Genetic diversity between native and improved Cattleya walkeriana Gardner famous clones Agronomy
Tambarussi, Evandro Vagner; Ann Veasey, Elizabeth; Menezes, Lou; Ibañes, Bruna; Lombardi, Kátia Cylene; Vencovsky, Roland.
The aim of this study was to evaluate the genetic diversity among native plants and some individuals obtained from crosses with unknown genealogy of C. walkeriana as well as C. loddigesii and C. nobilior and to advance towards solving the question of the genetic purity of the “Orchidglade” clone. Eight microsatellite loci were used to evaluate the genetic diversity between individuals of C. walkeriana. Microsatellites were not efficient in determining the genetic diversity between C. walkeriana groups (native and improved). The difficulty in determining the genetic distance between the different genotypes can be attributed to the complex mating system of the species and to a weak genetic barrier that facilitates the development of hybrids. Our analysis...

DNA barcode regions for differentiating Cattleya walkeriana and C. loddigesii Biological Sciences
Rivera-Jiménez, Hernando; Rossini, Bruno César; Tambarussi, Evandro Vagner; Ann Veasey, Elizabeth; Ibanes, Bruna; Marino, Celso Luis.
Growers appreciate Cattleya walkeriana and C. loddigesii due to striking shape and rarity. Thus, this study aimed to evaluate the feasibility of DNA barcode regions, namely ITS1, ITS2 and rpoC1, to discriminate between C. walkeriana and C. loddigesii species. DNA barcode regions were successfully amplified using primers designed to amplify plants. We also included sequences from public databases in order to test if these regions were able to discriminate C. walkeriana and C. loddigesii from other Cattleya species. These regions, and their combinations, demonstrated that the ITS1+ITS2 had the highest average interspecific distance (11.1%), followed by rpoC1 (1.06%). For species discrimination, ITS1+ITS2 provided the best results. The combined data set of...

Collection, Characterization and Use of Genetic Bariation in Chilean Bean Germplasm (Phaseolus vulgaris L.) Agricultura Técnica
Bascur B.,Gabriel; Tay U.,Juan.
Recently performed studies on the type of seed protein present from several origins and their morphological traits have shown that the common bean (Phaseolus vulgaris L.) is native to America, being a species without a specific center of origin and with two areas of domestication: Central America and South America. In this last center, three strains were determined, one of them is called "the Chilean strain", which as noted is a sub-center of genetic diversity for this species. With the purpose of collecting and preserving the genetic diversity present in the Chilean ecotypes, a germplasm collection was made, from Arica to Chiloe, visiting 207 locations in 28 expeditions and collecting 1239 accessions. This material was seeded in two Regional Center of...

Morphological and molecular characterization of common bean landraces cultivated in the semi-arid Mexican high plateau Boletín de la Sociedad
Gill Langarica,Homar Rene; Rosales Serna,Rigoberto; Hernandez Delgado,Sanjuana; Mayek Perez,Netzahualcoyotl.
The objective of the current research was to characterize the common bean in agricultural fields planted with common bean landraces from the semi-arid Mexican high plateau using both morphological and amplified fragment length polymorphism (AFLPs) data. The morphological traits were discriminating and exhibited the clustering of 150 accessions based on the geographic origin and seed coat color. AFLP primer combinations exhibited a polymorphic range between 0.292 (E-AGG + M-ACT) and 0.375 (E-ACA + M-AGA). The frequency and distribution of the polymorphic fragments allowed the detection of a larger number of rare fragments in accessions 121 and 111 (Flor de Mayo and black common beans, respectively). The analysis of genetic relationships, analysis of...

Pathogenicity mechanisms of prokaryotic cells: an evolutionary view BJID
Sousa,Cristina Paiva de.
The success of pathogenic microbes depends on their ability to colonize host tissues and to counter host defense mechanisms. Microorganisms can produce overwhelming infection because of their relatively short generation times, and because they have evolved powerful mechanisms for generating phenotypic diversity as an efficient strategy for adapting to rapidly responding immune system defenses and the broad range of polymorphisms characteristic of different host tissues. Bacterial evolution may not be a continuous process, but more of a succession of temporally spaced major events. These events cause a non-gradual sequence of adaptations to a given environment. The pathogenicity islands may be genetically unstable elements, and many of the genes coding for...

