ABSTRACT Endophyte microorganisms have great biotechnological interest, with features applicable to different areas and are potentially useful in agriculture. The current study determines the biotechnological potential of endophytic fungi, isolated from leaves of Sapindus saponaria, to control phytopathogenic fungi and evaluate their enzyme production. Molecular taxonomy was performed by sequencing of the ITS1-5.8S-ITS2 ribosomal DNA region, identifying the genera Phomopsis, Sordariomycetes, Diaporthe, and Colletotrichum. In vitro antagonism against phytopathogens showed better results against Fusarium solani and provided inhibition indices between 41.8 % and 67.5 %. The endophytic strain SS81 (Diaporthe citri) presented the highest antagonism index... |

ABSTRACT We studied the fungal endophyte community of the leaves of Myracrodruon urundeuva in Brazil along with their potential to produce L-asparaginase. In total, 187 endophytes were isolated. The colonization rate of leaf fragments from caatinga (10.41 %) was lower than those from brejo de altitude (39.58 %). Sequences of ITS rDNA from all endophytes revealed relationships with Ascomycota (Botryosphaeriales, Chaetothyriales, Diaporthales, Eurotiales, Glomerellales, Hypocreales, and Pleosporales) and Basidiomycota (Polyporales). The most frequent endophytes were members of the genus Diaporthe. Talaromyces was an indicator genus for caatinga while Phyllosticta, Diaporthe, and Colletotrichum were for brejo de altitude. The composition of endophytic fungi... |

The seeds of a sesame crop contain approximately 50% oil content, one of the main reasons for their cultivation. However, information regarding methodologies for the assessment of quality seeds of this culture are scarce. Therefore, the objective of this study was to adapt the methods of accelerated ageing tests for sesame seeds and investigate changes in the behaviour of isoenzymes. The physiological characterization of the seeds and electrophoretic analysis of the isoenzymes superoxide dismutase, esterase, catalase, alcohol dehydrogenase, malate dehydrogenase and isocitrate lyase were also performed. For the accelerated ageing test, the seeds were subjected to the traditional method, with a saturated NaCl solution, at ageing times of 0, 24, 48, 72, and... |

ABSTRACT In this review we discuss about the immobilization of enzymes in Langmuir-Blodgett films in order to determine the catalytic properties of these biomacromolecules when adsorbed on solid supports. Usually, the conformation of enzymes depends on the environmental conditions imposed to them, including the chemical composition of the matrix, and the morphology and thickness of the film. In this review, we show an outline of manuscripts that report the immobilization of enzymes as LB films since the 1980’s, and also some examples of how the surface properties of the floating monolayer prepared previously to the transfer to the solid support are important to determine the efficiency of the resulting device. |

This study tried to monitor the alterations of muscle enzymes activity - creatinokinase (CK), lactate dehydrogenase (LDH), and aspartate aminotransferase (AST) - in a group of horses that participated of 70 and 100km distance rides in five competitions of an annual endurance championship under tropical climate. Pre ride levels (U/l) were 245.13±9.84 for CK, 496.61±14.76 for LDH, and 328.95±8.65 for AST. When compared to these levels, the results revealed a significant decrease in all enzymes activities in the first moment of the rides. Peak levels, significantly different, were reached, immediately after rides by CK (413.591±50.75); 24 hours post rides by LDH (628.61±33.30); and 48 hours after rides by AST (389.89±16.96). The effects of varying temperatures (12 - 44° C) on the specific activity of cytoplasmic malate dehydrogenase ((cMDH), mitochondrial malate dehydrogenase (mMDH) and lactate dehydrogenase (LDH) of some earthworms (Metaphire posthuma, Perionyx sansibaricus and Lampito mauritii) were studied. The effects of different temperatures on supernatant and mitochondrial protein contents were also investigated. The specific activities of cMDH, mMDH and LDH of the earthworms decreased gradually as a function of increasing temperature from 12 to 44°C. Higher metabolic energy was needed to maintain the activity at low temperatures. Hence, the earthworms showed increased enzyme specific activity at low temperatures. However, the protein content increased upto 28°C.... |
