ABSTRACT The effect of pressure on the fluorescence of tryptophan in the presence of metal ions was studied by fluorescence spectrometry. It was found that at 60 MPa, the fluorescence intensity of M/Trp mixtures (M represented metal ions) increased compared to that at atmosphere pressure. The relative fluorescence efficiency of M/Trp mixtures increased with pressure. When the M/Trp ratio was above 10:1, the relative fluorescence efficiency in decreasing order was Cu2+/Trp mixtures, Ni2+/Trp mixtures and Mg2+ (K+)/Trp mixtures. The twospotted spider mite Tetranychus urticae Koch is a common pest on soybean plants. To clarify plant-arthropod interaction on mite-soybean system, leaf fluorescence, photosynthetic responses to variable carbon dioxide levels, and chlorophyll content were evaluated. Significant photosynthetic rate reduction was observed due to stomatal limitation. Stomatal closure was the major plant physiological response. As a consequence, there was reduction in photosynthetic rates. Surprisingly, plants did not show chlorophyll content reduction associated with photosynthetic impairment. Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Brazil is the largest world producer of papaya and the third largest exporter despite of only 1.5 to 2.0% of its production is exported. Such an underexplored exportation potential highlight the necessity for physiological studies on new cultivars and hybrids to verify their agronomic and commercial viability. Two Brazilian states, Bahia and Espirito Santo, are responsible for 80% of national production. Most studies of chromium have been realized in the sediments and in mangrove trees. However, little is known about its effect on growth and metabolism in this vegetation. L. racemosa plants were grown in solution with 20% of ionic strength of Hoagland and Arnon containing 100 mM NaCl and supplemented with 0.00, 0.05 and 0.50 mg L-1 of Cr+3. The growth, the pigments and gas exchange measurements showed no changes in response to doses of chromium. After 30 days the initiation of treatments, plants of L. racemosa had average 0.559 g of total dry weight, 1.34 mg L-1 of total chlorophyll and 7.9 µmol m-2 s-1 of A (CO2 photosynthetic assimilation). The effects of drought and rehydration on Canavalia ensiformis (L.) D.C. (jack bean) plants were evaluated using the following gas exchange parameters: net carbon assimilation rate (A), stomatal conductance (g s), Ci/Ca ratio and transpiration rate (E); chlorophyll a fluorescence: Fv/Fm and Fv/F0 ratio. Spathiphyllum spp. es conocida comúnmente como cuna de moisés, se utiliza como planta ornamental, presenta una hoja modificada (espata), de color blanco, y conforme madura enverdece. Este fenómeno típico de algunas Aráceas es poco conocido. En el presente trabajo se analizaron algunos parámetros que intervienen en el enverdecimiento. We investigated the influence of freshwater discharges on biogeochemistry and benthic communities of La Saline fringing reef, the largest coral reef system of La Réunion Island (France, Indian Ocean). Sampling was conducted in February–March 2012 along a land-ocean continuum, including rivers, groundwater, reef waters, and oceanic waters. Our results indicated the discharges of basalt groundwater (enriched in nitrates) in the southern part of the reef, and the discharges of sand groundwater and river waters (enriched in organic matter) in sites located northward. Upwelling of cold. saline, and nutrient rich water was observed in late September 1999 along an east-west transect in the SW Kattegat. The Kattegat forms part of the transitional zone between the high saline North Sea and the low saline Baltic Sea. The upwelling occurred after an extended period of northward flow and eastern winds in the Kattegat, that changed into a southward flow as wind ceased. The use of tissue homogenate has greatly aided the study of the functioning of mitochondria. However, the amount of ATP produced per oxygen molecule consumed, that is, the effective P/O ratio, has never been measured directly in tissue homogenate. Here we combine and refine existing methods previously used in permeabilized cells and isolated mitochondria to simultaneously measure mitochondrial ATP production (JATP) and oxygen consumption (JO(2)) in tissue homogenate. A major improvement over existing methods is in the control of ATPases that otherwise interfere with the ATP assay: our modified technique facilitates simultaneous measurement of the rates of uncorrected ATP synthesis and of ATP hydrolysis, thus minimizing the amount of tissue and time needed.... |