Carneiro,Valdirene Leão; Bendicho,Maria Teresita; Santos,Rosalina Guedes; Casela,Marilda; Netto,Eduardo M.; Mota,Scarlet Torres Moraes; Pina,Iza Cristina Araújo; Nascimento,Roberto Meyer; Freire,Songeli Menezes; Barbosa,Theolis.
ABSTRACT Introduction Latent tuberculosis infection diagnosis based on the release of interferon-gamma in cultures of peripheral blood cells stimulated with Mycobacterium tuberculosis antigens has replaced the tuberculin skin test in many countries with low tuberculosis prevalence. The IFN-γ production can be influenced by genetic polymorphisms, of which the IFNG + 874 (rs62559044) locus is the most studied. We investigated the possible influence of the IFNG + 874 A/T polymorphism on interferon-gamma test performance. Methods Patients diagnosed with pulmonary tuberculosis (75), volunteers with positive tuberculin skin test (70) and healthy volunteers with negative tuberculin skin test and no history of contact with tuberculosis (57) were evaluated...

Khatoon,Lubna; Khan,Inam Ullah; Shah,Shahid Ali; Jan,Muhammad Ishtiaq; Ullah,Farhat; Malik,Salman Akbar.
Malaria is one of the serious diseases threatening human health in Pakistan and contributes to a large proportion of the total malaria deaths in South Asia. However, little is known about the nature and extent of genetic diversity of the malarial parasites circulating in Pakistan. This study was designed to assess the infection status of Plasmodium and the genetic diversity of Plasmodium vivax and Plasmodium falciparum by analyzing msp-3α, msp-3β and msp-1, msp-2 genes respectively using allele specific nested PCR and RFLP assays. For this purpose, 130 field isolates were collected from the individuals who exhibited clinical symptoms associated with malaria in the Kohat region of Khyber Pakhtoonkhwa (KPK), Pakistan. Among 130 blood samples collected, P....

Genetic variations of EBV-LMP1 from nasopharyngeal carcinoma biopsies: potential loss of T cell epitopes BJMBR
Tang,Y.L.; Lu,J.H.; Cao,L.; Wu,M.H.; Peng,S.P.; Zhou,H.D.; Huang,C.; Yang,Y.X.; Zhou,Y.H.; Chen,Q.; Li,X.L.; Zhou,M.; Li,G.Y..
To find Epstein-Barr virus (EBV) strains with genetic variations of EBV latent membrane protein 1 (EBV-LMP1) from nasopharyngeal carcinoma (NPC), the full-length DNA of LMP1 genes from 21 NPC biopsies obtained in Hunan province in southern China was amplified and sequenced. Our sequences were compared to those previously reported by the Clustal V method. Results showed that all 21 sequences displayed two amino acid changes most frequently in LMP1 of CD4+ T cell epitopes at codons 144 (F®I, 21/21) and 212 (G®S, 19/21) or (G®N, 2/21). We also show that type A EBV strain is prevalent in the cases of NPC from Hunan province with a 30-bp 18/21 deletion, and we highlight that this deletion resulted in loss of one of the CD4+ T cell-restricted epitopes. The other...

Coping with genetic diversity: the contribution of pathogen and human genomics to modern vaccinology BJMBR
Lemaire,D.; Barbosa,T.; Rihet,P..
Vaccine development faces major difficulties partly because of genetic variation in both infectious organisms and humans. This causes antigenic variation in infectious agents and a high interindividual variability in the human response to the vaccine. The exponential growth of genome sequence information has induced a shift from conventional culture-based to genome-based vaccinology, and allows the tackling of challenges in vaccine development due to pathogen genetic variability. Additionally, recent advances in immunogenetics and genomics should help in the understanding of the influence of genetic factors on the interindividual and interpopulation variations in immune responses to vaccines, and could be useful for developing new vaccine strategies....

