Improved calibration of a solid substrate fermentation model Electron. J. Biotechnol.
Sacher,Johannes; Saa,Pedro; Cárcamo,Martín; López,Javiera; Gelmi,Claudio A; Pérez-Correa,Ricardo.
Background: Calibration of dynamic models in biotechnology is challenging. Kinetic models are usually complex and differential equations are highly coupled involving a large number of parameters. In addition, available measurements are scarce and infrequent, and some key variables are often non-measurable. Therefore, effective optimization and statistical analysis methods are crucial to achieve meaningful results. In this research, we apply a metaheuristic scatter search algorithm to calibrate a solid substrate cultivation model. Results: Even though scatter search has shown to be effective for calibrating difficult nonlinear models, we show here that a posteriori analysis can significantly improve the accuracy and reliability of the estimation....

Fast and reliable calibration of solid substrate fermentation kinetic models using advanced non-linear programming techniques Electron. J. Biotechnol.
Araya,M. Macarena; Arrieta,Juan J; Pérez-Correa,J. Ricardo; Biegler,Lorenz T; Jorquera,Héctor.
Calibration of mechanistic kinetic models describing microorganism growth and secondary metabolite production on solid substrates is difficult due to model complexity given the sheer number of parameters needing to be estimated and violation of standard conditions of numerical regularity. We show how advanced non-linear programming techniques can be applied to achieve fast and reliable calibration of a complex kinetic model describing growth of Gibberella fujikuroi and production of gibberellic acid on an inert solid support in glass columns. Experimental culture data was obtained under different temperature and water activity conditions. Model differential equations were discretized using orthogonal collocations on finite elements while model calibration...

Revival of ancient marine dinoflagellates using molecular biostimulation ArchiMer
Delebecq, Gaspard; Schmidt, Sabine; Ehrhold, Axel; Latimier, Marie; Siano, Raffaele.
The biological processes involved in the preservation, viability, and revival of long‐term dormant dinoflagellate cysts buried in sediments remain unknown. Based on studies of plant seed physiology, we tested whether the revival of ancient cysts preserved in century‐old sediments from the Bay of Brest (France) could be stimulated by melatonin and gibberellic acid, two molecules commonly used in seed priming. Dinoflagellates were revived from sediments dated to approximately 150 years ago (156 ± 27, 32 cm depth), extending the known record age of cyst viability previously established as around one century. A culture suspension of sediments mixed with melatonin and gibberellic acid solutions as biostimulants exhibited germination of 11 dinoflagellate taxa...

MOHAMED,N.; TAHA,R.M.; RAZAK,U.N.A.A.; ELIAS,H..
ABSTRACT: An efficient protocol for in vitro flowering was successfully established for Impatiens balsamina cv Dwarf Bush, an important medicinal plant, through tissue culture techniques. Shoot, stem and petiole explants obtained from 4 week-old aseptic seedlings cultured on MS medium supplemented with different concentrations of plant growth regulator (PGR) were used for in vitro flower induction. Gibberellic acid (GA3), benzylaminopurine (BAP) and kinetin (Kin) treatment singly applied in MS media (pH 5.8), could all stimulate flowering at 23-26 oC with photoperiod of 16 hours light and 8 hours dark. It was observed that shoot explants were more responsive than stem explants in floral formation. Regeneration was achieved via direct organogenesis. For...

Factors Affecting Seed Dormancy and Germination of Greater Bur-Parsley (Turgenia latifolia) Planta Daninha
REZVANI,M.; SADATIAN,S.A.; NIKKHAHKOUCHAKSARAEI,H..
ABSTRACT: Our knowledge about seed dormancy breaking and environmental factors affecting seed germination of greater bur-parsley (Turgenia latifolia) is restricted. This study has addressed some seed dormancy breaking techniques, including different concentrations of gibberellic acid (GA3) and potassium nitrate (KNO3), leaching duration, physical scarification as well as some environmental factors effective on seed germination such as salt and drought stresses, pH and seed planting depth. Seed germination was promoted with lower concentrations of KNO3 (0.01 to 0.02 g L-1), while higher concentrations reduced germination percentage. Seed dormancy was declined by low concentrations of GA3 up to 100 ppm. Seeds of greater bur-parsley germinated in a range of...

Leaf proline content and its relation to fruit load and flowering in citrus under field conditions Rev. Bras. Frutic.
Arias-Sibillotte,Mercedes; Borges,Alejandra; Díaz,Pedro; Ferenczi,Alejandra; Severino,Vivian.
Abstract Citrus species grown in temperate zones have sprouting inhibited in winter and this is retaken in spring when they also bloom. The main factor that defines the vegetative or reproductive destiny of the buds is the presence of fruits in development. Low winter temperatures slow plant metabolism, which has been reported as necessary for the expression of the reproductive program. During the winter period of the annual cycle, osmotically active compounds such as proline are increased as part of the cold tolerance response. We investigated whether the presence of developing fruit and the application of exogenous gibberellins affect the proline level in citrus leaves. In an orchard of adult ‘Montenegrina’ mandarin trees, leaf proline contents were...

