of the four life stages of the female. The doses tested were 10(5), 10(6), 10(7) and 10(8) conidia mL-1. Also fecundity, egg size, fertility, and longevity of adult females were evaluated at doses of 10(5) and 10(6) conidia...

Volcanic materials as carriers for the formulation of mycoinsecticides using the fungus Beauveria bassiana Ciencia e Investigación Agraria
Sy,Victoria E; Schalamuk,Santiago; Scorsetti,Ana C; Botto,Irma L.
Improvements in the formulation of biological insecticides are crucial to increase their stability and competitiveness in the market. The aim of this work was to evaluate the use of volcanic materials with different hydric retention capacities to increase the thermotolerance of B. bassiana conidia. Two pyroclastic rocks and a zeolite clinoptilolite were selected for this study. In addition, a commercial silica gel was used due to its reported capacity to increase conidial thermotolerance. Dry conidia were mixed with the materials to obtain a 2% w/w granular formulation, while conidial powder alone served as the control. Mixtures were stored at room temperature for 30 days and then exposed to 50 °C for two hours, and the viability was measured before and...

Genetic diversity of Hirsutella thompsonii isolates from Thailand based on AFLP analysis and partial beta-tubulin gene sequences Genet. Mol. Biol.
Tigano,Myrian S.; Adams,Byron; Maimala,Saowanit; Boucias,Drion.
Amplified fragment length polymorphism (AFLP) was used to investigate polymorphism among 43 Hirsutella thompsonii isolates (33 from Thailand) obtained from various mite species. The outgroups were an unidentified Hirsutella isolate along with Hirsutella nodulosa and Hirsutella kirchneri. Phylogenetic analyses of the AFLP data showed significant variation among isolates and the existence of three H. thompsonii clades. We also investigated the isolates using PCR with specific primers for the Hirsutella exotoxin gene Hirsutelin A (HtA) and 18 of these isolates were used for sequencing of the partial beta-tubulin gene. Phylogenetic analyses of beta-tubulin sequences showed two distinct H. thompsonii clades, one of which included AFLP clades I and II. For both...

Populational parameters of Spalangia endius Walker (Hymenoptera: Pteromalidae) on Pupae of Musca domestica L. (Diptera: Muscidae) treated with two strains of Beauveria bassiana (Bals.) Vuil. (Deuteromycetes) Neotropical Entomology
Lecuona,Roberto; Crespo,Diana; La Rossa,Francisco.
The parasitoid Spalangia endius Walker is an efficient controller of Dipteran pupae, such as Musca domestica L. The entomopathogenic fungus Beauveria bassiana (Bals.) Vuil. is a regulator of insect populations, including these synanthropic pests. The aim of this work was to explore the possibilities of utilizing both agents in a combined form for the biocontrol of the domestic fly. Recently formed M. domestica pupae were inoculated by immersion in conidia suspension (10(8) conidia/ml) with two strains of B. bassiana (Bb6 and Bb10). The inoculated pupae were offered to the female parasitoid. In one bioassay they were offered pupae inoculated a single day and in other, pupae inoculated the following day as well. In both bioassays non inoculated (control)...

Seasonal Variations of Metapleural Secretion in the Leaf-Cutting ant Atta sexdens piriventris Santschi (Myrmicinae: Attini), and Lack of Fungicide Effect on Beauveria bassiana (Bals.) Vuillemin Neotropical Entomology
DIEHL,ELENA; JUNQUEIRA,LUCIANE K..
Mechanisms involved in maintaining colonies of social insects free of pathogens are among the main aspects of interest in the evolutionary biology of this group. For ants, it has been suggested that secretion from the metapleural glands play an important role in colony asepsis. In this study, different worker subcastes of the fungus-growing ant Atta sexdens piriventris Santschi were compared in relation to presence of metapleural secretion during a year. At the same time, the topical activity of the secretion was tested on workers treated with the entomopathogenic fungus Beauveria bassiana (Bals.) Vuillemin. Based on the mean width of the cephalic capsule, six worker subcastes were defined: gardener, minor, media, large, major and soldier. A positive...
