Evaluation of a portable watermelon juice extracting machine CIGR Journal

A small-scale machine for extracting watermelon juice was developed with locally available materials and evaluated to determine its best extracting operating factors. The machine slices a whole watermelon and extracts its juice content. This machine operates on the principle of compressive and shearing force exerted through an auger.  Component parts includes feeding hopper, conveyor shaft, auger conveyor housed in a cylindrical chamber, juice sieve, juice collector, seed - pulp outlet, gear and main frame. Criteria for the Extraction of Fish Oil CIGR Journal

The effect of pre-treatment of mackery (Scomber scombrus) on oil recovery and quality was investigated. Fish oil was extracted using the screw expeller press after heat pre-treatment of the fish sample. The effect of cooking temperatures and cooking times was evaluated. It was observed that the oil yield has a positive relationship with cooking temperature. The oil yield increased with increasing cooking temperature at all the cooking times investigated and also increased with increasing cooking time from 5-15 minutes and then decreased for 20 minutes cooking time at all the cooking temperatures investigated. Effect of Expeller Press Parameters on Fish Oil Extraction CIGR Journal

An investigation was carried out on the effect of expeller press parameters such as choke clearances (1, 2 and 3 mm), screw clearances (1, 2 and 3 mm) and screw (worm) shaft speeds (50, 60 and 70 rpm) on fish oil extraction. The oil extraction rate and extraction efficiency had a negative relationship with choke clearances and screw clearances but had a positive relationship with screw (worm) shaft speeds. The oil extraction rate and extraction efficiency were found to decrease with increase in choke clearances and also decreased with increase in screw clearances. Mathematical modeling of mechanical horizontal screw oil extractor CIGR Journal

A theory using mechanical expression was proposed from the physics of oil expression in a screw press to characterize a mathematical model. The general oil expression equation was solved for three machine parameters (choke clearances, screw shaft clearances and screw shaft speeds) which simulate the machine parameters effects in the overall screw expeller process. The mathematical model was validated by the experimental data obtained from pressing fish oil with a screw press, using regression analysis. In the current study, response surface methodology (RSM) was used to assess the effects of extraction time and temperature on the content of bioactive compounds and antioxidant activity of purple basil leaf (Ocimum basilicum L.) extracts. The stability of anthocyanins in relation to temperature, light and copigmentation was also studied. The highest anthocyanin content was 67.40 mg/100 g extracted at 30 °C and 60 min. The degradation of anthocyanins with varying temperatures and in the presence of light followed a first-order kinetics and the activation energy was 44.95 kJ/mol. Purification and some properties of pectinesterase from potato (Solanum tuberosum L.) alpha cultivar BABT

Pectinesterase was extracted from potato alpha cultivar, purified and partially characterized The used protocol resulted in a 58.8-fold purification (51 850.2 units/mg protein) with 15.5% recovery of pectinesterase activity. The purified enzyme had a molecular weight of 27 kDa and its isoelectric point was around 4.5 with pH and temperature optima of 8.0 and 60°C, respectively. The purified enzyme had a single symmetric peak of specific activity after chromatographic steps. The homogeneity of the purified pectinesterase was confirmed by gel filtration and polyacrylamide electrophoresis gel.

Optimization of extraction parameters for recovery of alpha-amylase from the fermented bran of Bacillus circulans GRS313 BABT

The extraction of alpha-amylase in the solid state fermentation of wheat bran by Bacillus circulans GRS313 was optimized. Among various solvents tested, maximum extraction was achieved when 2.5% (v/v) glycerol was added. The optimum conditions for extraction were 2.5 hrs soaking time at 30° C under recirculation and agitated condition, which yielded 590 U/g bran of enzyme activity. Whereas under stationary soaking condition the activity of the enzyme was 439.58U/g bran. With repeated wash under the above optimum conditions showed that 600 U/g and 184.2 U/g of enzyme extracted in the 1st and 2nd washes respectively and only 36.84 U/g was recovered in the 3rd wash

Extraction of pectin from apple pomace BABT

As apple-processing units are now in expansion in Brazil, industrial by-products like pomace play an important role in pectin manufacture. The objective of this article was to determine a pratical follow-up to the extraction of pectin from apple pomace and to characterize it in a laboratory, on a small scale, aiming at establishing the optimum conditions for acid extraction. Extraction of red cabbage anthocyanins: optimization of the operation conditions of the column process BABT

The aim of this work was to extract anthocyanins from the red cabbage. Batch studies under several extraction conditions indicated that acetic acid in aqueous solution (10% V/V) was the best solvent, used in the proportion of 0.25 g of red cabbage mL-1. At this condition, column assays were carried out to evaluate the influence of the ionic force, pH, solvent flow rate, recirculated volume of red cabbage juice and the mass of red cabbage. Results showed that the pH, recirculation and mass of red cabbage had statistically significant effects, where the optimum operation conditions found for the process were pH 2.3, recirculation volume of the solvent 0.83 L and mass of red cabbage 50 g.