In vitro germination to overcome dormancy in seeds of ‘Red Globe’, ‘Italia’ and ‘Niagara Rosada’ grapes Rev. Bras. Frutic.
Generoso,Andressa Leal; Viana,Alexandre Pio; Carvalho,Virginia Silva; Costa Júnior,Otalício Damásio da.
Abstract Dormancy in grape seeds slows the progress of many breeding programs of the crop, culminating in low uniformity and low germination percentages. Conventional methods used to overcome dormancy are time-consuming. Thus, in vitro seed germination emerges as a promising alternative to ensure the germination of grape seeds. In this study, we examined the in vitro germination and vigor of seedlings originating from seeds of ‘Red Globe’, ‘Italia’ and ‘Niagara Rosada’ grapes in growth media supplemented with five concentrations of gibberellic acid (GA3) (0, 1.41, 2.83, 4.24 and 5.66 µmol L-1) for 47 days. The use of GA3 increased seed germination percentage and seedling vigor, in the three varieties. Therefore, for the in vitro germination of seeds of...

Gibberellin and cytokinin effects on soybean growth Scientia Agricola
Leite,Vagner Maximino; Rosolem,Ciro Antonio; Rodrigues,João Domingos.
Soybean is an important crop in Brazil. Nonetheless, there are no reports on the use of plant growth regulator potential in relation to this crop in the national literature. To better understand the role of these compounds, a pot experiment was carried out to study effects of GA3 and cytokinin on the vegetative growth of the soybean. GA3 (50 mg L-1) was applied as seed treatment, leaving plants with water application as control. GA3 (100 mg L-1) and cytokinin (30 mg L-1) were sprayed on leaves at the physiological stage V3/V4, and 15 days after, cytokinin (30 mg L-1), also as foliar spray. Seed treatment decreased plant emergence and initial soybean root growth, but as the season progressed, differences in root growth disappeared; plants were shorter, and...

Effects of chemical treatments and environmental factors on seed dormancy and germination of shepherd's purse (Capsella bursa-pastoris (L.) Medic.) Acta Botanica
Rezvani,Mohammad; Zaefarian,Faezeh; Amini,Vajihe.
Shepherd's purse (Capsella bursa-pastoris) is a problematic weed in citrus orchards and crop fields in northern Iran. In a series of laboratory and greenhouse experiments, we evaluated the effects that treatment with gibberellic acid (GA3) and potassium nitrate (KNO3), as well as environmental factors, including temperature, the duration of pre-chilling (wet and dry), drought stress, salt stress, pH, and sowing depth, have on seed dormancy breaking and germination in C. bursa-pastoris. Treatment with GA3 strongly stimulated germination of C. bursa-pastoris in conditions of light/dark and continuous darkness. The germination rate was highest (40.08%) for seeds treated with 400 ppm of GA3 in the light/dark condition. Treatment with KNO3 did not significantly...

Abstract: Somatic embryogenesis from explants from hermaphrodite papaya mother plants is an alternative for the production of true-to-type plants without the need for sexing. This study aimed to analyze hormonal and osmotic inducers in different somatic embryogenesis stages in the commercial hermaphrodite hybrid papaya UENF/Caliman 01. Leaf disks from in vitro shoots originated from ex vitro hermaphrodite plants were cultured in induction medium supplemented with different concentrations of 2,4-D (6, 9, 12, 15, and 18 μM) and 4-CPA (19, 22, 25, 28, and 31 μM). After 90 days, the formation of somatic embryos was verified. The 2,4-D induced the formation of light brown calli with low frequency (20%) of somatic embryogenesis. However, 4-CPA (25 μM) induced...

Effects of Storage and Exogenous Ga3 on Lychee Seed Germination BABT
Ono,Elizabeth Orika; Leonel,Sarita; Duarte Filho,Jayme; Rodrigues,João Domingos.
The effects of storage time and exogenous gibberellic acid on lychee seeds germination were studied. The seeds were removed from ripe fruits, washed, dried, stored at 8°C for 0, 15 and 30 days, and soaked during 24 hours in water and GA3 at 50, 100 and 200 mg.L-1 solutions. As the storage period increased, the germination capacity was lost, and the seeds had a short germinability after 30-day storage period. Gibberellic acid had not significant effect on enhancing both the germination percentage and rate.
