The pioneering use of ISSR (Inter Simple Sequence Repeat) in Neotropical anurans: preliminary assessment of genetic diversity in populations of Physalaemus cuvieri (Amphibia, Leiuperidae) 48
Moresco,Rafaela M; Maniglia,Thiago C; De Oliveira,Classius; Margarido,Vladimir P.
The greatest diversity of anurans in the world is in Brazil and one of the major challenges is to reconcile the accelerated economic development with strategies that aim to maintain this diversity in forest fragments, often representing ESUs of some biomes. This study aimed to obtain data that will support conservation projects through the pioneering use of ISSR analysis in Neotropical anurans, estimating the intra- and interpopulation genetic diversity of four populations of P. cuvieri (Paraná and São Paulo regions). Of the 65 loci scored 58 were polymorphic, with 0.797 intrapopulation variation and 0.203 interpopulation variation. The index of interpopulation genetic differentiation (Fst) proved to be high among the population of Marmeleiro-PR and the...

Genetic structure of natural populations of Cryptocarya moschata Nees (Lauraceae) from southeastern Brazilian Atlantic rain forest 49
Moraes,Pedro Luís Rodrigues de; Derbyshire,Maria Teresa Vitral de Carvalho.
This study was accomplished on a more comprehensive basis to evaluate previous questions that were raised from a preliminary article about the genetic structure of Cryptocarya moschata populations. Thus, through the analysis of 40 polymorphic allozyme loci, allele frequencies were estimated from 335 individuals of 11 natural populations of C. moschata from six hydrographic basins of São Paulo state and Serra da Estrela, Rio de Janeiro, Brazil. Estimates of Wright's F statistics were done through the analysis of variance, presenting average values of <img border=0 width=32 height=32 id="_x0000_i1026" src="../../../../../../img/revistas/bn/v4n1/img/a04car(f).jpg" align=absmiddle > or = 0.352, <img border=0 width=32 height=32 id="_x0000_i1027"...

Genetic diversity in oil palm (Elaeis guineensis Jacq) using RAM (Random Amplified Microsatellites) 53
Cardona,Christian Camilo Castañeda; Coronado,Yacenia Morillo; Conronado,Ana Cruz Morillo; Ochoa,Iván.
ABSTRACT The oil palm (Elaeis guineensis Jacq) plays an important role in the economy of some tropical countries; Colombia ranks first for production in Latin America and fifth in the world. The aim of this research was to characterize the genetic variability of 51 oil palm genotypes from the Congo with seven randomly amplified microsatellite markers RAM. As a result, 241 alleles were generated andthe number of polymorphic loci ranged from 46 to 14 for the ACA and CGA primers, respectively. High genetic diversity was found, with a total heterozygosity of 0.64, along with a high percentage of polymorphic loci, 89%. The coefficient of genetic differentiation (Fst) was 0.15, indicating that there was moderate genetic differentiation. The genetic diversity...

Variation and its distribution in wild cacao populations from the Brazilian Amazon 52
Dias,Luiz Antônio dos Santos; Barriga,Júlio Pontes; Kageyama,Paulo Yoshio; Almeida,Caio Márcio Vasconcellos Cordeiro de.
A sample of 64 progenies (320 cacao trees as a whole) from four Brazilian Amazon basins was collected and evaluated on the basis of 15 fruit and seed traits. Nested univariate analyses of variance showed significant variation across progenies and basins. However, most of the variability appeared to be due to among trees and basins differences. The multivariate analysis showed that the differentiation in cacao populations occurred among basins. Since cacao diversity was predominantly found in trees within basins and among basins, one should optimize the collecting process by taking as many trees as possible starting from few progenies and many river basins. These findings seemed to validate gene conservation efforts made to date to preserve the cacao...

Genetic diversity in natural populations of Hancornia speciosa Gomes: Implications for conservation of genetic resources 64
Fajardo,Cristiane Gouvêa; Costa,Daniel Ferreira da; Chagas,Kyvia Pontes Teixeira das; Vieira,Fábio de Almeida.
ABSTRACT The continuing fragmentation of forests has been a threat to the maintenance of genetic resources. Genetic diversity is fundamental to the survival of species in natural environments in the long term, as well as being the basis for genetic improvement. The objective of this study was to evaluate the genetic diversity in natural populations of Hancornia speciosa and to contribute to the development of conservation strategies. We sampled 105 individuals of H. speciosa, distributed in seven populations. The ISSR (Inter-Simple Sequence Repeat) markers provided 70 loci, of which 81% were polymorphic. The mean genetic diversity of Nei (h) was 0.19, and the Shannon index (I) was 0.27. The h and I diversity indices ranged respectively from 0.16 to 0.24 in...