Extraction time for soybean isoflavone determination BABT

Studies were carried out on the optimization of the extraction procedures for isoflavones determination in soybean by HPLC. Isoflavones were extracted from 100 mg samples of full fat soybean flour "Kinako" (milled toasted grains). Recovery of average total isoflavones was higher when extraction was performed with agitation(217.2 mg/100g), than without agitation(191.0 mg/100g). Isoflavone extraction without agitation for 1,4, 20 and 24 hours, were equally efficient for total and individual compounds. These results suggested that an efficient isoflavone recovery could be achieved with extraction for one hour with agitation.

Neural modeling of bromelain extraction by reversed micelles BABT

A pulsed-cap microcolumn was used for bromelain extraction from pineapple juice by reversed micelles. The cationic micellar solution used BDBAC as the surfactant, isooctane as the solvent and hexanol as the co-solvent. In order to capture the dynamic behavior and the nonlinearities of the column, the operating conditions were modified in accordance with the central composite design for the experiment, using the ratio between the light phase flow rate and the total flow rate, and the time interval between pulses. Development and optimization of extractives from Astronium urundeuva (allemão) Engl. by factorial design BABT

The purpose of this study was to evaluate the influences from variables of preparation on dry residue and total tannins from macerates of Astronium urundeuva barks. Thus, a 3 ² factorial design was used to study the importance of plant proportion (10, 15 and 20 %, w/v) and the ethanol concentration (40, 60 and 80 %, v/v) on the selected response. Mathematical models were fitted according to experimental data. The validated equations were used to generate response surfaces. The analysis of the surfaces showed that the optimum conditions to obtain extractive preparations with high extractive efficiency for tannins were: 10 % (w/v) of plant with ethanol 40 % (v/v) as solvent.

Abstract The present study aims to evaluate the influence of extraction methods on the quality of bioactive compounds and antioxidant capacity in Pterodon emarginatusVogel (sucupira) oils. The oils were extracted from the sucupira seeds by Soxhlet and by the extraction system of Bligh & Dyer. The oils were analyzed as to fatty acid profile, tocopherols, phytosterols, carotenoids, total phenolics, and total antioxidant capacity. The extraction by Soxhlet showed better yield of total lipids and was more efficient to extract tocopherols, phytosterols, and carotenoids, besides presenting better antioxidant activity by DPPH•. Extraction model of low methoxyl pectin from apple pomace effects of acid concentration and time on the process and the product BABT

In Brazil, one of the top apple producing countries in the world, apple processing is an increasing activity, with pomace as the main by-product. To extract pectin from pomace, factors affecting process and product should be studied for optimization. A model to produce LMP directly from dried apple pomace was established observing the effects of HNO3 concentration and the time of reaction at 97ºC, analyzed from a statistical and practical point of view. Extraction and in vitro antioxidant activity of exopolysaccharide by Pleurotus eryngii SI-02 BJM

The extraction parameters for Pleurotus eryngii SI-02 exopolysaccharide (EPS) produced during submerged culture were optimized using response surface methodology (RSM). The optimum conditions for EPS extraction were predicted to be, precipitation time 20.24 h, ethanol concentration 89.62% and pH 8.17, and EPS production was estimated at 7.27 g/L. The actual yield of EPS under these conditions was 7.21 g/L. The in vitro antioxidant results of the EPS showed that the inhibition effects of EPS at a dosage of 400 mg/L on hydroxyl, superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals were 59.63 ± 3.72%, 38.69 ± 2.59%, and 66.36 ± 4.42%, respectively, which were 12.74 ± 1.03%, 8.01 ± 0.56%, and 12.19 ± 1.05% higher than that of butylated...

Assessment of four different detergents used to extract membrane proteins from Xylella fastidiosa by two-dimensional electrophoresis BJM

Four different detergents, ASB 14, SB 3-10, CHAPS and Triton X100, were utilized to determine the optimal detergent for the solubilization of membrane proteins from the phytopathogenic bacterium Xylella fastidiosa. These proteins were differentially solubilized in distinct buffers containing the detergent and subjected to bidimensional electrophoresis within the non-linear pH range of 3-10. The detergents ASB 14 and SB 3-10 were the most effective revealing 221 and 157 spots, respectively. CHAPS and Triton X100 were less effective and revealed only 72 and 43 spots, respectively. Abstract This study investigated the extraction of matrine from Vietnamese sophora root and its in vitro and in vivo inhibitory effects on oral squamous cell carcinoma (OSCC) cells. Matrine was extracted from Vietnamese sophora root using microwave-assisted extraction technology and purified using macroporous adsorption resin. Human OSCC Tca8113 cells were treated with matrine with concentration of 0, 0.5, 1, 2, 4 and 8 g/L. The proliferation and apoptosis of cells were detected.
