Experimental ostreid herpesvirus 1 infection of the Pacific oyster Crassostrea gigas: Kinetics of virus DNA detection by q-PCR in seawater and in oyster samples ArchiMer

Herpes- and herpes-like viruses are known to infect a wide range of bivalve mollusc species throughout the world. Abnormal summer mortalities associated to the detection of ostreid herpesvirus 1 (OsHV-1) have been currently reported in France among larvae and spat of the Pacific cupped oyster Crassostrea gigas. In the present work, we have developed an experimental protocol of horizontal transmission based on the cohabitation between healthy and experimentally infected oysters. Through a cohabitation trial, the kinetics of OsHV-1 detection in different oyster organs and seawater samples were investigated and characterized for the first time using real time quantitative PCR.

Bonamiosis due to the parasite Bonamia ostreae has been associated with massive mortality outbreaks in European flat oyster stocks in Europe. As eradication and treatment are not possible, the control of the disease mainly relies on transfer restriction. Experimental herpes-like viral infections in marine bivalves: demonstration of interspecies transmission ArchiMer

Since 1972, herpes-like virus infections have been reported in several marine bivalve species around the world. Viral detection was often associated with high mortality rates in larvae and spat. To determine whether a single virus is able to infect different bivalve host species, we carried out experimental transmission assays. As a first step, 8 assays were performed to infect axenic Crassostrea gigas larvae with virus from infected C. gigas larvae using a previously described protocol. Experimental infection of adult Scapharca broughtonii with Ostreid herpesvirus SB strain ArchiMer

We investigated the susceptibility of ark shell, Scapharca broughtonii, adults to Ostreid herpesvirus SB strain (OsHV-1-SB) through experimental infection by intramuscular injection assays. Results showed the onset of mortality occurred at 3 days post injection, one day after the water turbidity became evident in rearing tanks. The mortality curves for the challenged group were similar to those observed at affected hatcheries. Histological lesions, herpesvirus-like particles and high OsHV-1-SB quantities were detected in challenged ark shells. This is the first study to successfully reproduce OsHV-1 disease in Arcoida species, and very few studies in adult bivalves (over 24 months old).

The oyster farming constitutes one of the major components of the global aquaculture. However, this sector is facing abnormal mortalities outbreaks that affect diploid and triploid oyster at their different life stages, in the hatcheries and in the field. During bivalve molluscs mortality events in France, bacteria initially classified into Harveyi group, were regularly isolated along with herpes virus, V. splendidus or V. aestuarianus. Siderophore production and probiotic effect of Vibrio sp. associated with turbot larvae, Scophthalmus maximus ArchiMer

The proportion of vibrio E, dominant in healthy turbot larvae, was artificially increased in the rotifer enrichment medium. The maid effect of this vibrio enrichment was to improve the resistance of larval turbot challenged with a pathogenic strain of Vibrio splendidus, vibrio P. The improvement of the survival rates was significant with 1.5 x 10(6) CFU of vibrio E added per mi of enrichment medium, and it was still maximum at the hi hest dose tested (5 x 10(7) CFU.ml(-1); 80 +/- 2% of survivors after 48 h of challenge, versus 50 +/- 2 % for the control group). The susceptibility of BALB/c, C57Bl/6, DBA/2, NMRI, NIH and CD1 mouse strains to experimental infection with a Venezuelan equine isolate of Trypanosoma evansi was examined through hemoglobin concentration, packed red cell volume, number of erythrocytes per blood μl, parasitemia, body weight, and mice survival time. Encephalitozoonosis is a common infectious disease caused by Encephalitozoon cuniculi in rabbits. The purpose of this study was to examine the levels of antibody in urines of experimentally E. cuniculi-infected rabbits. Humoral immune response and spore excretion in urines were also examined. Rabbits were orally infected with E. cuniculi, and sera and urines were collected. Levels of serum IgG, IgM, IgA and urinary IgG were examined. This study evaluated the pathogenic characteristics of bacteria isolated from Litopenaeus vannamei during an outbreak at the Laboratory of Marine Shrimp, UFSC, Santa Catarina State, Brazil. Their virulence potential in larvae and juvenile shrimp and the effects on the total haemocyte count, phenoloxidase activity and serum agglutinate titre were examined after experimental infection. An immunoistochemical (IHC) test was developed to detect bovine respiratory syncytial virus (BRSV) in cell cultures and tissues of experimentally infected mice and calves, using a commercial monoclonal antibody (Mab) against human respiratory syncytial virus (HRSV), as a less expensive alternative, instead of producing specific monoclonal antibodies to BRSV. Clinical samples from calves suffering respiratory disease were also submitted to this test. Five adult donkeys were experimentally infected with Brazilian strain of Trypanosoma evansi originally isolated from a naturally infected dog to study the hematological biochemical and histopathological alterations during the evolution of the disease. The course of the experimental infection was followed up to 145 days. Hematological analyses of the infected donkeys revealed a marked decline in hemoglobin, packed-cell volume, and erythrocyte count. Anemia was observed after successive peaks of parasitemia.