Proximal causes of genetic variation between and within populations of rauli (Nothofagus nervosa) 118
Carrasco,Basilio; Garnier,Maurice; Eaton,Lafayette; Guevara,Rafael; Carú,Margarita.
Random Amplified Polymorphic DNA(RAPD)markers were used to assess the genetic diversity of 587 individuals, belonging to 22 populations of Nothofagus nervosa that were distributed through the Coastal (38°S to 41 °S) and Andes Mountains in Central-Southern Chile (36°S to 40°S). The objective of this study was to complement the genetic inferences previously determined by isozyme analysis, in order to obtain more accurate genetic diversity estimations. We scored 81.8% of the polymorphic loci of the samples tested. The average incidence of genetic polymorphism within populations was high, with values ranging between 33% and 63%. Analysis of molecular variance (AMOVA) showed most of the genetic variation was distributed within populations (87.6%), but FST...

Heterogeneous genetic structure in a natural population of Raulí (Nothofagus nervosa) 118
Carrasco,Basilio; Eaton,Lafayette; Letelier,Luis; Díaz,Carole; García-Gonzáles,Rolando.
Heterozygote deficiencies in natural populations of outbreeding tree species are common and thought to be due mainly to biparental inbreeding. Inbreeding is believed to be caused by family structure within populations, a product of limited seed dispersal and probably limited pollen dispersal. Although both theory and simulation studies predict that structure should be apparent where trees are isolated by distance, most studies of structure in natural populations have detected only a weak spatial genetic structuring. In this contribution, we compare the use of spatial autocorrelation methodology and F statistics with the concept of relatedness to examine the spatial genetic structure in the natural population of a native southern beech and to explore the...

The genetic diversity of Sardinian myrtle (Myrtus communis L.) populations 69
Melito,Sara; Chessa,Innocenza; Erre,Patrizia; Podani,János; Mulas,Maurizio.
Background: The myrtle (Myrtus communis) is a common shrub widespread in the Mediterranean Basin. Its fruit and leaves exhibit antioxidant, antibacterial and antifungal properties, and are used for their content of essential oils and for their medicinal properties, but most commonly as an ingredient in locally made liquor. The uncontrolled exploitation of natural stands has reduced both the species' geographical coverage and the size of individual populations. The selection of genotypes for controlled cultivation requires a characterization of the genetic diversity present both within and between populations. Results: Genotypic variation was evaluated using ISSR profiling and genetic diversity characterized using standard population genetics approaches....

Genetic diversity in two Italian almond collections 69
Rigoldi,Maria Pia; Rapposelli,Emma; De Giorgio,Donato; Resta,Paolo; Porceddu,Andrea.
Background Sweet-seeded domesticated almonds were brought to the Mediterranean Basin from central Asia about 4000 years ago. In Italy, most of the almonds produced are cultivated in the southern part of the country. Local populations of the tree in Sardinia are largely seed-derived and mostly self-incompatible, so have developed extensive genetic diversity. The need to protect biodiversity has prompted a revived interest in local genetic materials in almond. Two Italian collections have been established, one in Sardinia and the other in Apulia. These collections were the focus of the present evaluation of genetic diversity. Results Eleven SSRs (microsatellites) were used for fingerprinting. The Sardinian germplasm was highly polymorphic, revealing a mean...

Genetic structure of red-handed howler monkey populations in the fragmented landscape of Eastern Brazilian Amazonia 74
Bastos,Heitor B.; Gonçalves,Evonnildo C.; Ferrari,Stephen F.; Silva,Artur; Schneider,Maria Paula C..
We genotyped 15 microsatellite loci in order to evaluate the effects of habitat fragmentation, caused by flooding of the Tucuruí reservoir, on the genetic structure of Alouatta belzebul in eastern Amazonia. The analysis included two populations sampled in 1984, representing both margins of the Tocantins river, and three populations sampled 18 years later. Minimal differences in the diversity levels between present-day (Ho = 0.62-0.69 and A R = 6.07-7.21) and pre-flooding (Ho = 0.60-0.62 and A R = 6.27-6.77) populations indicated there was no significant loss of genetic variability, possibly because of successful management strategies applied during the flooding. The changes observed were limited to shifts in the composition of alleles, which presumably...

Genetic structure of Argentinean hexaploid wheat germplasm 74
Vanzetti,Leonardo S.; Yerkovich,Nadia; Chialvo,Eugenia; Lombardo,Lucio; Vaschetto,Luis; Helguera,Marcelo.
The identification of genetically homogeneous groups of individuals is an ancient issue in population genetics and in the case of crops like wheat, it can be valuable information for breeding programs, genetic mapping and germplasm resources. In this work we determined the genetic structure of a set of 102 Argentinean bread wheat (Triticum aestivum L.) elite cultivars using 38 biochemical and molecular markers (functional, closely linked to genes and neutral ones) distributed throughout 18 wheat chromosomes. Genetic relationships among these lines were examined using model-based clustering methods. In the analysis three subpopulations were identified which correspond largely to the origin of the germplasm used by the main breeding programs in Argentina.

