Post-thaw acrosomal viability and reaction in sperm obtained from equine epididymis tail Rev Salud Anim.

In order to evaluate the post-thaw effect on acrosomal viability and reaction of sperm collected from equine epididymis tail, 62 testes were collected and transported at 4°C. Epididymis was dissected to perform the retrograde flushing of the epididymis tail , using 10 mL of a diluent based on skim milk (INRA 82). The diluent obtained with the spermatic content was recovered in graduated collecting tubes and 1mL of each sample was taken to be evaluated. For evaluations, the samples having a minimum of 30% of progressive motility were selected. The evaluations were on: percentage of live sperm, normal morphology, sperm concentration, percentage of live sperm without acrosomal reaction (VsRA) and live with acrosomal reaction (VcRA), evaluating this indicator...

Efecto de la criopreservación de semen de conejo Nueva Zelanda (Oryctolagus cuniculus) sobre su viabilidad y estado acrosomal Rev Salud Anim.

El objetivo del estudio fue investigar el efecto de la criopreservación sobre la viabilidad y estado acrosomal de espermatozoides de conejo de la raza Nueva Zelanda (Oryctolagus cuniculus). Se utilizaron 46 eyaculados, obtenidos mediante vagina artificial, evaluándose las siguientes características en semen fresco y post-descongelado: % de viabilidad sin y con reacción acrosomal (VsRA y VcRA, respectivamente), y % de mortalidad sin y con reacción acrosomal (MsRA y McRA, respectivamente), mediante la tinción de Isotiocianato de fluoresceína conjugada con la lectina de Arachis Hypogaea y Ioduro de Propidio (FITC-PNA/IP). In the present research, the effects of cryopreservation on the viability and acrosomal status of sheep sperms were analyzed. Forty five ejaculates were obtained by the artificial vagina method for analysis, evaluating in fresh semen the following characteristics: volume (Vol), progressive motility (PM), viability percentage (Viab), normal morphology (NM), spermatic concentration (Concentr); as well as its viability and acrosomal status, The last two parameters were evaluated staining with fluorescein isothiocyanate conjugated with Arachis hypogaea lectin and propidium iodide (FITC-PNA/IP), in which there were different patterns of staining: live sperms without acrosomal reaction (VsRA), live with acrosomal reaction (VcRA), dead without acrosomal reaction...
