Variation over time in the prevalences of three viruses of the grapevine leafroll complex in a commercial vineyard in south-central Chile Ciencia e Investigación Agraria

A vineyard in south-central Chile was surveyed during five growing seasons to analyze the increase over time in the number of plants infected with Grapevine leafroll-associated virus-1, -2 and -3. At the end of the study, GLRaV-3 was found to be the most prevalent virus, with 46% of plants infected by this pathogen. GLRaV-2 infected 13% of plants, and GLRaV-1 infected 12%. Considering all three viruses together, the prevalence of infection increased exponentially from 14% of the plants at the beginning of the study period to 68% at the end. The patterns of the spread of GLRaV-2 and GLRaV-3 were mostly along the rows, whereas the pattern of spread for GLRaV-1 could not be determined.

Detecção e identificação molecular de vírus associados a videiras sintomáticas e assintomáticas Ciência Rural

A propagação vegetativa da videira favorece infecções virais múltiplas, com expressão diferencial de sintomas em função da combinação da cultivar ou espécie da hospedeira com a espécie viral. Os objetivos deste trabalho foram detectar e identificar as espécies virais presentes em duas espécies/cultivares de videira: uma sintomática e outra assintomática. Production of polyclonal antisera using recombinant coat proteins of Grapevine leafroll-associated virus 2 and Grapevine virus B PAB

The objective of this work was to produce and characterize specific antisera against Brazilian isolates of Grapevine leafroll-associated virus 2 (GLRaV-2) and Grapevine virus B (GVB), developed from expressed coat proteins (CPs) in Escherichia coli, and to test their possible use for the detection of these two viruses in diseased grapevines. The coat protein (CP) genes were RT-PCR-amplified, cloned and sequenced. The CP genes were subsequently subcloned, and the recombinant plasmids were used to transform E. coli cells and express the coat proteins. The recombinant coat proteins were purified, and their identities were confirmed by SDS-PAGE and Western blot and used for rabbit immunizations.
