 The objective of the present work was to investigate the antioxidant enzyme activities and the occurrence of lipid peroxidation during storage of J. curcas seeds obtained from fruits at different maturity stages. The seeds were subsequently stored for 18 months in Kraft paper bags in a laboratory environment. The pineapple is a fruit that has wide acceptance worldwide both in natural form, as industrialized. Your peel is a residue generated by food industries and from this residue can obtain a protein extract which is a good source of bromelain. This study aimed to obtain a protein extract from pineapple peel, evaluate its enzyme activity and its healing properties in skin lesions in rats. ABSTRACT The lipases have ability to catalyze diverse reactions and are important in different biotechnological applications. The aim of this work was to isolate and characterize microorganisms that produce lipases, from different food industry effluents localized in Pelotas, RS/Brazil. Bacteria were identified using Gram stain and biochemical tests (Vitek 2(r)). Fungi were identified according to macro and micromorphology characteristics. Polycyclic aromatic hydrocarbons (PAH) are carcinogenic compounds which contaminate water and soil, and the enzymes can be used for bioremediation of these environments. This study aimed to evaluate some environmental conditions that affect the production and activity of the catechol 1,2-dioxygenase (C12O) by Mycobacterium fortuitum in the cell free and immobilized extract in sodium alginate. Production of moderately halotolerant, SDS stable alkaline protease from Bacillus cereus MTCC 6840 isolated from lake Nainital, Uttaranchal state, India BJM

A moderately cold active, extracellular alkaline protease producing bacterium was isolated from a fresh water lake. The isolate was found to be a gram-positive, rod shaped organism later identified as Bacillus cereus MTCC 6840. The bacterium produced the maximum amount of enzyme when allowed to grow for 24 h at temperature 25º and pH 9.0. Among a variety of substrates used, fructose as a carbon source and a combination of yeast extract and peptone as nitrogen source, supported the maximum protease production by the organism (120 U/ml). Fe++ and Co++ stimulated the enzyme activity whereas Ca++, Cu++, K+, Mg++ and Mn++ inhibited it to different extents. Aspergillus niger F7 isolated from soil was found to be the potent producer of cellulase and xylanase. The residue of forest species Toona ciliata, Celtris australis, Cedrus deodara and Pinus roxburghii was selected as substrate for biodegradation study due to its easy availability and wide use in industry. It was subjected to alkali (sodium hydroxide) treatment for enhancing its degradation. Production of CGTase by a Bacillus alkalophilic CGII strain isolated from wastewater of a manioc flour industry BJM

GCTase production by a new strain of Bacillus alkalophilic CGII isolated from Brazilian wastewater of manioc flour industry was examined. The growth medium used was composed by 1.5% starch, 1.5% nitrogen and 1% Na2CO3. Higher activity was obtained with starch, maltodextrin and galactose. When glucose was added to the medium, no enzyme production was observed. High enzyme activity and growth were reached when aeration was increased (88.6 U/mL). The enzyme characterization showed an optimum pH and temperature 8.0 and 55ºC for starch hydrolyses, respectively. Mg+ and Ca++ showed small activation; however, Hg+ and Cu+ showed a strong enzyme inhibition.

The NADP+-dependent glutamate dehydrogenase of the yeast Kluyveromyces marxianus responds to nitrogen repression similarly to Saccharomyces cerevisiae BJM

NADP+-dependent glutamate dehydrogenase (NADP+-Gdh) is the first step in ammonia assimilation pathway in Saccharomyces cerevisiae and the knowledge of its regulation is the key for many biotechnological purposes such as single cell protein production. The regulation of NADP+-Gdh activity in Kluyveromyces marxianus cells was evaluated under different ammonia supply in batch cultivations. The results showed that K. marxianus NADP+-Gdh activity is induced over a narrow range of extracellular ammonia supply, being repressed by both high ammonia concentration and the glutamate formed. Abstract The commercial enzyme (E.C. = 3.2.1.23) from Kluyveromyces lactis (liquid) and Aspergillus oryzae(lyophilized) was investigated for its hydrolysis potential in lactose substrate, UHT milk, and skimmed milk at different concentrations (0.7; 1.0 and 1.5%), pH values (5.0; 6.0; 6.5 and 7.0), and temperature (30; 35; 40 and 55 ºC). High hydrolysis rates were observed for the enzyme from K. lactis at pH 7.0 and 40 ºC, and from A. oryzae at pH 5.0 and 55 ºC. The enzyme from K. lactis showed significantly higher hydrolysis rates when compared to A. oryzae. The effect of temperature and β-galactosidase concentration on the lactose hydrolysis in UHT milk was higher than in skimmed milk, for all temperatures tested. Abstract Immobilization process is used to facilitate the enzyme recovery and reusability. The aims of this study were to evaluate the effect of different amounts of chitosan on immobilization efficiency of Glucose Oxidase (GOX), pH effect on enzyme activity and changes of kinetic characteristics of enzyme immobilized on Magnetic Chitosan Nanoparticles (MCNP). GOX was immobilized on MCNP with cross bonds and Schiff base covalent connections. The sample with 0.3 g of chitosan had the highest immobilization efficiency (41.30%). ABSTRACT: The influences of byproduct amendments, containing silicon, calcium, magnesium and potassium, on acidic soil quality in Jiaodong Peninsula of China had been studied and compared with that of lime through monitoring physicochemical properties and enzyme activities of acidic soil over a 120-day period. Byproduct amendments (1125, 2250, 4500 and 9000 kg ha-1) and lime 2250 kg ha-1 was applied in the acidic soil. Results showed that both byproduct amendments and lime significantly increased the pH, EC and enzyme activities of soil. The by-product amendments inhibited microbial biomass carbon and soil respiration. En el presente trabajo se investigo la producción extracelular de celulasas en medio líquido con 12 cepas nativas mexicanas y una comercial de Pleurotus. También se determino el contenido de proteína en los extractos y el peso seco de las colonias. Y se determino el coeficiente de correlación entre las variables producción de celulasa y proteína. Abstract Two Amazonian closely related tetras – cardinal Paracheirodon axelrodi and green neon P. simulans – were artificially acclimatized to environmental chambers mimicking future climate change scenarios (mild, moderate and extreme), using a microcosm facility. P. simulans survived (100%) to all scenarios after 30 days exposure, while P. axelrodi presented decreasing survival percentages according to environmental severity. These differences may be the reflection of distinct natural acclimatization to microhabitats between the species, which differ in thermal conditions. Effect of growth hormone on fatty acid synthase gene expression in porcine adipose tissue cultures Genet. Mol. Biol.

We describe an efficient in vitro assay to test growth hormone effects on mRNA levels and fatty acid synthase (FAS, EC. 2.3.1.85) activity. Swine adipose tissue explants were long-term cultured in medium containing growth hormone and FAS mRNA levels and enzyme activity were measured. We quantified FAS transcripts by competitive reverse transcriptase PCR (RT-PCR) using total RNA from cultured adipose tissue explants and RT-PCR standard-curves were constructed using a cloned 307 bp segment of native FAS cDNA and a shorter fragment from which a 64 bp (competitor, 243 bp) internal sequence had been deleted.