A wide array of molecular markers has been used to investigate the genetic diversity among common bean species. However, the best combination of markers for studying such diversity among common bean cultivars has yet to be determined. Few reports have examined the genetic diversity of the carioca bean, commercially one of the most important common beans in Brazil. In this study, we examined the usefulness of two molecular marker systems (simple sequence repeats - SSRs and amplified fragment length polymorphisms - AFLPs) for assessing the genetic diversity of carioca beans. The amount of information provided by Roger's modified genetic distance was used to analyze SSR data and Jaccards similarity coefficient was used for AFLP data. Seventy SSRs were...

High genetic diversity and connectivity in Colossoma macropomum in the Amazon basin revealed by microsatellite markers 74
Fazzi-Gomes,Paola; Guerreiro,Sávio; Palheta,Glauber David Almeida; Melo,Nuno Filipe Alves Correa de; Santos,Sidney; Hamoy,Igor.
Abstract Colossoma macropomum is the second largest scaled fish of the Amazon. It is economically important for commercial fisheries and for aquaculture, but few studies have examined the diversity and genetic structure of natural populations of this species. The aim of this study was to investigate the levels of genetic variability and connectivity that exist between three natural populations of C. macropomum from the Amazon basin. In total, 247 samples were collected from the municipalities of Tefé, Manaus, and Santarém. The populations were genotyped using a panel of 12 multiplex microsatellite markers. The genetic diversity found in these populations was high and similar to other populations described in the literature. These populations showed a...

Evidence of two lineages of the dengue vector Aedes aegypti in the Brazilian Amazon, based on mitochondrial DNA ND4 gene sequences 74
Lima Júnior,Raimundo Sousa; Scarpassa,Vera Margarete.
Genetic variation was estimated in ten samples populations of Aedes aegypti from the Brazilian Amazon, by using a 380 bp fragment of the mitochocondrial NADH dehydrogenase subunit 4 (ND4) gene. A total of 123 individuals were analyzed, whereby 13 haplotypes were found. Mean genetic diversity was slightly high (h = 0.666 ± 0.029; π = 0.0115 ± 0.0010). Two AMOVA analyses indicated that most of the variation (~70%-72%) occurred within populations. The variation found among and between populations within the groups disclosed lower, but even so, highly significant values. F ST values were not significant in most of the comparisons, except for the samples from Pacaraima and Rio Branco. The isolation by distance (IBD) model was not significant (r = 0.2880; p =...

Population genetic analysis of Caiman crocodilus (Linnaeus, 1758) from South America 74
Vasconcelos,William R.; Hrbek,Tomas; Silveira,Ronis da; Thoisy,Benoit de; Marioni,Boris; Farias,Izeni P..
The genetic structure of Caiman crocodilus was investigated using a 1085 bp mtDNA fragment of the cytochrome b gene. Inferences were based on 125 individuals from nine localities in Peru, Brazil and French Guiana. With the exception of Mamirauá Lake, Anavilhanas Archipelago and the Tapará Community which show a signal of demographic expansion, the sampled localities are in a mutation-drift genetic equilibrium. Divergence between the Amazon basin and extra-Amazon basin localities is significant; however, inference from Nested Clade Analysis cannot distinguish between continuous range expansion, long distance colonization or past fragmentation; however, past fragmentation is unlikely due to low number of mutational steps separating these two regions. The...

Dipteryx alata is a native fruit tree species of the cerrado (Brazilian savanna) that has great economic potential because of its multiple uses. Knowledge of how the genetic variability of this species is organized within and among populations would be useful for genetic conservation and breeding programs. We used nine simple sequence repeat (SSR) primers developed for Dipteryx odorata to evaluate the genetic structure of three populations of D. alata located in central Brazil based on a leaf sample analysis from 101 adults. The outcrossing rate was evaluated using 300 open-pollinated offspring from 25 seed-trees. Pollen dispersal was measured by parentage analysis. We used spatial genetic structure (SGS) to test the minimal distance for harvesting seeds...

Analysis by RAPD of the genetic structure of Astyanax altiparanae (Pisces, Characiformes) in reservoirs on the Paranapanema River, Brazil 74
Leuzzi,Maria Sueli Papa; Almeida,Fernanda Simões de; Orsi,Mário Luís; Sodré,Leda Maria Koelblinger.
In this study, the RAPD technique was used to analyze the genetic structure of populations of the fish Astyanax altiparanae (Characidae, Tetragonopterinae) living in the lower, middle and upper Paranapanema River, Brazil. The aim was to assess this structure regarding fish handling and conservation programs. The genetic variability (P) was found to be 42.64%, 75% and 75% in the low, middle and upper reaches, respectively. The dendrogram of genetic similarity, obtained by comparative analysis of the sets of samples from the three sites, showed the formation of three clusters. All of the genetic parameters used indicate that the population in the lower Paranapanema is genetically different from those in the middle and upper sections. The theta P test shows...