Random amplified polymorphic DNA profiles as a tool for the characterization of Brazilian keratitis isolates of the genus Acanthamoeba BJMBR
Alves,J.M.P.; Gusmão,C.X.; Teixeira,M.M.G.; Freitas,D.; Foronda,A.S.; Affonso,H.T..
The genus Acanthamoeba comprises free-living amebae identified as opportunistic pathogens of humans and other animal species. Morphological, biochemical and molecular approaches have shown wide genetic diversity within the genus. In an attempt to determine the genetic relatedness among isolates of Acanthamoeba we analyzed randomly amplified polymorphic DNA (RAPD) profiles of 11 Brazilian isolates from cases of human keratitis and 8 American type culture collection (ATCC) reference strains. We found that ATCC strains belonging to the same species present polymorphic RAPD profiles whereas strains of different species show very similar profiles. Although most Brazilian isolates could not be assigned with certainty to any of the reference species, they could...

ABSTRACT Group A rotaviruses are the main causative agent of infantile gastroenteritis. The segmented nature of the viral genome allows reassortment of genome segments, which can generate genetic variants. In this study, we characterized the diversity of the VP7, VP4 (VP8*), VP6, NSP4, and NSP5 genes of the rotaviruses that circulated from 2005 to 2011 in the Triângulo Mineiro (TM) region of Brazil. Samples with genotypes G2 (sublineages IVa-1 and IVa-3), G1 (sublineage I-A), G9 (lineage III), G12 (lineages II and III), G8 (lineage II), G3 (lineage III), P[4] (sublineages IVa and IVb), P[8] (sublineages P[8]-3.6, P[8]-3.3, and P[8]-3.1), I2 (lineage VII), E2 (lineages VI, XII, and X), and H2 (lineage III) were identified. Atypical phenotypic characteristics of klebsiella pneumoniae isolates from an outbreak in a neonatal intensive care unit in Brazil BJM
Otman,J.; Perugini,M.E.; Tognim,M.C.B.; Vidotto,M.C..
Extended-spectrum beta-Lactamase-producing (ESBL) Klebsiella sp.isolates from an outbreak in a Neonatal Intensive Care Unit (NICU) at a teaching hospital in Londrina, Paraná State, Brazil, presented atypical phenotypic characteristics that hampered their identification and the distinction between Klebsiella and Enterobacter species. Ten isolates were identified as K. pneumoniae due to negative reactions for motility and inducible beta-lactamase test (ESBL and AmpC) despite being positive for ornithyne descarboxilase. These isolates were genotyped by ribotyping and polymerase chain reaction (PCR) with repetitive extragenic palindromic sequences (REP). Ribotyping by means of an automated instrument and EcoRI and Pvu II as restriction enzymes resulted...

Genetic variation among and within tritipyrum (Thinopyrum bessarabicum x Triticum durum) lines using PCR-based molecular markers Ciencia e Investigación Agraria
Siahsar,Barat Ali; Allahdoo,Maryam; Shahsevand Hassani,Hossein.
Primary hexaploid tritipyrum lines are amphiploids between Triticum durum and Thinopyrum bessarabicum, which can set seed in at least 250 mM NaCl. Genetic variation within and among 14 lines of primary tritipyrum were investigated by using 2 random and 6 semi random primers that produced clear and reproducible DNA amplicons. Average values for Nei's gene diversity (h) and Shannon indices (I) were 0.115 and 0.178, respectively. These results indicated low variation within primary tritipyrum lines, probably related to unbiased chromosome and partial aneuploidy. Total genetic diversity (Ht) and gene diversity within lines (Hs) were 0.2114 and 0.1156, respectively. The coefficient of genetic differentiation (Gst) was equal to 0.4534. AMOVA showed highly...
